Goldnanopartikel besitzen im Vergleich zu makroskopischem Gold veränderte physikalische und chemische Eigenschaften. Dabei ist die Änderung der optischen Eigenschaften auf das Auftreten von lokalisierte Oberflächenplasmonen (kollektive Elektronendichteschwankungen) zurückzuführen. Aufgrund der Abhängigkeit der Plasmonik von der äußeren Form der Nanopartikel lassen sich die optischen und elektronischen Eigenschaften über post-synthetische Formmodifikation direkt manipulieren. So zeigen sphärische Goldnanopartikel (AuNPs) lediglich eine plasmonische Resonanzbande im sichtbaren Spektrum, wohingegen eindimensional elongierte Goldnanorods (AuNRs) eine transversale (t-LSPR) und eine longitudinale lokalisierte Oberflächenplasmonenresonanz (l-LSPR) aufweisen. Die lineare Abhängigkeit der Resonanzwellenlänge der l-LSPR von dem Verhältnis der Länge zum Durchmesser der AuNRs (Aspektverhältnis) ermöglicht vielfältige Anwendungen der AuNRs durch die Möglichkeit der einfachen Manipulation der plasmonischen
Resonanzbande im sichtbaren und nahinfraroten Spektralbereich des Extinktionsspektrums.

In dieser Dissertation wurde die annähernd stufenlose Manipulation der Länge der AuNRs bei konstantem Durchmesser untersucht. Diese auf einer milden Oxidationsmethode durch Komproportionierung beruhende Verkürzung ermöglichte den Erhalt der klaren Unterscheidbarkeit der beiden plasmonischen Resonanzbanden bei der Einstellung des Aspektverhältnisses bis auf einenWert von 1.4 sowie die entsprechende Feinjustierung der l-LSPR-Resonanzwellenlänge bis auf einen Wert von 568nm im Extinktionsspektrum. Basierend auf empirischen Daten der systematischen Verkürzung sowie einem geometrischen Ansatz wurde ein Modell entwickelt,
das für Aspektverhältnisse AR > 1.4 vollständige Kontrolle über die Verkürzung der AuNRs und damit die Einstellung des Aspektverhältnisses sowie die Lage der l-LSPR-Resonanzwellenlänge der AuNRs ermöglicht.

Im zweiten Teil dieser Dissertation wurden biofunktionalisierte AuNRs mit einer plasmonischen Resonanzbande im biologischen Transparenzfenster der menschlichen Haut (650 nm bis 900 nm) als Energiekonverter für plasmonische Photothermaltherapie in BaF3-Zellen eingesetzt. Das Verhalten der individuell mit einem Laser bestrahlten Zellen wurde lichtmikroskopisch verfolgt. Dabei konnte gezeigt werden, dass die Schädigung der Zellmembran durch die lokale Temperaturerhöhung des photothermalen Effekts der AuNRs zeitverzögert zum Anschwellen der Zelle einsetzt. Dieses Verhalten spricht für die primäre Schädigung von inneren Zellkompartimenten und die Störung von essentiellen Zellfunktionen wie der Homöostase und gegen die direkte Schädigung der Zellmembran durch die lokale Temperaturerhöhung. Weiterhin konnte eine signifikante Verbesserung der Effizienz dieser Methode durch die Nutzung von spezifischer Aptamer-Rezeptor-Wechselwirkung erzielt werden.

Compared to macroscopic gold, gold nanoparticles (AuNPs) have altered physical and chemical properties. The modified optical properties base on localized surface plasmons (collective oscillation of electron density). Due to the dependence of plasmonics on the external shape of the nanoparticles, it is possible to tune the optical and electronic properties by shape control. Spherical AuNPs, for example, only show one plasmonic peak in the visible spectrum, whereas one-dimensionally elongated goldnanorods (AuNRs) show a transversal (t-LSPR) and a longitudinal localized surface plasmon resonance (l-LSPR). The linear dependence of the l-lspr
wavelength on the ratio between length and diameter of the AuNRs (aspect ratio) allows a wide variety of applications through the possibility of manipulation of the plasmonic resonance band in the absorbance spectrum over the entire visible and near infrared range.

In this thesis, the almost continous variable manipulation of the length of the AuNRs at constant diameter was investigated. The manipulation is based on a mild oxidation method by means of comproportionation and made it possible to reduce the aspect ratio to 1.4 preserving both plasmonic bands distinguishable. In accordance with the aspect ratio, the spectral position of the l-LSPR band was fine-tuned up to 568nm in the absorbance spectrum. Based on empirical data of the systematic shortening and a geometric approach, a model has been developed that provides full control of the aspect ratio for AR > 1.4 and thus the spectral position of the l-LSPR absorbance band of the AuNRs.

In the second part of the thesis, biofunctionalized AuNRs with a plasmonic resonance band in the biological transparency window of the human skin (650 nm to 900 nm) have been used as energy converter for plasmonic photothermal therapy in BaF3 cells. The behaviour of the single cells individually treated with laser irradiation was tracked via light microscopy. It was shown that the damage of the cell membrane caused by a local temperature increase due to photothermal behaviour of the AuNRs started delayed compared to the swelling of the cell. This behaviour speaks in favour of the primary damage of inner cell compartments and the disturbance of essential cell functions as homeostasis. Apparently, direct damage to the cell membrane caused by the local temperature increase seems excluded. Fur thermore, a significant improvement was achieved in the efficiency of this method through the use of specific aptamer-receptor interaction.
