Die Insulin-ähnlichen Wachstumsfaktoren (IGF-I und IGF-II) stimulieren Wachstum, Differenzierung und Überleben von verschiedenen Zelltypen über endokrine, parakrine und autokrine Mechanismen. Neben den Hauptkomponenten des IGF-Systems, den IGFs, den IGFBPs sowie den IGF-Rezeptoren und den IGFBP-Proteasen, interagieren eine Reihe von weiteren Proteinen mit IGFs und IGFBPs, was möglicherweise zur zell- und gewebespezifischen Feinregulation von IGF-Wirkungen beiträgt. In der vorliegenden Arbeit wurden verschiedenste biochemische und zellbiologische Methoden eingesetzt, um neue Proteine der IGF-Achse aus Plasmafraktionen und Serum zu identifizieren und zu charakterisieren, die mit humanen IGFs und IGFBPs interagieren. • Mit Hilfe einer IGF-II Affinitätschromatographie wurden vier Proteine gereinigt und als IGFBP-3, IGFBP-5, Transferrin und Plasminogen identifiziert. Westernblot Analysen wiesen zusätzlich die Präsenz von Antithrombin III, Präkallikrein und von IGF II/Mannose-6-Phosphat-Rezeptor-Formen im Eluat der Affinitätssäule nach. • Zusätzlich zu den klassischen IGFBPs gehört Plasminogen zu der Familie der hochaffinen Bindungsproteine für IGF-II. • BIAcore Analysen zeigten, daß Plasminogen auch mit einer hohen Affinität an IGFBP-3 bindet und potentiell mit anderen IGFBP-3-assoziierten Proteinen wie Transferrin an der Bildung von neuen dimeren, trimeren oder oligomeren IGF/IGFBP Komplexen beteiligt sein kann. • Von der IGF-II Affinitätsmatrix wurde auch eine 92 kDa IGFBP-3 Protease eluiert. Ob die Protease direkt oder indirekt über IGFBP oder IGFBP-assoziierte Proteine an die IGF-II-Matrix bindet, ist unklar. • IGFBP-Proteaseaktivität läßt sich im Schwangerenserum durch Gelfiltration in zwei Fraktionen auffinden, die eine molare Masse von 600 kDa (Pool I) und 50 kDa (Pool II) aufweisen. In Pool I konnte neben einer 50 kDa Protease aus der Familie der Disintegrin- und Metalloproteasen (ADAMs), die sich auch in Pool II fand, eine 100 kDa Protease charakterisiert werden, deren proteolytische Aktivität nicht mit Serin- und Metalloproteaseinhibitoren hemmbar war. • Zellbiologische Arbeiten an Transferrin- und IGFBP-3-überexprimierenden MDCK-Zellen zeigten, daß die IGF-I-abhängige Zellproliferation und Aminosäure-Aufnahme im Vergleich mit nicht-transfizierten MDCK-Zellen vermindert war. • In polarisiert wachsenden MDCK-Zellen wird IGFBP-3 bevorzugt apikal und Transferrin bevorzugt basolateral sortiert. Die vorliegende Arbeit zeigt die Existenz von Proteinen in der Zirkulation mit multiplen Funktionen (wie z.B. Fe 3+-Transportmolekül Transferrin; proteolytische Vorstufenform Plasminogen; Integrinrezeptor interagierendes Vitronectin), die gleichzeitig durch die Bindung von IGF-II oder Assoziation mit IGFBP-3 oder beidem an der Bioverfügbarkeit von IGFs beteiligt sein können bzw. auf zellulärer Ebene physiologische Wirkungen der IGFs wie Stimulation der Proliferation bzw. Aminosäure-Aufnahme modulieren.

The insulin-like growth factors (IGF-I and IGF-II) stimulate growth, differentiation and survival of various cell types in an endocrine, paracrine and autocrine fashion. Besides the main components of the IGF system like IGFs, IGFBPs, IGF receptors and IGFBP proteases, several other proteins interact with IGFs and IGFBPs which may regulate cell and tissue specific IGF actions. In this work, several biochemical and cellbiological methods were used to identify and characterize new proteins of the IGF-axis from plasma fractions and serum that interact with human IGFs and IGFBPs. Via IGF-II affinity chromatography four proteins could be separated and identified as IGFBP-3, IGFBP-5, transferrin and plasminogen. Western blot analyses revealed the additional presence of antithrombin III, prekallikrein and the IGF-II/Mannose-6-phosphate receptor in the eluate of the affinity matrix. In addition to the classical IGFBPs, plasminogen is a member of the high-affinity binding proteins of IGF-II. BIAcore analysis showed that plasminogen binds also IGFBP-3 with high affinity and participates with other IGFBP-3-associated proteins like transferrin in the formation of dimeric, trimeric and oligomeric IGF/IGFBP complexes. A 92 kDa IGFBP-3 protease eluted from the IGF-II affinity matrix. Whether this protease binds directly to the IGF-II matrix or indirectly via IGFBP or IGFBP-associated proteins is unclear. IGFBP protease activity can also be detected in human pregnancy serum in two fractions after gelfiltration with two molar masses of 600 kDa (pool I) and 50 kDa (pool II). Besides a 50 kDa protease of the family of disintegrin- and metalloproteases (ADAMs) in pool I, there could be characterized a 100 kDa protease. The proteolytic activity could be inhibited with neither serine nor metalloprotease inhibitors. Cellbiological investigations with transferrrin- and IGFBP-3-overexpressing MDCK cells showed that IGF-dependent cell proliferation and amino acid uptake are lessened in comparison to none-transfected MDCK cells. In polarized growing MDCK cells, IGFBP-3 is sorted preferentially to the apical and transferrin to the basolateral side. This work shows the existence of proteins in the circulation with multiple functions (like ion-transport molecule transferrin; proteolytic zymogen plasminogen; integrin-receptor binding vitronectin) that are involved simultanously in the bioavailability of IGFs through IGF-II binding or association with IGFBP-3 or both or modulation of the physiological role of IGFs like proliferation and amino acid uptake on a cellular level.
