Zusammenfassung: 
In der Arbeit wird die Darstellung konformativ flexibler Nucleosid-5'-monophosphat-Analoga beschrieben. Diese werden für enzymkinetische Untersuchungen an Thymidylatkinase benötigt, dem verantwortlichen Enzym für die verminderte Bioaktivierung des anti-HIV-Wirkstoffes AZT. In Folge einer abstoßenden Wechselwirkung zwischen der 3'-Azidgruppe des konformativ starren AZT-Monophosphates und dem Aspartat-14 des Enzyms kommt es in der Thymidylatkinase zu einer Strukturverzerrung, die eine effektive Phosphorylierung des Substrats behindert. Durch Erhöhung der Flexibilität im Glykon des AZTMPs soll diese abstoßende Wechselwirkung ausgeglichen und somit eine verbesserte Phosphorylierungsrate erreicht werden.

Die Darstellung der carba-Nucleotide erfolgte nach einer konvergenten Synthesestrategie. Sie gliederte sich in die Synthese eines enantiomerenreinen carbocyclischen Grundgerüsts, die Kupplung des Grundgerüsts mit der Nucleobase, Modifikationen an der 3'-Position sowie abschließend die Phosphorylierung der Nucleoside an der 5'-Hydroxygruppe zum entsprechenden carba-Nucleotid.

Als Ausgangsverbindung wurde der chirale Alkohol 2-Benzyloxymethylcyclopent-3-enol gewählt, da hier die benötigte Grundstruktur von 2'-Desoxynucleosiden bereits vorgegeben ist. Das Cyclopentenol 32 wurde durch Alkylierung von Cyclopentadienylnatrium mit Benzylchlormethylether und anschließender enantioselektiver Hydroborierung mit (–)-Diiso-pinocampheylboran in einer Eintopfreaktion mit Gesamtausbeuten zwischen 35 – 45 % erhalten. Das Konzept der konvergenten Synthese beinhaltet die Kupplung eines aktivierten carbocyclischen Grundgerüsts mit der entsprechenden Nucleobase. Hierzu wurde das Cyclopentenol 32 an der freien Hydroxygruppe als Benzylether blockiert und das resultierende Cyclopenten im Anschluss daran regioselektiv hydroboriert. Durch nucleophile Substitution der Hydroxygruppe nach einem modifizierten MITSUNOBU-Protokoll durch N-3-benzoyliertes Thymin und anschließender Abspaltung der Schutzgruppen wurde die Schlüsselverbindung carba-dT 2 in einer Gesamtausbeute von 43 % aus dem Cyclopentenol 1 erhalten. In diesem Zusammenhang wurden umfangreiche Studien zur N-1-/O-2-Selektivität der Kupplungs-reaktion zwischen der Nucleobase und der Alkoholkomponente durchgeführt. Anhand dieser neuen Synthesestrategie konnten erstmals auch carbocyclische iso- und a-Nucleoside dargestellt werden.

Die folgenden Modifikationen an der 3'-Position von carba-dT 2 wurden analog der am natürlichen Furanosesystem beschriebenen Methoden durchgeführt. Die Einführung des Azid-und Thiocyanat-Substituenten gelang hierbei durch nucleophile Substitutionsreaktionen, der Allyl-Rest hingegen wurde durch eine diastereoselektive Radikalreaktion eingebracht. Der Propargyl-Rest wurde durch Bromierung und anschließende doppelte Dehydrohalogenierung aus dem Allyl-Substituenten erhalten.

Die Darstellung der korrespondierenden Nucleosid-5'-monophosphate erfolgte durch Hydrolyse der jeweiligen cycloSal-Phosphattriester. Diese wurden nach einer neuen Variante durch Reaktion des Nucleosids mit cycloSaligenylphosphorchloridat in Pyridin dargestellt. Die Ausbeuten konnten hierbei im Vergleich zur konventionellen Darstellungsmethode mittels eines phosphitylierenden Reagenzes und anschließender Oxidation gerade bei Nucleosiden mit einer 3'-Hydroxygruppe merklich gesteigert werden. Bei der Bestimmung der Hydrolysehalbwertszeiten der Phosphattriester in Phosphatpuffer bei einem pH-Wert von 7.3 konnte eine starke Abhängigkeit der Hydrolysegeschwindigkeiten vom Substituenten in der 3'-Position des Nucleosids beobachtet werden. Demnach führen unpolare Substituenten zu einer Stabilisierung des Triesters, polare Substituenten hingegen beschleunigen die Hydrolyse durch einen assistierenden Effekt.

Die synthetisierten carbocyclischen Nucleosidanaloga sowie deren cycloSal-Phosphattriester wurden auf ihre Aktivität gegen das HI-Virus untersucht. Hierbei zeigt das carbocyclische Analogon des Thymidins carba-dT 25 in CEM/0-Zellen eine dem d4T 2 vergleichbare antivirale Aktivität gegen HIV-1 und HIV-2. Dies ist ein bemerkenswertes Ergebnis, da Nucleosidanaloga mit einer 3'-Hydroxygruppe nicht zu den klassischen anti-HIV-Wirkstoffen zählen.

Nucleoside-based inhibitors of reverse transcriptase (RT) were the first drugs to be used in the chemotherapy of AIDS. After entering the cell, these substances need to be activated to their triphosphates by cellular kinases. Unfortunatelly, for the most extensively used drug, 3'-azido-3'-deoxythymidine (AZT), whereas phosphorylation is facile, the product (AZTMP) is a very poor substrate for the second kinase, thymidylate kinase (TmpK). Due to the steric demand of the azido group in the 3'-position of AZTMP, compared to the 3'-hydroxyl group of the natural substrate, 2'-deoxythymidine monophosphate (dTMP), the structure of the enzyme changes, which causes a 200-fold decrease in the rate of phosphorylation.

In our group, we are interested in investigating the structural requirements of nucleotides, in order to be phosphorylated efficiently by thymidylate kinase (structure/activity relationship). In a first attempt we replaced the azido group in AZTMP by isosteric substituents, e.g. the thiocyanato, the allyl and propargyl group and we increased the flexibility of the nucleotide by using acyclic analogues of thymidine. Surprisingly, none of the prepared nucleotides were substrates for TmpK. Apparently, the acyclic nucleotides were too flexible to bind to the active site of the kinase. In a second attempt we now prepared carbocyclic nucleotides, whose flexibility is reduced compared to the acyclic system, but still increased compared to the natural furanose ring.

By using a new convergent synthetic strategy, enantiomerically pure carbocyclic thymidine (carba-dT) 25 was obtained as a key intermediate for further variations at the 3'-position. As starting material, we chose the chiral cyclopentenol 32, which can easily be prepared from cyclopentadiene by alkylation and a subsequent asymmetric hydroboration. After protection of the free hydroxy group in 32, the resultung chiral olefin can be hydroxylated by different methods, leading to chiral cyclopentanols. Additionally, after inversion of the configuration and protection of the hydroxy group in 32, the chiral olefin can also be hydroxylated, leading to carbocyclic precursors with 2'-deoxy-threo-configuration. The condensation of the carbocyclic moiety and the heterocyclic nucleobase was then achieved by using a modified Mitsunobu reaction, using N-3-protected bases as nucleophiles. Detailed investigations regarding the N-1- vs. O-2-alkylation at the pyrimidine nucleobase were performed. This strategy was also used for the synthesis of carbocyclic BVdU, a nucleoside analogue known to be antivirally active against HSV-1. Additionally, carbocyclic a-nucleosides and carbocyclic iso-nucleosides were prepared by this reaction sequence.

After protection of the 5'-hydroxy group in carba-dT 25, the secondary hydroxy group was substituted by the azido, thiocyanato, allyl and propargyl group. These reactions were performed in accordance with the published data for the natural 2'-deoxyribose system. Finally, the carbocyclic nucleosides 3 were transferred into their monophosphate esters (nucleotides) by hydrolysis of the corresponding cycloSal-phosphate triesters.

Hydrolysis studies of the synthesised cycloSal-phosphate triesters revealed a strong dependence of the hydrolytic stability on the nature of the substituent in the 3'-position of the nucleoside. While unpolar substituents increase the hydrolysis half life of the triester, the hydrolytic stability for 3'-hydroxylated derivatives is decreased. Apparently, polar substituents create a more polar environment, which supports the nucleophile in attacking the phosphate unit. A similar effect was observed for the aminal oxygen in natural nucleosides.

The carbocyclic nucleosides and the corresponding cycloSal-phosphate triesters were evaluated for their ability to inhibit the replication of HIV-1 and HIV-2 in CEM cells. From all compounds tested, carba-dT 2 showed the best antiviral data, with activities similar to those of d4T. This result is particulary interesting with respect to structure/activity relations, since nucleosides with a 3'-hydroxy group do not belong to the class of typical anti-HIV compounds.
