Zusammenfassung: 
Die vorliegende Arbeit beschäftigt sich mit der Charakterisierung von Zielsteuerungen und damit zusammenhängender Funktionen zweier wichtiger Enzyme im Inositolphosphatmetabolismus. Die Inositol-1,4,5-trisphosphat 3-Kinase Isoform B ist ein Schlüsselenzym in der Regulation von Kalziumsignalen, indem es den sekundären Botenstoff Ins(1,4,5)P3 einer Metabolisierung zu höherphosphorylierten InsPs zuführt. Die Inositol-1,3,4,5,6-pentakisphosphat 2-Kinase ist ebenfalls ein Inositolphosphat umsetzendes Enzym, über dessen Funktionen, neben der Phosphorylierung von Ins(1,3,4,5,6)P5 zu InsP6, zu Beginn dieser Arbeit noch nichts bekannt war.

In der vorliegenden Arbeit ist es gelungen, die Aktinbindungsdomäne der Inositol 1,4,5-trisphosphat 3-Kinase Isoform B vollständig zu charakterisieren. Es konnte belegt werden, dass die von Dr. Isabell Schreiber identifizierte Aktinbindungsdomäne (Aminosäuren 108-170) der Ratten Inositol-1,4,5-trisphosphat 3-Kinase Isoform B auch in der humanen Form (Aminosäuren 103-193) hinreichend und notwendig für die F-Aktinbindung des Enzyms ist. Die Aktinbindungsdomäne ist die einzige funktionale F-Aktinzielsteuerungsdomäne in der Inositol-1,4,5-trisphosphat 3-Kinase Isoform B, ihre Bindung an F-Aktin ist reversibel, benötigt keine weiteren unterstützenden Proteine und ist abhängig von der korrekten Sekundärstruktur zweier prädizierter a-Helices. Innerhalb der Aktinbindungsdomäne konnte außerdem ein aktives kanonisches monopartites Kernlokalisationssignal identifiziert werden, das an Aminosäureposition 128 beginnt. Diese duale Zielsteuerungsdomäne besitzt damit die interessante Eigenschaft einer alternativen Zielsteuerung zwischen F-Aktin und Kern.
Die Inositol-1,3,4,5,6-pentakisphosphat 2-Kinase konnte erstmals als ein hochaktives eukaryotisch exprimiertes Enzym angereichert und auf seine Substratspezifität hin untersucht werden. Als Hauptaktivität bestätigte sich die bereits bekannte Phosphorylierung von Ins(1,3,4,5,6)P5 zu InsP6 mit einer spezifischen Aktivität von 0,5 µmol/min/mg. Zusätzlich wurde die Umsetzung von Ins(1,3,4,6)P4 zu Ins(1,2,3,4,6)P5 mit niedrigerer Aktivität von ca. 0,02 µmol/min/mg nachgewiesen. Eine Überexpression der Kinase in COS7-Zellen rief neben dem erwarteten Anstieg der InsP6- und einem Abfall der Ins(1,3,4,5,6)P5-Konzentration eine Erniedrigung des Ins(1,4,5,6)P4- sowie eine Erhöhung des Ins(1,2,3,4,5)P5-Pegels hervor.
Im Rahmen dieser Arbeit wurde außerdem die intrazelluläre Lokalisation der Inositol-1,3,4,5,6-pentakisphosphat 2-Kinase umfassend charakterisiert. Die Kinase weist sowohl zytosolische als auch nukleäre Lokalisation unter Colokalisation mit Euchromatin und nukleärer neusynthetisierter mRNA auf. Für einen dynamischen Wechsel zwischen diesen Lokalisationen sind Zielsteuerungsdomänen notwendig, die das hier beschriebene nukleozytoplasmatische Pendeln der Kinase vermitteln. Sowohl das basische nicht-kanonische Kernlokalisationssignal, dessen Aktivität von der Funktionalität eines putativen Zink bindenden Motivs abhängt, als auch ein Exportin unabhängiges Kernexportsignal konnten beschrieben werden. Außerdem wurde durch die Colokalisation mit zytosolischen mRNA-Akkumulationen und den mRNA-Bindeproteinen PABP und TIAR erstmals die Beteiligung der Inositol-1,3,4,5,6-pentakisphosphat 2-Kinase an Stressgranula nachgewiesen. Anscheinend ist die Inositol-1,3,4,5,6-pentakisphosphat 2-Kinase zusammen mit dem mRNA-Bindeprotein TIAR an der Produktion, der Prozessierung, dem Kernexport und dem Translationsarrest von bestimmten mRNAs in Stressgranula beteiligt. Außerdem konnte TIAR als ein Bindungspartner der IP5-2K identifiziert werden, dessen Kernimport nach Herunterregulation der Kinase blockiert ist.
Die physiologische Relevanz dieser neu identifizierten Funktionen der Inositol-1,3,4,5,6-pentakisphosphat 2-Kinase liegt möglicherweise im Schutz vor Apoptose. In gestressten Zellen könnte die Kinase an der Lagerung bestimmter mRNAs in Stressgranula beteiligt sein, deren Translation zur Einleitung von Apoptose führen würde. Gleichzeitig erfolgt vermutlich die Apoptoseinhibierung durch Erhöhung der InsP6-Konzentration durch die Kinaseaktivität der Inositol-1,3,4,5,6-pentakisphosphat 2-Kinase. Auf diese Weise könnte den Zellen die Gelegenheit gegeben werden den Stress unter Erhalt der Option auf Apoptose zu bewältigen.

In this work localization and the functions of two essential human enzymes of the inositol phosphate metabolism were characterized. Inositol-1,4,5-trisphosphte 3-kinase isoform B is a key player in regulation of Ca2+-signalling starting phosphorylation of the second messenger Ins(1,4,5)P3 to higher phosphorylated inositol phosphates. Inositol-1,3,4,5,6-pentakisphosphate 2-kinase is also an inositol phosphate metabolising enzyme. This work describes novel functions of this enzyme in addition to its phosphorylation of Ins(1,3,4,5,6)P5 to InsP6.
Actin Binding Domain of Inositol-1,4,5-trisphosphte 3-kinase isoform B was characterized. It is shown that the ABD from rat (aa 108-170), first described by Dr. Isabell Schreiber, is also in the human enzyme (aa 103-165) and is essential for binding of the kinase to F-actin. The ABD investigated here is the only Actin Binding Domain in the full-length enzyme. Its binding to F-actin is reversible and no other proteins are needed. Binding requires the correct folding of the two predicted a-helices in the ABD. Interestingly, the ABD contains an active classical monopartite Nuclear Localization Signal beginning at aa 128.
The Inositol-1,3,4,5,6-pentakisphosphate 2-kinase was for the first time expressed in an mammalian expression system and then enriched as an highly active protein and its substrate specificity was investigated. The phosphorylation of Ins(1,3,4,5,6)P5 to InsP6 was confirmed as the main activity of the kinase. The enzyme showed a high specific activity of 0.5 µmol/min/mg. Additionally the conversion of Ins(1,3,4,6)P4 to Ins(1,2,3,4,6)P5 with lower specific activity of 0.02 µmol/min/mg was identified. Furthermore, overexpression of Inositol-1,3,4,5,6-pentakisphosphate 2-kinase in COS7 cells led to the expected loss of Ins(1,3,4,5,6)P5 and an increase in InsP6 concentration. Unexpectedly the amount of Ins(1,2,3,4,5)P5 was also increased, whereas Ins(1,4,5,6)P4 concentration was decreased.
The intracellular localization of Inositol-1,3,4,5,6-pentakisphosphate 2-kinase was fully described for the first time in this work, and a novel function of IPKs was identified. IP5-2K was found to co-localize with mRNA. Furthermore, Inositol-1,3,4,5,6-pentakisphosphate 2-kinase was identified as a component of stress granules by its co-localization with cytosolic mRNA accumulations and the mRNA binding proteins PAPB and TIAR.
The RNA associated functions of Inositol-1,3,4,5,6-pentakisphosphate 2-kinase necessitate targeting domains mediating nuclear import as well as nuclear export of the protein. Here the nuclear localization sequence as well as the nuclear export signal of Inositol-1,3,4,5,6-pentakisphosphate 2-kinase were identified.
Knock-down of Inositol-1,3,4,5,6-pentakisphosphate 2-kinase by siRNA led to a modified nuclear morphology and nuclear import of TIAR was inhibited. Together with TIAR, Inositol-1,3,4,5,6-pentakisphosphate 2-kinase seems to be involved in production, processing, nuclear export and translational arrest of mRNA.
To explain the physiological relevance of these novel functions the following thesis is suggested: In cells exposed to stress Inositol-1,3,4,5,6-pentakisphosphate 2-kinase, together with other proteins, might regulate the delay of apoptosis by the storage of special mRNAs (e.g. TNFa and Fas) in stress granules where translation is inhibited. At the same time, the kinase activity of Inositol-1,3,4,5,6-pentakisphosphate 2-kinase increases InsP6 levels, thus inhibiting apoptosis. This provides the stressed cell time to either recover from stress or induce cell-death by apoptosis.
