Zusammenfassung: 
Ein entscheidender Prozess der Alzheimer Krankheit ist die pathogene Bildung von Neurofibrillenbündel und das dadurch ausgelöste vermehrte Absterben der Nervenzellen im menschlichen Gehirn. Diese Neurofibrillenbündel bestehen aus Aggregaten des Tau-Proteins, das die Mikrotubuli stabilisiert und damit für die intrazelluläre Versorgung der synaptischen Region wichtig ist. Bei der Aggregation des Tau-Proteins spielen zwei Minimalmotive innerhalb der repeat-Regionen des Proteins eine bedeutende Rolle. Im ersten Teil dieser Arbeit wird die Interaktion eines dieser Motive, des t3a (272GGKVQIINKK281), zu dem Tau-Proteinkonstrukt K18, bestehend aus den vier repeat-Regionen und einer Molmasse von 13.2 kDa, untersucht. Zusätzlich wird auch der Einfluss des polyanionischen Heparins auf die Aggregation analysiert. Die Zugabe von Heparin zu einer Lösung des Peptids t3a führt zur sofortigen Aggregation. Durch die Veränderung der Reihenfolge der Präparation, indem zuerst t3a, dann das Protein K18 und erst zum Schluss Heparin hinzugegeben werden, erhält man eine homogene Phase. Die Analyse der NOE Spektren zeigt, dass die Bindung des Peptids im Komplex zu Veränderungen der Signalvolumina in den NOESY Experimenten führt. Da die Signale jedoch stark überlagert sind, konnten keine sequenziellen oder weiterreichenden NOE-Kontakte identifiziert werden. Die Bindung zwischen Protein und Peptid kann durch die Heparinzugabe beschleunigt werden, so dass sie mittels STD NMR-Methode Bindungsprozesse detektiert und charakterisiert werden können. Das Bindungsepitop zeigte den großen Einfluss der Aminosäuren Asn279, Ile277 oder 278, Val275, Gln276 und Lys274, 280 oder 281. Nach der Synthese und Charakterisierung des 15N-markierten Peptids t3a15N konnte das Bindungsepitop durch die Veränderungen der chemischen Verschiebungen im 15N,1H-HSQC-Experiment nach Zugabe des Proteinkonstrukts K18 bestätigt und erweitert werden. Ferner wurde gezeigt, dass die Seitenketten von drei Lysinen Lys274, 280 und 281 ebenfalls an der Bindung beteiligt sind. Durch die Etablierung eines SPR-Systems zur Untersuchung des Komplexes aus immobilisiertem Protein und Peptid stand eine weitere Methode zur Untersuchung der Komplexbildung zur Verfügung. Die Dissoziationskonstante des Komplexes aus Peptid und Proteinkonstrukt konnte zu KD = 1.9 *10-6 M bestimmt werden. Die Sensorgramme zeigten hierbei ein negatives Vorzeichen. Die Dissoziation verlief sehr langsam, so dass keine Auswertung möglich war. Durch Vorgabe der mittels der Sättigungswerte bestimmten Dissoziationkonstanten konnte die Geschwindigkeitskonstante kon zwischen 801 und 69 M-1s-1 bestimmt werden. Hieraus ergibt sich eine off-rate von koff = 10-3 bis 10-4 s-1, die zu der Tatsache passt, dass die STD-Effekte nur mit sehr großer Proteinkonzentration zu beobachten waren. Der zweite Teil der Arbeit beschäftigt sich mit der Untersuchung von drei niedermolekularen Substanzen, welche die Aggregation des Tau-Proteins in vitro und im Zellsystem effektiv verhindern und sogar in der Lage sind, bereits gebildete Aggregate aufzulösen. Bei B4D3 stellte sich heraus, dass die angegebenen Summen- und Strukturformeln nicht zutreffend waren. Nach der Bestimmung der korrekten Summen- und Strukturformel wurde die Bindung des B4D3 an das Proteinkonstrukt K18 mittels STD NMR-Spektroskopie bewiesen und näher charakterisiert. Hierbei wurde das Bindungsepitop der Substanz beschrieben. Durch Titration der Inhibitorkonzentration konnte die Dissoziationskonstante der Komplexbildung aus K18 und B4D3 zu KD = 11.3 *10-6 M bestimmt werden. Das Bindungsereignis konnte durch SPR-Experimente bestätigt werden. Die kinetische Auswertung lieferte für die Geschwindigkeitskonstante der Assoziation 250 M-1s-1, während die Geschwindigkeitskonstante der Dissoziation mit 1.2 • 10-3 s-1 ermittelt wurde. Daraus ergab sich eine Dissoziationskonstante von 6.1 *10-6 M. Die Auswertung der Sättigungswerte RUmax bei verschiedenen B4D3-Konzentrationen lieferte eine Dissoziationskonstante von 36 *10-6 M, was jedoch mit einem deutlich höheren Streuungswert Chi2 verbunden war, so dass die beiden ersteren Methoden wohl die präziseren Aussagen liefern. Abschließend wurde untersucht, ob der Inhibitor B4D3 die Aggregation direkt am Minimalmotiv (272GGKVQIINKK281) verhindert. Geringe Veränderungen im 15N,1H-HSQC-Experiment einer Probe mit t3a15N und B4D3 deuten auf eine Bindung hin und liefern ein Bindungsepitop der Peptidseite. Die gleichen Untersuchungen wurden auch an der zweiten Substanz B4A1 durchgeführt. Via STD NMR-Studien konnte das Bindungsepitop und durch Auswertung von Konzentrationsreihen die Dissoziationskonstante der Komplexbildung von B4A1 und K18 auf 52 *10-6 M bestimmt werden. Im SPR-Experiment wurden die kinetischen Parameter zu koff = 9.57 •10-3 s-1 und kon = 444 M-1s-1 bestimmt. Daraus ergibt sich eine Dissoziationskonstante von 29.3 *10-6 M. Die Auswertung der Sättigungswerte RUmax bei verschiedenen B4A1-Konzentrationen lieferte eine Dissoziationskonstante von 19 *10-6 M. Auch beim zweiten Inhibitor wurde untersucht, ob er die Aggregation direkt am Minimalmotiv (272GGKVQIINKK281) verhindert. Auch hier deuteten geringe Veränderungen der chemischen Verschiebungen im 15N,1H-HSQC-Experiment auf eine Bindung und lieferten Vorstellungen eines Bindungsepitops auf Peptidseite. Die Untersuchungen des dritten Inhibitors führten zu keiner Beschreibung einer spezifischen Wechselwirkung zum Proteinkonstrukt, da alle Protonen im STD NMR-Experiment die gleichen Signalintensitäten zeigten. Dies ist nur bei unspezifischen Wechselwirkungen mit dem Protein zu beobachten.

The formation of neurofibrillary tangles is a pathological process of Alzheimer’s disease. These tangles consist of aggregates of tau protein that therefore cannot properly stabilize microtubules and maintain neuronal transport. Consequently the neurons die of axonal undersupply. Two minimal binding motifs within the repeat regions of the protein play a significant role in forming aggregates. In the first part of this thesis the interaction of one of these peptide motives t3a (272GGKVQIINKK281) with a tau protein construct K18 consisting of the four repeat regions (MW = 13.2 kDa) is investigated. Additionally the influence of the polyanionic compound heparin on the aggregation is analyzed. Addition of heparin to a sample of peptide t3a in buffer solution immediately caused the aggregation of the peptide. Changing the order of sample preparation in first dissolving t3a followed by addition of K18 and finally heparin resulted in a homogenous phase. The binding event of the peptide in the complex induced observable changes of the signal volumes in the NOESY-spectra. Strong signal overlap prohibited the identification of sequential or long-range NOE-contacts. The binding kinetics between protein and peptide is accelerated by the presence of heparin which in turn made the binding process fast enough to allow the characterization of the system by STD NMR experiments. The binding epitope comprises amino acids Asn279, Ile277 or 278, Val275, Gln276 and Lys274, 280 or 281. After the successful synthesis and characterization of a 15N-labelled peptide t3a15N the binding epitope was described in more detail by additional data obtained by chemical shift changes in the 15N,1H-HSQC-spectra after the addition of the protein construct K18. The influence of the side chain of Asn279 on the binding event was confirmed. It was possible to show that the side chains of the three lysine residues Lys274, 280 and 281 are also involved in the binding event. The establishment of a SPR-system for the description of the complex consisting of the immobilized protein and the peptide resulted in the dissociation constant of KD = 1. 9 *10-6 M. The sensorgrams showed a negative sign, though. Dissociation of the complex was very slow. Therefore, kinetic data of the dissociation were not directly accessible. Presetting the dissociation constant obtained from the saturation values resulted in an on-rate between 801 and 69 M-1s-1. This results in an off-rate in the range of 10-3 to 10-4 s-1. This, in turn, is in agreement with the data obtained from the STD NMR spectra. Subsequently, inhibition of the aggregation of tau protein by three organic compounds was investigated. The quoted structure of B4D3 turned out to be incorrect. After determination of the correct structure the binding of the molecule to the protein construct K18 was confirmed and specified by STD NMR spectroscopy. In this context the binding epitope of the compound was assigned. Titration of the compound yielded a dissociation constant of KD = 11.3 *10-6 M. Binding was also confirmed by SPR-experiments. Kinetic analysis provided the association rate constant of 250 M-1s-1 and the dissociation rate constant of 1.2 • 10-3 s-1, resulting in a dissociation constant KD = 6.1 *10-6 M. Analysis of the amount of saturation at different concentrations of B4D3 resulted in a dissociation constant of KD = 36 *10-6 M. The latter value had significantly higher margins of error. Therefore, the dissociation constant can be assigned to KD = 8.7 *10-6 M. The inhibitor B4D3 seems to prevent aggregation by binding directly at the minimal binding motif (272GGKVQIINKK281), which could be assessed by small changes in the 15N,1H-HSQC-spectra of a sample consisting of t3a15N and B4D3. The same analysis was done with the second inhibitor B4A1. Evaluation of STD NMR-studies delivered the binding epitope and the dissociation constant of KD = 52 *10-6 M in complex with K18. The event was characterized with additional SPR-studies which yielded an association rate constant of 444 M-1s-1 and a dissociation rate constant of 9.6 • 10-3 s-1. This provided a dissociation constant of KD = 29.3 *10-6 M. The analysis of the saturation values at different concentrations of B4A1 resulted in a dissociation constant of KD = 19 *10-6 M. It was shown that the second inhibitor also prevents aggregation directly at the minimal binding motif and the binding epitope of the peptide was described by characterization of the 15N,1H-HSQC-spectra of t3a15N and B4A1. The investigation of the 3rd inhibitory compound bb-5 did not result in the description of a specific interaction with the protein construct. Here, all protons of the inhibitor showed the same signal intensities in the STD NMR spectra. This is only observed if the interaction to the protein is unspecific.
