Monoklonale Antikörper (mAbs) mit Affinität bezüglich der V3-Region des HIV-1 Glycoproteins gp120 können potentiell die Infektion humaner CD4+ Lymphozyten mit dem Virus verhindern. Dieses beruht darauf, dass die V3-Region essentiell für die Interaktion mit dem auf Makrophagen präsenten HIV-Korezeptor CCR5 ist. Da existierende anti-V3 mAbs nahezu ausschließlich Sequenzen eines b-Schleifen bildenden Motivs an der Spitze der V3-Loop erkennen und oft keine breit neutralisierende Eigenschaften aufweisen, wurde im Rahmen dieser Arbeit ein vollständig neuer Ansatz zur Selektion von V3-Antikörpern verfolgt.
Zielstruktur für einen Phage Display Ansatz war zunächst ein V3-Glycopeptid der Sequenz 291SVEINSTRPNNNTRKSIHI309 mit einer GlcNAc-GlcNAc-Glycosylierung an Position N301 (Chi), stellvertretend für ein im viralen Kontext vorkommendes komplexes Oligosaccharid. Die Selektion gegen dieses Antigen wurde in Lösung mit einer Bibliothek semisynthetischer single-chain Antikörperfragmente (scFv) humanen Ursprungs im Phagemidvektorformat durchgeführt (Griffin.1). Es gelang durch unterschiedliche Selektionsvarianten monoklonale Antikörperfragmente gegen das V3-Glycopeptidantigen zu isolieren, von denen der am besten charakterisierte Klon D26 ist. Der Selektion mit dem V3-Glycopeptid schlossen sich zusätzliche Selektionsschritte gegen das Volllängenprotein gp120 an, wobei aus diesem Ansatz Klon P2 hervorging. Die Sequenzen der scFv dieser beiden Klone und die vier weiterer spezifisch an das V3-Glycopeptid bindender Klone sind ermittelt worden.
Zur Charakterisierung der Bindungseigenschaften von isolierten scFv ausgewählter Klone wurden ELISA-Studien sowohl mit an BSA konjugierten V3-Peptiden als auch mit gp120 erfolgreich durchgeführt. In auf Oberflächenplasmonenresonanz (SPR) beruhenden Experimenten wurden für die scFv D26 und P2 Affinitäten gegenüber V3-Peptiden, einem V3-Proteinkonstrukt sowie gegenüber den gp120-Isolaten IIIB, SF162 und Bal bestimmt. Zusätzlich konnte durch SPR-Kompetitionsstudien das Epitop von P2 auf den Bereich von S296 bis I309 eingegrenzt werden. Die Methode der STD NMR Spektroskopie ermöglichte durch Titrationsexperimente mit V3-Glycopeptiden die Bestimmung von thermodynamischen Dissoziationskonstanten bezüglich von V3-Peptiden unterschiedlicher Länge (~2 µM) sowie einer atomar aufgelösten Epitopkartierung auf Seiten eines verkürzten V3-Glycopeptids bezüglich scFv P2. Bei geringen Peptidkonzentrationen traten bei der Durchführung dieser Experimente allerdings deutliche Limitierungen auf. Das Epitop von scFv P2, konnte durch STD NMR Experimente auf 298RPNN(Chi)NTRKS306 eingegrenzt werden. Zudem wurden erstmalig STD-Experimente mit scFv-präsentierenden Phagen durchgeführt, deren Ergebnisse jedoch nicht interpretiert werden konnten.
Eine Differenzierung jeglicher Klone in der Erkennung von V3-Peptid und V3-Glycopeptid stellte sich bei keinem der durchgeführten Experimente heraus. Es konnte jedoch eindeutig eine hohe Affinität gegenüber den glycosylierten gp120-Isolaten nachgewiesen werden, so dass eine Toleranz gegenüber einer komplexen Glycosylierung an N301 vorliegt. Zugleich beinhaltet das Epitop Aminosäurepositionen in direkter Umgebung zur Glycosylierungsstelle, welche subtypübergreifend größtenteils hochkonserviert vorliegen. Zusammenfassend gelang die Selektion und Charakterisierung von monoklonalen scFv-Antikörperfragmenten mit thermodynamischen Bindungskonstanten von etwa 100 nM an das virale Oberflächenprotein gp120. Aufgrund von Epitopen, die hochkonservierte Aminosäurepositionen in direkter Umgebung einer sterisch anspruchsvollen Glycosylierung beinhalten, unterscheiden sich diese deutlich von bisher veröffentlichten V3-Antikörpern.

During the entry of the human immunodeficiency virus type 1 (HIV-1) the viral envelope glycoprotein gp120 binds to CD4 molecules on host cells followed by an interaction with a coreceptor which is CCR5 on macrophages. In this secondary interaction the third hypervariable loop (V3 loop) of gp120 plays a critical role. Anti-V3 monoclonal antibodies (mAbs) provide insight into the mechanism of HIV-1 entry, are key in the development of passive immunization concepts and could promote novel vaccine approaches.
Monoclonal Abs that bind to the V3 loop are numerous while many of these bind to residues of a highly conserved motif which is responsible for a type II b turn at the V3 loop tip (310GPGR313 for subtype B). They infrequently neutralize other clades with GPGR-motif variations. Only a few mAbs that bind to the N-terminal region excluding the (QR)GPGR motif are documented of which all of them showed limited neutralizing activities. So far approaches with linear V3 peptides or fully glycosylated gp120 did not lead to broadly neutralizing mAbs binding to residues of the highly conserved region 298RPXNNTR304 which includes a complex type oligosaccharide at N301. It can be assumed that mAbs selected on linear peptides would not be able to bind these residues in the presence of the complex oligosaccharide. In the course of selections with fully glycosylated gp120 on the other hand, these conserved residues might be masked by the voluminous carbohydrate moiety. The idea of this work was to arrive at mAbs that recognize the highly conserved amino acids and that additionally tolerate the glycan moiety.
Therefore a screening of a naive phagemid vector based library of human monoclonal single-chain Fv antibody fragments (scFv) whose the CDR3 are partially randomized (Griffin.1) was performed. In contrast to conventional V3 peptide screens a synthetic N-type glycopeptide of the subtype B V3 N-terminal region (291SVEINSTRPNNNTRKSIHI309, C296S) that excluded the immunodominant 310GPGR313-motif was used as antigen. Instead of a complex glycan at N301 the chitobiosyl-moiety (Chi; GlcNAc-GlcNAc) representing the part of the glycan that is closest to the peptide backbone was used. Several selections were carried out in solution with the biotinylated V3 glycopeptide and led to the isolation of clone scFv D26. Additional rounds were performed with immobilized gp120 (clone scFv P2).
In surface plasmon resonance (SPR) experiments scFv D26 and P2 showed about 100 nM binding affinity to the V3 glycopeptide as well as to gp120 isolates IIIB, SF162 and Bal. Competition experiments with V3 peptide fragments narrowed the Epitope of scFv P2 to the region 296STRPNN(Chi)NTRKSIHI309. Besides SPR studies, saturation transfer difference (STD) NMR was used as a tool for interaction analysis of scFv P2 with different V3 glycopeptides. Because of insufficient sensitivity at low peptide concentrations these experiments proved out to be borderline. The lowest determined KD-values of proton groups where in the range of 2 µM. An epitope based on STD experiments with the V3 glycopeptide (298RPNN(Chi)NTRKS306) by relative STD-percent values as well as by KD values for individual proton groups was determined.
In all experiments none of the isolated clones showed a significant differentiation between the V3 peptide and the V3 glycopeptide. Nevertheless, because the gp120 isolates bear a complex type glycosylation at N301, the SPR-experiments revealed tolerance respective to the carbohydrate moiety. Isolated scFv bind to highly conserved residues in close proximity to the glycosylation site N301.
The scFv clones represent an optimal starting point for in vitro evolution. Furthermore these scFv could be part of new retrovaccinology approaches based on the preparatively well accessible V3 glycopeptides. The V3 glycopeptide selection strategy can also be used for a screening of HIV-1 positive patient libraries.
Summarizing this work, scFv specific for the V3 region of subtype B HIV-1 gp120 could be isolated by a simple and direct phage display approach. SPR experiments revealed binding of scFv D26 and P2 to the V3 glycopeptide as well as to gp120 isolated with thermodynamic dissociation constants of about 100 nM. A detailed epitope mapping occurred via STD NMR spectroscopy. Anti-V3 scFv are presented which bind to highly conserved residues of the 298RPNNNTRKS304 region while tolerating a complex type glycosylation at N301.
