Zusammenfassung: 
Die Störung zellulärer Proteinkinase C (PKC) Aktivität kann in vitro zu Tumorpromotion
führen und ist möglicherweise ursächlich an Karzinogeneseprozessen in vivo beteiligt. In
vorangegangenen Arbeiten wurde gezeigt, dass Fragmente des Nichtstrukturproteins 3 (NS3)
des Hepatitis C Virus (HCV) mit dem katalytischen Zentrum der PKC interagieren, wodurch
es zur Inhibition der enzymatischen Aktivität der Kinase kommt (Borowski et al. 1999). Die
Inhibition wird von einem auf der Oberfläche von Domäne 2 der NS3 NTPase/Helikase
gelegenen, argininreichen Sequenzabschnitt vermittelt, der starke Sequenzhomologie zur
autoinhibitorische Pseudosubstratregion der PKC aufweist. Diese Beobachtungen gaben
Anlass zur Frage, ob eine NS3-vermittelte PKC-Inhibition an der molekularen Pathogenese
der HCV-Infektion, insbesondere an der HCV-assoziierten Hepatokarzinogenese beteiligt ist;
ihre Aufklärung war Ziel dieser Arbeit.
Es konnte nachgewiesen werden, dass nicht nur NS3-Fragmente, sondern auch die katalytisch aktive NS3 NTPase/Helikase in vitro zu starker PKC-Inhibition führt. Die IC50-Werte
für die Inhibition der PKC katalysierten Phosphorylierung von Histon HIIIS durch verschiedene NS3 Konstrukte wurden für alle 11 PKC-Isoformen bestimmt. Sie liegen für die
NTPase/Helikase defiziente Mutante und die isolierte Domäne 2 der NS3 NTPase/Helikase,
für die meisten Isoformen, im submikromolaren Bereich.
Die PKC-Inhibition kommt unabhängig von der enzymatischen NTPase-Aktivität
zustande, wie anhand einer NTPase/Helikase defizienten Mutante und durch den Einsatz
eines ATP-regenerierenden Systems im PKC-Assay demonstriert werden konnte. Die
kinetische Analyse der PKC-NS3-Wechselwirkung ergab, dass die Inhibition im Modus einer
mixed type Hemmung erfolgt und hauptsächlich aus der spezifischen Interaktion von
Domäne 2 der NS3 NTPase/Helikase mit dem aktiven Zentrum der PKC resultiert.
Um die biologische Relevanz dieser Ergebnisse in intakten Zellen zu überprüfen, sowie
generelle onkogene Einflüsse von der NS3 NTPase/Helikase zu untersuchen, wurden
NIH 3T3 Zellen mit stabiler Expression von NS3-Konstrukten generiert. Zur Bestimmung
von Einflüssen der rekombinanten Proteine auf ihre intrazelluläre PKC-Aktivität wurde der
Phosphateinbau in das PKC-Substratprotein p80/MARCKS in diesen Zellen nach Stimulation
mit dem PKC-Aktivator TPA (12-O-Tetradecanoylphorbol-13-Acetat) gemessen. Die NS3-Expression hatte hier keinen signifikanten Einfluss; der gemessene Parameter kann jedoch nur
als grober Indikator für die intrazelluläre Gesamtaktivität der meisten PKC-Isoformen
angesehen werden, so dass nicht auszuschließen ist, dass die PKC-Inhibition durch NS3 eine
Rolle in der Pathogenese der HCV-Infektion spielt.
Zur Prüfung möglicher onkogener Effekte durch die NS3 NTPase/Helikase wurden verschiedene zelluläre Eigenschaften transfizierter Zellen untersucht, die im Zusammenhang mit
neoplastischer Transformation stehen. Stabil mit der NS3 NTPase/Helikase transfizierte
Zellen besitzen keine Koloniebildungsfähigkeit in Soft Agar und auch keine erhöhte DNA-Syntheserate bei Konfluenz. Demzufolge weist die NS3 NTPase/Helikase keine direkt transformierenden Eigenschaften auf. Zur Untersuchung tumorpromovierender Effekte durch die
NS3 NTPase/Helikase kamen zwei verschiedene Zellkulturmodelle zum Einsatz. Die Untersuchung transient transfizierter Zellen ergab, dass die NS3 NTPase/Helikase weder in
JB6-Cl41-5a-Zellen, noch in c-Src überexprimierenden 3Y1-Rattenfibroblasten eine
gesteigerte Koloniebildungsfähigkeit in Soft Agar bewirkt. Die erstgenannte Zelllinie stellt ein
gut etabliertes Modell für die Untersuchung von tumorpromoterinduzierter neoplastischer
Transformation in vitro dar (Colburn et al. 1980), letztere lässt sich durch Inhibition von
PKC-delta zu matrixunabhängiger Proliferation in Soft Agar anregen (Lu et al. 1997). Folglich
verursacht die NS3 NTPase/Helikase keine Tumorpromotion und die Ausgangshypothese,
nach der sie durch Störung von PKC-Funktionen tumorpromovierend wirkt, wurde nicht
bestätigt.
Wie in dieser Arbeit erstmalig gezeigt wurde, führt die NS3 NTPase/Helikase jedoch zu
Morphologieveränderungen und einer starken Steigerung der Zellproliferation. Diese Einflüsse kommen allerdings unabhängig von einer Inhibition der katalytischen Aktivität von
Kinasen der PKC-Familie zustande. Derartige phänotypische Veränderungen werden häufig
im Verlauf der Induktion eines Tumorphänotyps beobachtet. Bei der sehr langfristigen, chronischen Einwirkung von NS3 auf Hepatocyten im Verlauf einer chronischen Hepatitis C ist es
durchaus wahrscheinlich, dass proliferationsfördernde Faktoren maßgeblich an der Karzinogenese beteiligt sind.

A broad range of biological and biochemical studies document that disturbing protein
kinase C (PKC) function induces tumor promotion and may promote carcinogenesis in vivo.
In foregoing works it has been demonstrated that fragments of the NTPase/helicase domain of
the nonstructural protein 3 (NS3) of hepatitis C virus (HCV) interact with the active site of
PKC and reduce catalytic activity of the enzyme (Borowski et al. 1999). The inhibition is
mainly mediated by a short arginine rich amino acid stretch localized on the surface of
Domain 2 of the NS3-NTPase/helicase. The HCV amino acid sequence of this motif strongly
resembles the pseudosubstrate sequence within the autoregulatory domain of PKC. Thus,
NS3-mediated PKC-inhibition may be involved in the molecular pathogenesis of the HCV infection, particularly in regard to HCV-associated carcinogenesis leading to hepatocellular
carcinoma. The presented thesis sets out to scrutinize this hypothesis.
The acquired data clearly demonstrates, that not only fragments of NS3, but catalytically
active NS3-NTPase/helicase acts as a potent PKC-inhibitor in vitro. IC50-values for the
inhibition of PKC-catalyzed Histone HIIIS phosphorylation by an NTPase/helicase-inactive
mutant (NS3h-D1316A) or the isolated domain 2 of the NS3-NTPase/helicase were determined for all 11 PKC-isoforms. They fall within the submicromolar range for the inhibition
of most PKC-isoforms. A PKC-activity assay comprising an ATP-regenerating System was
applied to confirm these results for wildtype NS3-NTPase/helicase and consequently rule out
influences of the D1316A point mutation on the accessibility of the arginine rich stretch.
PKC-inhibition by NS3h-D1316A displays a mixed type kinetic pattern and mainly results
from a specific interaction between domain 2 and the PKC active site.
To elucidate the biological relevance of these findings in intact cells, NIH 3T3 cells with
stable expression of the NS3-constructs were generated. These cells were subjected to an assay
for intracellular PKC-activity, that employed measuring phosphorylation of the PKC-substrate p80/MARCKS after stimulation with the PKC-activator TPA (12-O-Tetradecanoylphorbol-13-acetate). The NS3-expression had no significant effect in this assay. However, the
possibility that PKC inhibition plays an important role in HCV pathogenesis can not be ruled
out, as the employed method must be regarded as a rough measurement of the overall activity
of most PKC isoforms. To scrutinize possible oncogenic effects caused by the NS3-NTPase/helicase, NS3-transfected cells were assayed for several cellular properties that are commonly associated with
neoplastic transformation. Stably transfecting NIH 3T3 cells with NS3-constructs did not
confer the ability to form colonies in soft agar and did not lead to an elevated DNA-synthesis
rate in cells that were grown to confluence. Thus NS3-NTPase/helicase does not exhibit
directly transforming properties. To address the question if the NS3-NTPase/helicase exerts
tumorpromoting effects, two different cell culture models were utilized. The determination of
colony fomation of JB6-Cl41-5a cells is commonly used to assay general tumorpromoting
effects in vitro, whereas in c-Src overexpressing 3Y1 cells, anchorage independent growth can
specifically be induced by inhibition of PKC-delta activity (Lu et al. 1997). Transient transfection
with NS3-constructs did not confer the ability to form colonies in softagar to neither of the
two cell lines. These findings strongly suggest that NS3-NTPase/helicase does not cause
tumorpromotion.
Nonetheless it was shown, that NS3-NTPase/helicase expression in NIH 3T3 cells causes
morphological alterations and stongly stimulates cell proliferation, albeit independently from
PKC inhibition. To date, this is the first report that attributes the induction of these
phenotypic changes to the NTPase/helicase portion of HCV-NS3. Proliferative and morphological changes are often observed in the course of neoplastic transformation. Considering the
long term exposure of hepatocytes to NS3 during chronic hepatitis C, it seems eminently
reasonable, that proliferation promoting factors substantially contribute to HCV-associated
hepatocarcinogenesis.
