Zusammenfassung: 
Die Resistenzbildung von Neuroblastomzellen gegen Oxiradikalbildende Zytostatika ist ein ungelöstes Problem und verantwortlich für die weiterhin unbefriedigende Prognose von Hochrisiko-Neuroblastomerkrankungen. Vor diesem Hintergund war das Ziel dieser Arbeit, die Frage zu klären, ob BSO und EA in der Lage sind über die Modulation des GSH-GSSG-Redox-Systems die Empfindlichkeit von Tumorzellen gegen Oxyradikalbildner zu erhöhen und somit deren Wirksamkeit in-vitro an der Neuroblastomzelllinie LA-N-1 zu verbessern.
Als oxyradikalbildende Zytostatika wurden Doxorubicin, Cisplatin, Melphalan und Arsentrioxid untersucht. Zusammenfassend ergaben sich folgende Befunde:
Die Kombination der von uns getesteten Chemotherapeutika mit BSO an LA-N-1-Zellen steigert die Wirksamkeit von Cisplatin, Melphalan und insbesondere As2O3.
Die Kombination mit EA steigert die Effektivität von Doxorubicin, Melphalan und Cisplatin. Die Zytotoxizität von As2O3 wird durch Zugabe von EA abgeschwächt.
Die Inkubation mit BSO + EA verstärkt den Wirkmechanismus von As2O3 über einen addierenden Effekt der Einzelwirkungen hinaus.
Auf die Wirksamkeit von As2O3 als direktem Radikalbildner mit drei Sauerstoffatomen in der Verbindung haben sowohl BSO als auch EA ausgeprägtere Auswirkungen als auf die anderen getesteten Zytostatika.
Die in-vitro-Befunde an der Neuroblastomzelllinie LA-N-1 weisen auf eine Einsatzmöglichkeit von As2O3 alleine oder in Kombination mit BSO und EA zur Behandlung des Neuroblastoms hin.
Die erwiesene Wirksamkeit von Doxorubicin, Cisplatin und Melphalan in der Therapie des Neuroblastoms könnte durch die kombinierte Gabe mit EA profitieren, wenn auch nur im Grenzbereich hoher klinisch einsetzbarer EA- Konzentrationen. Hier betrug die maximale Wirkungssteigerung durch die kombinierte Gabe mit EA im Vergleich zur Einzelgabe bis zu 100 % für Doxorubicin, 75 % für Cisplatin und bis zu 55 % für Melphalan.
Aufgrund der erhobenen Befunde ergibt sich die Notwendigkeit für weitere in-vitro- und in-vivo-Untersuchungen zur optimalen Anwendung von Arsentrioxid in der Behandlung des Neuroblastoms.

The growth of chemical resistant neuroblastoma cells against radical-forming cytostatic drugs still is an unsolved problem. It is responsible for the persistent dissatisfying prognosis of high-risc neuroblastoma diseases. On this backround, the aim of this work was to clearify, if BSO and EA are capable of affecting the sensibility of tumor cells against radical-forming agents by modulating the GSH-GSSG-redox system, and thus, to improve the potency against the neuroblastoma cell line LA-N-1 in-vitro.
Doxorubicin, cisplatin, melphalan, and arsenic trioxide were investigated as radical-forming cytostatics. In summary, the following investigations were made:
The combination of the tested chemotherapeutics with BSO on LA-N-1 cells could increase the potency of cisplatin, melphalan, and in particular As2O3.
The combination with EA improves the potency of doxorubicin, melphalan and cisplatin. The cytotoxity of As2O3 decreases in the combination with EA.
The incubation with both, BSO and EA, increases the potency of As2O3 exceeding the sum of the single effect of each chemosensitizer alone.
BSO and EA both have a stronger effect on As2O3 as a radical-forming drug with three oxygen atoms in its compound than on any other cytostatic drug tested.
The in-vitro results at the neuroblastoma cell line LA-N-1 indicate a possible use of As2O3 alone or in combination with BSO and EA in the treatment of the neuroblastoma.
The approved effectiveness of doxorubicin, cisplatin, and melphalan in the therapy of the neuroblastoma can possibly improve in combination with EA but only at clinical maximum dose rates. Here, the maximum increase of the effect through combination with EA in comparison to the effect of the cytostatic alone was up to 100% for doxorubicin, 75% for cisplatin, and up to 55% for melphalan.
Because of the present results, further in-vitro and in-vivo investigation has to be performed to observe the optimal use of arsenic trioxide in the treatment of the neuroblastoma.
