Die vorliegende Arbeit hatte zum Ziel, Kenntnisse über die Carcinogenität und Mutagenität der bis dato wenig untersuchten Arylamin-Addukte von 2' Desoxyadenosin zu erlangen. Hierbei sollte ein besonderes Augenmerk auf die möglichen Unterschiede zwischen Grenzcarcinogenen und starken Carcinogenen gelegt werden. Aus diesem Grund war eine synthetische Erschließung der Arylamin-Addukte nötig, um sie im weiteren Verlauf in sequenzspezifisch modifizierte Oligonucleotide einzubauen. Die Arbeit gliederte sich in vier Teilgebiete: die Synthese C8- und N6-Arylamin-modifizierter Phosphoramidite, die Darstellung von C8-Arylamin-modifizierten-5'-Monophosphaten zur Konformationsanalyse und die Synthese, Charakterisierung und Untersuchung von sequenzspezifisch modifizierten Oligonucleotiden.
Die Synthese C8-Arylamin-modifizierter Addukte von 2'-Desoxyadenosin konnte in dieser Arbeit erfolgreich durchgeführt und verbessert werden. Den Schlüsselschritt der Synthese stellte dabei die Darstellung der Addukte über eine Palladium-katalysierte Kreuzkupplung dar. Es konnten unter Verwendung von 10 mol% Pd2(dba)3 und 30 mol% rac. BINAP und einer Vorreaktion des Katalysators und des Liganden Ausbeuten zwischen 67 und 80% erreicht werden. Um diese Addukte in der automatisierten Festphasensynthese von Oligonucleotiden einsetzen zu können, war eine orthogonale Schutzgruppenstrategie nötig. Die C8-Arylamin-modifizierten Phosphoramidite konnten in Gesamtausbeuten zwischen 17 und 26% erzielt werden. Die synthetische Route zu den N6 Addukten, die eine freie Aminofunktion in der ortho- bzw. para-Position tragen, unterscheidet sich von der der C8-Addukte. Als optimale Reaktionsbedingungen zur Darstellung derselbigen hat sich die Verwendung von 10 mol% Pd2(dba)3, 15 mol% Xantphos, TBDMS dA, dem entsprechenden Nitroarylbromid und Cs2CO3 als Base in Dimethoxyethan herausgestellt. Wichtig war hierbei der Einsatz des Nucleosides im Überschuss (1.3 Äq.), um eine doppelte Arylierung der Aminofunktion zu verhindern. Die N6 Addukte konnten in Ausbeuten bis 98% erhalten werden. Die bisher synthetisch unerschlossenen Phosphoramidite konnten erhalten werden, indem die Aminofunktion bis nach erfolgter DNA-Synthese als Nitrofunktion vorlag und erst dann mit Zink in einem Citrat-Puffer reduziert wurde. Die Synthese der C8-Arylamin-modifizierten Monophosphate zur Konformationsanalyse wurde über die Methode nach SOWA und OUCHI realisiert. Die Auswertung der NOESY-Spektren zeigte weder für die freien Addukte noch für die C8-Arylamin-modifizierten Monophosphate eine Verschiebung des Konformationsgleichgewichtes von anti zu syn an.
Die C8- und N6-Arylamin-modifizierten sequenzspezifischen Oligonucleotide wurden sequenzspezifisch in drei Sequenzen eingebaut. Die EcoRI-Oligonucleotide wurden einem Restriktionsabbau durch das Enzym EcoRI unterzogen um den Einfluss der Arylamin-Modifikationen zu untersuchen. Es zeigte sich, dass eine Arylamin-Modifikation direkt an der Schnittstelle bzw. eine Base davon entfernt, einen so erheblichen Störfaktor darstellte, dass das Enzym nicht mehr mit dem Doppelstrang interagieren konnte und somit kein Abbau erfolgte. Befand sich die Modifikation außerhalb der Erkennungssequenz aber dennoch in der Nähe der Schnittstelle, so kam es zu einem leicht verlangsamten Abbau durch das Enzym. Der Schmelzpunkt der DNA ist ein Indikator für die Stabilität des Doppelstrangs und kann somit etwas über den Einfluss von Modifikationen aussagen. Den größten Verlust an thermischer Stabilität konnte bei den N6-Arylamin-modifizierten EcoRI-Oligonucleotides mit Schmelzpunkten von weniger als 25 °C im Vergleich zum unmodifizierten Doppelstrangs mit 42 °C beobachtet werden. Zur genaueren Untersuchung des Einflusses von Arylamin-Modifikationen wurde die NarI-Sequnez ausgewählt. Neben den Arylamin-modifizierten Oligonucleotiden wurden drei mismatch-Oligonucleotide synthetisiert sowie ein ABASIC-Oligonucleotid, welches an der eigentlichen Position der Modifikation gar keine Base enthält. Die Ergebnisse zeigten, dass nicht nur Watson-Crick Wasserstoffbrücken zur Stabilität des Doppelstrangs beitragen sondern ebenso pi-stacking Wechselwirkungen, so dass Arylamin-Modifiaktionen den Doppelstrang durch sterische Hinderung nicht nur destabilisieren sondern auch durch pi-stacking stabilisieren.
DNA-Addukte können die Selektivität und Wirkungsweise von DNA-Polymerasen beeinträchtigen können. Aus diesem Grund wurden in Kooperation mit Prof A. MARX von der Universität Konstanz C8-Arylamin-modifizierte Oligonucleotide in Primer-Verlängerungsstudien als Template eingesetzt. Untersucht wurden die Polymerasen Pyrococcus pfuriosus (Pfu) DNA Polymerase (exo-), Polymerase PolB) und Dpo4. Allgemein lässt sich für das starke Carcinogen 4-Aminobiphenyl ein stärkerer Einfluss hinsichtlich Replikationsabbruch und Fehleinbau von Nucleotiden feststellen als es beim Grenzcarcinogen p-Anisidin der Fall war.

The herein presented work had the aim to gain knowledge of the carcinogenesis and mutagenesis of aryl amine adducts of 2'-deoxyadenosine with special regards concerning the carcinogenic and mutagenic potential of borderline and strong carcinogens. For this reason a synthetic route towards this adducts and the site-specific incorporation of the corresponding phosphoramidites into oligonucleotides is a prerequisite.
The work was divided into four sections: The synthesis of both C8-dA-aryl amine adducts and N6-dA-aryl amine adducts, the synthesis of C8-dA-aryl amine modified 5' monophosphates for conformational analysis and the synthesis and investigation of C8 and N6-dA-aryl amine site-specifically modified oligonucleotides.
The synthesis of the C8-aryl amine modified dA-adducts and their corresponding phosphoramidites could successfully be accomplished and optimized. The key step of the synthesis was a palladium catalyzed cross coupling, the so called Buchwald-Hartwig coupling. Under usage of 10 mol% Pd2(dba)3, 30 mol% racemic BINAP, silyl protected dA, aryl amine and cesium carbonate as base the C8-adducts were obtained in yields between 67% and 80%. In order to apply these adducts in the automated solid phase synthesis of oligonucleotides an orthogonal protection group strategy was necessary. The C8-arylamine modified phosphoramidites, suitably protected for the automated solid phase synthesis of oligonucleotides, could be obtained in seven steps out of 2’ deoxyadenosine in overall yields from 17% to 26%.
The synthetic route towards the N6-adducts, bearing a free amino function at the aromatic ring of the aryl amine in the ortho- or para-position, is quite different. The usage of 10 mol% Xantphos and 15 mol% Pd2(dba)3 as catalytic system with a nitroarylbromide and 3',5'-di-TBDMS-dA turned out to be the best conditions for the synthesis of N6-aryl amine modified adducts of 2'-dA. The corresponding phosphoramidites, that have not been synthesized so far, could be obtained by keeping the ortho- respectively para-amino function as a nitro function until after DNA synthesis and a following reduction with zinc in a citrate buffer.
The C8-dA-aryl amine modified 5'-monophosphates could be synthesized according to the method of SOWA and OUCHI. NOE-analysis of the modified nucleotides as well as of the adducts with free 3'- and 5'-OH group showed no indication for a conformational change regarding the glycosidic bond from anti to syn.
The C8- und N6-modified phosphoramidites were site-specifically incorporated into three sequences by means of the automated solid phase synthesis of oligonucleotides. The EcoRI-oligonucleotides containing a recognition site for the EcoRI-enzyme were submitted to an enzymatic restriction to investigate the effects of the different aryl amine modifications. It turned out that the modifications situated in the recognition site and at the cleavage site impair the activity of the enzyme completely. The same results were obtained for modifications located inside the recognition site but not directly at the celavage site. Still a decreased activity was observed for modifications in a few bases distance from the recognition site of the enzyme. The melting point of DNA is an indication for the stability of the double strand and therefore can be used to estimate the impact of modifications. The highest decrease in thermal stability could be observed for the N6-aryl amine modified EcoRI-oligonucleotides with melting points less than 25 °C in comparison to the unmodified double strand with 42 °C. For a better investigation of the impact of arylamine modifications on the stability of the duplexes the NarI sequence was chosen. Beside the aryl amine modified oligonucleotides several mismatch oligonucleotides as well as an oligonucleotide with an abasic site were synthesized. The results showed that not only the Watson-Crick hydrogen bonds contribute to the stability of the double strand it as well is the pi-stacking interactions. As a result an arylamine modification does not only destabilize the double strand based on the steric hindrance and in case of the N6-modification the loss of one hydrogen bond it also gives a certain stability due to pi-stacking interactions.
It is known that DNA-adducts can hamper the selectivity and efficiency of lesion bypass synthesis by replicative DNA polymerases, while other DNA polymerases are effective in performing DNA synthesis beyond the site of damage. In cooperation with Prof. A. MARX from the University of Constance modified 30mers were used as templates in primer-extension assay to study the effects of C8-arylamine modifications on several DNA-polymerases. It was found that the strong carcinogen modification has a higher impact on replication error and misinsertion of nucleotides as it was found for the borderline carcinogen modification.
