Die vorliegende Arbeit behandelt die Darstellung von carbocyclischen Nucleosiden in Hinblick auf eine antivirale Anwendung gegenüber ausgewählten Viruserkrankungen. Ein besonderes Augenmerk wurde dabei auf die Synthese von carbocyclischen L-Nucleosiden gelegt. Gerade bei der Behandlung von Hepatitis B finden zahlreiche nucleosidische Wirkstoffe in der L-Konfiguration Anwendung. Dabei handelt es sich um die Nucleosidanaloga, die in ihrer Glyconkonfiguration von der L-Furanose abgeleitet werden. L-Nucleoside sind die Enantiomere der natürlich vorkommenden D-Nucleoside. Das Ziel war es, das Konzept der L-Nucleoside aufzugreifen und mit den Vorteilen von carbocyclischen Nucleosiden zu verbinden. So wurden neue, konvergente Syntheserouten zu enantiomerenreinen, carbocyclischen L-Nucleosiden entwickelt, die es ermöglichen, durch ihre Allgemeingültigkeit zahlreiche potentiell aktive carbocyclische Nucleosidderivate zu generieren.
Die Darstellung von carbocyclischen L-2’-Desoxynucleosiden erfolgte unter Anwendung eines konvergenten Syntheseschemas, bei dem ein entsprechend funktionalisiertes Cyclopentanolderivat mit Nucleobasen verknüpft wurde. Es wurde gezeigt, dass unterschiedlichste Heterocyclen unter Mitsunobu-Bedingungen an das Cyclopentanolgerüst gekuppelt werden können. Neben der Verwendung der gängigen Nucleobasen zur erfolgreichen Darstellung von L-carba-dT, L-carba-dU,
L-carba-dC, L-carba-dA oder L-carba-dG wurden auch ungewöhnliche Nucleobasen eingesetzt, um die Vielseitigkeit dieser Syntheseroute zu verdeutlichen. So wurden erstmalig biologisch viel versprechende Modifikationen von L-carba-dC synthetisiert, halogenierte Derivate wie L-carba-FdU, L-carba-IdU oder L-carba-BVDU erhalten und mit der Darstellung von L-carba-2’-Desoxyribavirin gezeigt, dass auch Heterocyclen eingesetzt werden können, die von Pyrimidin- bzw. Purinstrukturen abweichen.
Der Schwerpunkt dieser Arbeit lag auf der Entwicklung und Erprobung von neuen Synthesewegen zur Darstellung von hochfunktionalisierten, carbocyclischen Grundgerüsten für eine Anwendung in einem konvergenten Synthesekonzept.
Neben Untersuchung der Synthese von carbocyclischen 2’,3’-Didehydro-2’,3’-didesoxy-Nucleosiden (d4-Nucleosiden) wurden in dieser Arbeit auch die Modifikationen geeigneter d4-Grundkörper durchgeführt. Durch Cyclopropanierung der Doppelbindung wurden bicyclische Cyclohexangrundgerüste synthetisiert, die je nach Konfiguration der Hydroxyfunktion eine endo- oder exo-Konfiguration des annelierten Cyclopropanrests steuern. Es wurden erstmalig L-carba-2’,3’-endo-Methylen-beta-thymidin und L-carba-2’,3’-endo-Methylen-alpha-thymidin synthetisiert. Dabei handelt es sich um eine besonders interessante Klasse von Nucleosiden, da über die Konfiguration des bicyclischen Systems Vorzugskonformationen des Glycons eingestellt werden können, was einzigartige biologische Aktivitäten mit sich bringen kann.
Des Weiteren wurde in dieser Arbeit der synthetische Zugang zu carbocyclischen Ribonucleosiden untersucht. Dazu wurde ein entsprechender Cyclopentanolbaustein für einen konvergenten Syntheseansatz entwickelt, der sowohl an der 2’- und 3’-Position die erforderlichen OH-Gruppen mit sich bringt. Nach Mitsunobu-Kupplung und Abspaltung der Schutzgruppen wurde so erfolgreich L-carba-Ribothymdin erhalten.
Neben den carbocyclischen L-Nucleosiden gibt es auch interessante Eigenschaften bei den D-Derivaten. Nach genauerer Untersuchung des anti-HIV aktiven carbocyclischen Nucleosids D-carba-dT zeigte sich, dass es sich hierbei sowohl um ein Substrat für die Thymidinkinase als auch in seiner Triphosphatform für die HIV-RT handelt. Es wird zwar langsamer als sein natürlicher Konkurrent D-Thymidin phosphoryliert, zeigt aber dafür einen neuen, wirksamen Mechanismus bei der Inhibition der viralen Transkription. Für weiterführende Untersuchungen der antiviralen Aktivität wurde ein in dieser Arbeitsgruppe entwickeltes Prodrugsystem angewandt. Im Rahmen dieser Arbeit wurde die Synthese von D-carba-dT in Bezug auf die Schutzgruppenstrategie so angepasst, dass erstmalig mit BAB-D-carba-dTDP ein potentielles Diphosphatprodrug eines antiviral aktiven carbocyclischen Nucleosids synthetisiert werden konnte.

This thesis deals with the syntheses of carbocyclic nucleosides analogues as drugs for an application in the treatment of certain viral diseases. A special emphasis herein is the synthesis of carbocyclic L-nucleosides. Especially in the treatment of hepatitis B there are used numerous nucleosidic drugs in the L-configuration. These are nucleoside analogues which glycon configuration is derived from L-furanose.
L-nucleosides are the enantiomers of the naturally occurring D-derivatives. The intention of this thesis is the combination of the L-nucleoside concept with the advantages of carbocyclic nucleosides. Thus there were developed new convergent synthetic ways to enantiomerically pure carbocyclic L-nucleoside analogues. Due to their proven generality these routes allow syntheses of various potentially active carbocyclic nucleoside derivatives.
The preparation of those carbocyclic L-2’-deoxynucleosides occurred by application of a convergent synthetic strategy in which a functionalized cyclopentanol derivative is condensed with a nucleobase. It was shown that several hetero cycles could be coupled to a cyclopentanol moiety according to a Mitsunobu protocol.
Beside the usage of usual nucleobases for the successful synthesis of L-carba-dT, L-carba-dU, L-carba-dC, L-carba-dA or L-carba-dG the strategy was applied to various nucleobases to demonstrate the flexibility of this route. In this way there were synthesized promising biological active modifications of L-carba-dC, halogenated derivatives like L-carba-FdU, L-carba-IdU or L-carba-BVDU. Within the preparation of von L-carba-2’-deoxyribavirin it was shown that it is possible to use uncommon hetero cycles, which differ from pyrimidines or purines, respectively.
The focus of this work was the development and testing of new synthetic routes for the preparation of highly functionalized carbocyclic moieties for the application in a convergent synthetic strategy.

In addition to the study of the syntheses of carbocyclic 2’,3’-Didehydro-2’,3’-dideoxy-nucleosides (d4-nucleosides) there were generated several modifications of applicable d4-moieties. Via cyclopropanation of the double bond there were synthesized bicyclic cyclohexane derivatives, which built up an endo- or exo-configuration of the annulated cyclopropane residue subject to stereogenic centre bearing the hydroxyl function. The first time synthesis of L-carba-2’,3’-endo-methylene-beta-thymidine und L-carba-2’,3’-endo-methylene-alpha-thymidine led to notable interesting class of nucleosides, due to their controllable configuration of the glycon’s bicyclic system. This possibility may affect unique potential biological activity.
Moreover this work dealt with the investigation of synthetic access to carbocyclic ribonucleosides. For that purpose it was developed a certain cyclopetanol building block for the application in a convergent synthesis strategy, which bore the needed OH-group at the 2’- and 3’-position. After Mitsunobu coupling and cleavage of the protecting groups L-carba-Ribothymdine could successfully be obtained.
A precise research into the anti-HIV active carbocyclic nucleoside D-carba-dT showed that it is a substrate for the thymidine kinase as well as a substrate in its triphosphate form for the HIV-reverse transcriptase. It is indeed phosporylated even slower as its natural counterpart D-thymidine, but therefore it displayed a novel effective mechanism of inihibition of the viral transcription. For further investigations of the antiviral activity it was applied a prodrug strategy which was developed in this group. As part of this work the synthesis of D-carba-dT was adapted concerning the protective groups. Following this route BAB-D-carba-dTDP, a potential diphosphate prodrug of an antiviral active carbocyclic nucleoside could be yielded for the first time.
