Zusammenfassung: 
Natürliche Vanadat-abhängige Bromoperoxidasen aus marinen Algen enthalten in den periphären Bereichen der Proteinmatrix partiell bromierte Tyrosylreste. Im aktiven Zentrum solcher Peroxidasen, die mit Bromid versetzt wurden, werden Brom-Vanadium-Abstände zwischen 3.6 und 4.1 Å gefunden. EXAFS-Untersuchungen ergaben einen Brom-Kohlenstoffabstand von 1.88-1.91 Å. Das entspricht dem, was für eine Bindung von Br an einen sp2-hybridierten (aromatischen) Kohlenstoff erwartet wird, erfordert allerdings einen elektrophilen Angriff des Broms am Aromaten, der in Abwesenheit von Peroxid aber nicht erfolgen kann. Mit den in dieser Arbeit durchgeführten Untersuchungen an Komplexen des Vanadiums mit Liganden, die Brom an aromatischem sp2- und aliphatischem sp3-Kohlenstoff gebunden enthalten, sollten die Abstände V-Br und C-Br modelliert und damit plausible Aussagen zur Bindung des Substrats Bromid im aktiven Zentrum der Enzyme gemacht werden.
In Abwesenheit von Peroxid kann Bromid nur im Sinne eines nukleophilen Angriffs an ein Kohlenstoffatom geknüpft werden. Ein plausibles Ziel ist Serin, das sich in unmittelbarer Nähe des Reaktionszentrums des Enzyms befindet; die Substitution der alkoholischen Funktion des Serins durch Brom führte dann zu Bromalanin. Zur Modellierung dieses Vorganges wurde nun Bromalanin- methylester (Brala) in einer Mehrstufensynthese dargestellt. Der kationische Ester konnte durch Kristall- strukturanalyse charakterisiert werden. Der Abstand Br C(sp3) beträgt 1.944 Å.
Mit dem Coliganden Salicylaldehyd (sal) entsteht VO2+-templiert ein Schiffbase-Ligand (Brala-sal), der an VO2+ koordiniert. Da für den so generierten Oxidovanadium(IV)-Komplex [VO(Brala-sal)THF] weder Röntgenbeugungs- noch Röntgenabsorptionsdaten vorlagen, wurde ein relativ neues Verfahren zur vorläufigen Strukturermittlung derartiger Komplexe eingesetzt: Mit Hilfe von DFT-Rechnungen wird für den paramagnetischen (d1) Vanadiumkomplex eine optimierte Struktur berechnet. Als Qualitätskriterium für diese Struktur dient der Vergleich der gemessenen ESR-spektroskopischen Hyperfeinkopplungskonstanten mit denen, die unter Zugrundelegung der berechneten Struktur erhalten werden. Die in der Peroxidase gefundenen Brom-Vanadium-Abstände konnten mit der so generierten Struktur des Komplexes [VO(brala-sal)THF] allerdings nicht modelliert werden. Lässt man aber im aktiven Zentrum des Proteins Deformationen des Diederwinkels Br C C N im Bromalanin zu, so lassen sich die experimentellen V-Br Abstände durchaus modellieren. Die Energie, die erforderlich ist, um eine gespannte Konformation im Komplex einzustellen, die den Abstand V-Br in der Peroxidase widerspiegelt liegt bei 32 kJ/mol.
Während also mit aliphatisch gebundenem Brom eine Modellierung des Brom-Vanadium-Abstandes möglich ist, gelingt dies mit aromatisch gebundenem Brom nicht. Es wurden zahlreiche Komplexe mit Brom in den möglichen Positionen am Aromaten mit unterschiedlichen Aminkomponenten untersucht. Der kleinstmögliche Abstand von ca. 4.9 Å wurde hier im Oxidovanadiumkomplex Gly-3-Br-THF-VO gefunden.
Zur Validierung des Verfahrens wurden zahlreiche berechnete Strukturen mit denen aus Röntgenbeugungsexperimenten verglichen. Die Abweichungen lagen sowohl bei den Bindungslängen wie –winkeln im Marginalbereich. Ebenso lagen die Abweichungen zwischen berechneten und gemessenen Hyperfeinkopplungs- konstanten unter einem Prozent, sodass die mit diesem Verfahren erzielten Ergebnisse als hinreichend abgesichert gelten dürfen.

Naturally occurring vanadate-dependent bromoperoxidases from marine algae contain partially brominated tyrosyl residues in the peripheral regions of the protein matrix. In the active centre of these peroxidases, treated with bromide, bromine-vanadium distances between 3.6 and 4.1 Å have been found. EXAFS investigations revealed bromine-carbon distances between 1.88 and 1.91 Å. This corresponds to bromine bonded to a sp2 hybridised (aromatic) carbon, which requires an electrophilic attack of bromine to the aromatic system, a reaction which only occurs in the presence of peroxide. The present investigations were intended to model the experimentally detected V-Br and V-C bond lengths and, thus, to arrive at plausible information on the binding of the substrate bromide to the active centre. To this effect, vanadium complexes were synthesised with ligands containing bromine bonded to sp2 and sp3 hybridised carbon.
In the absence of peroxide, bromide can be bound to a carbon only by nucleophilic attack. A plausible target for such an attack is serine, which resides directly in the proximity of the enzyme’s reaction centre. Substitution of the alcoholic function of serine leads to the formation of bromoalanine. In order to model this process, the methyl ester of bromoalanine (Brala) was synthesised by multi-step synthesis. The cationic ester could be characterised by X-ray diffraction analysis. The distance Br-C (sp3) is 1.944 Å. In the presence of salicylaldehyde (sal) as co-ligand and VO2+ as a template, the Schiff base ligand Brala-sal forms, which is coordinated by VO2+. Since the structure of the oxidovanadium(IV) complex thus generated could not be obtained by X-ray diffraction or absorption, a comparatively novel procedure for the provisional structure elucidation was applied: by means of DFT calculations, an optimised structure is calculated for a paramagnetic (d1) vanadium complex. As a criterion for the quality of this structure, EPR-spectroscopic hyperfine coupling constants obtained on the basis of the calculated structure are compared with those from the experimental spectrum. The structure thus generated for [VO(Brala-sal)THF] did not model the bromine vanadium distances found in the peroxidase, though. However, if the dihedral angle Br-C-C-N in the bromoalanine moiety is subjected to some deformation, the experimental V-Br distances are obtained. The energy afforded to arrive at a strained conformation of the complex reflecting the V-Br distance amounts to 32kJ/mol.
While being able to model the experimental V-Br distances by aliphatic bromine as part of the ligand, this modelling does not work with aromatic bromine. Numerous complexes containing bromine in various positions of the aromatic system and different amino components have been investigated. The smallest distance was found in the complex Gly-3-Br-THF-VO with 4.9 Å.
In order to prove the reliability of this procedure of structure elucidation, calculated and measured EPR-parameters were, on the one hand, compared for all complexes showing negligible differences. On the other hand, calculated structures were checked against structures obtained from X-ray diffraction; see, for instance, X-ray and calculated structures of an aminophenol complex with vanadium as a chiral center.
