Zusammenfassung: 
Die GlcNAc-1-Phosphotransferase ist das Schlüsselenzym bei der Bildung des Mannose-6-Phosphatrestes an Zuckerketten von löslichen lysosomalen Proteinen, der das Signal für den rezeptorabhängigen Transport dieser Proteine zum Lysosom darstellt. Der hexamere GlcNAc-1-Phosphotransferase-Komplex besteht aus drei verschiedenen Untereinheiten (α2β2γ2), die durch zwei Gene kodiert werden. Defekte im Gen für das α/β Vorstufenprotein der GlcNAc-1-Phosphotransferase führen zu einer schweren Erkrankung, Mukolipidose II, die biochemisch durch Fehlsortierung lysosomaler Enzyme und einer lysosomalen Dysfunktion gekennzeichnet ist. Als Folge kommt es zur zellulären Akkumulation von nicht-abbaubarem Speichermaterial. Skelettale Abnormalitäten, psychomotorische Retardierung und kardiorespiratorische Komplikationen führen zu einem frühzeitigen Tod der Patienten. Die α- und β-Untereinheit werden als gemeinsames Transmembran-Typ III-Vorstufenprotein synthetisiert, das eine modulare Domänenstruktur aufweist. Die Bedeutung der einzelnen Domänen für die GlcNAc-1-Phosphotransferase-Aktivität ist bisher unbekannt. Die α- und β-Untereinheiten enthalten das katalytische Zentrum des Komplexes und weiterhin die Substratbindungsstellen 1. für den Phosphatdonor UDP-GlcNAc, 2. für lysosomale Enzyme und 3. für mannosereiche Oligosaccharide an lysosomalen Enzymen. Die proteolytische Spaltung des α/β Vorstufenproteins in die reifen Untereinheiten ist essentiell für die enzymatische Aktivität der GlcNAc-1-Phophotransferase. Die für die Spaltung verantwortliche Protease ist bisher nicht bekannt.

(A) Aus Sequenzanalysen ist bekannt, dass die Region, die die Aminosäuren 60-430 des α/β Vorstufenproteins umfasst, Ähnlichkeiten zu dem N-terminalen Teil von Kapselbiosyntheseproteinen von Amöben und Bakterien aufweist, die UDP-GlcNAc für den Aufbau der Kapsel nutzen. Daher wird angenommen, dass die N-terminale Region des α/β-Vorstufenproteins für die Bindung des UDP-GlcNAc verantwortlich ist, das das Substrat der enzymatischen Reaktion darstellt. Es konnte gezeigt werden, dass die N-terminale Region des α/β-Vorstufenprotein an einen sogenannten „Reaktiven Farbstoff“ bindet, der dafür bekannt ist, Nukleotid-bindende Proteine zu erkennen. Die strukturelle Untersuchung der N-terminalen Domäne ergab, dass die eingefügte säugerspezifische Domäne (Aminosäuren 102-324), sowie die N Glykane in dieser Region für die Bindung an UDP-GlcNAc nicht von Bedeutung sind. Es konnte außerdem gezeigt werden, dass der Cysteinrest 70 im N-terminalen Teil des α/β-Vorstufenproteins an der Dimerisierung der α-Untereinheit beteiligt ist.

(B) Die Protease, die das α/β-Vorstufenprotein zu den reifen α- und β-Untereinheiten der GlcNAc-1-Phosphotransferase spaltet, konnte in dieser Arbeit identifiziert werden. Mutationsanalysen, subzelluläre Lokalisation und Sequenzvergleiche mit bekannten Spaltsequenzen lieferten die ersten Hinweise, dass das α/β-Vorstufenprotein von der im Golgi-Apparat lokalisierten Site-1-Protease gespalten wird. Die siRNA-vermittelte Downregulation und Untersuchungen in Site-1-Protease-defizienten Zellen bestätigten die Spaltung des α/β-Vorstufenproteins durch die Site-1-Protease. Es wurde gezeigt, dass die N-Glykane der β-Untereinheit wichtig für die Stabilität des α/β-Vorstufenproteins sind, jedoch keine Bedeutung für die Erkennung und die Spaltung durch die Site-1-Protease haben. Die Site-1-Protease ist durch die Spaltung von spezifischen membranintegralen Transkriptionsfaktoren an der Lipid- und Cholesterolhomöostase beteiligt. Site-1-Protease-defiziente Zellen sind nicht in der Lage das α/β-Vorstufenprotein proteolytisch zu aktivieren und zeigen einen Mukolipidose II-ähnlichen Phänotyp. Die Ergebnisse belegen eine neue Funktion der Site-1-Protease in der Biogenese von Lysosomen und weisen darauf hin, dass lipidunabhängige Veränderungen in Site-1-Protease-defizienten Zellen oder Organen zumindest teilweise durch lysosomale Fehlfunktion verursacht werden können.

GlcNAc-1-phosphotransfease is the key enzyme in the generation of the mannose-6 phosphate residue on soluble lysosomal proteins, which functions as a signal for receptor-mediated transport to the lysosome. The hexameric GlcNAc-1-phosphotransfease complex (α2β2γ2) consists of three different subunits that are encoded by two different genes. Defects in the gene encoding the α/β-precursor of the GlcNAc-1-phosphotransfease lead to the severe disorder mucolipidosis II that is biochemically characterized by missorting of lysosomal enzymes and lysosomal dysfunction. The cellular accumulation of non-degradable storage material results in skeletal abnormalities, psychomotor retardation and cardiorespiratory complications accompanied by an early death of the patients.
The α- and β-subunits are synthesized as a single type III-membrane protein showing a complex modular structure. The α- and β-subunits contain the catalytic portion of the complex and substrate binding sites for 1. the phosphate donor UDP-GlcNAc, 2. lysosomal enzymes and 3. high-mannose type oligosaccharides on lysosomal enzymes. The proteolytic cleavage of the α/β-precursor to mature α- and β-subunits is a prerequisite for the enzymatic activity of the GlcNAc-1-phosphotransfease. The protease responsible for the cleavage is not known so far.

(A) Sequence comparison showed that a region (amino acids 60 to 430) of the α/β precursor has similarities to the N-terminal part of capsule biosynthesis proteins of amoeba and bacteria. Therefore the N-terminal region of the α/β-precursor might be responsible for the binding of UDP-GlcNAc, the substrate of the enzymatic reaction. It was shown that the N-terminal part of the α/β-precursor bind to a so-called reactive dye that is known to recognize nucleotide-binding proteins. Structural analysis of the N-terminal domain revealed that the inserted mammalian-specific domain (amino acids 102-324) as well as the N-glycans in this region is not necessary for the binding. Furthermore, cysteine residue 70 seems to be responsible for the dimerization of the α-subunit of the GlcNAc-1-phosphotransferase.

(B) The protease cleaving the α/β-precursor to mature α- and β-subunits of the GlcNAc 1-phosphotransfease was identified. Mutational analysis, subcellular localization and sequence comparison with known cleavage sites gave the first hints, that the α/β-precursor is cleaved by the Golgi-resident site-1 protease. The siRNA mediated knockdown and the analysis of site-1 protease deficient cells confirmed the cleavage of the α/β-precursor by the site-1 protease. It was shown that N-glycans of the β-subunit are required for folding and stabilisation of the α/β-precursor protein but are not necessary for the recognition and cleavage by site-1 protease. Site-1 protease is known to be involved in the lipid- and cholesterol homoeostasis by cleaving specific membrane-bound transcription factors. Site-1 protease-deficient cells are not able to proteolytically activate the α/β precursor and show a mucolipidosis II-like phenotype. These results reveal a new function of site-1 protease in the biogenesis of lysosomes and suggest that lipid-independent phenotypes of site-1 protease-deficient cells and organs are at least partially caused by lysosomal dysfunction.
