Zusammenfassung: 
Die Analoga natürlicher Nucleoside haben breite Anwendung als Wirkstoffe in der antiviralen Chemotherapie sowie in der Krebsbehandlung. Um biologische Aktivität zu zeigen, müssen sie in der Regel intrazellulär in die 5’-Triphosphate überführt werden. Diese Phosphorylierungen erfolgen in drei einzelnen Schritten, die von unterschiedlichen Kinasen katalysiert werden. Aufgrund der hohen Substratspezifität dieser Enzyme finden die Phosphorylierungsreaktionen der Nucleosidanaloga häufig nur ineffizient statt, was eine niedrige intrazelluäre Triphosphatkonzentration und entsprechend niedrige antivirale Aktivität zur Folge hat. Eine Möglichkeit, dieses Problem zu umgehen, ist die Verwendung lipophil maskierter, phosphorylierter Nucleosidanaloga (Pronucleotide), die in der Zelle durch Abspaltung der maskierenden Gruppen das jeweilige Nucleosidphosphat freisetzen können. Ziel dieser Arbeit war es, Methoden zu entwickeln, um die intrazelluläre Freisetzung von Nucleosidmono- und -diphosphaten aus cycloSal- und DiPPro-Nucleotiden untersuchen zu können.

Anhand der antiviralen Aktivität der DiPPro-Nucleotide von 2’,3’-Didesoxyuridin (ddU) und 2’,3’-Didesoxy-2’,3’-didehydrouridin (d4U) sollte die Freisetzung der Nucleosiddiphosphate in der Zelle untersucht werden. Dazu wurden von beiden Nucleosiden jeweils vier DiPPro-Nucleotide mit unterschiedlichen Masken (iPr-, C6-, C9-, C11- substituiert) synthetisiert. Als Vergleichssubstanzen wurden zusätzlich die 3-Me-cycloSal-Triester sowie die Nucleosiddiphosphate beider Nucleoside dargestellt. Die Untersuchung des Hydrolyseverhaltens der DiPPro-Nucleotide in unterschiedlichen Medien zeigte, dass die chemische Hydrolyse in Phosphatpuffer (pH 7.3) nach dem bekannten Mechanismus über das einfach maskierte Intermediat abläuft, aus dem das Nucleosiddiphosphat freigesetzt wird. Durch konkurrierende Spaltung der Anhydridbindung entstehen zusätzlich das Nucleosidmonophosphat sowie der entsprechende Phosphatdiester. Die Untersuchung der antiviralen Aktivität gegen HIV-1 und HIV-2 zeigte erwartungsgemäß keine Aktivität der Nucleoside sowie für die cycloSal-Verbindungen. Doch auch für die DiPPro-Nucleotide wurde keine oder nur moderate Aktivität gefunden (EC50 = 23 µM - 42 µM).

Mit dem Ziel, die Aufnahme der Prodrugs in Zellen mittels Fluoreszenzmikroskopie verfolgen zu können, wurden cycloSal- und DiPPro-Nucleotide bicyclischer Nucleosidanaloga (BCNA) synthetisiert. Neben ihrer antiviralen Aktivität beispielsweise gegen das Varicella-Zoster-Virus zeichnen sich die BCNA durch starke Autofluoreszenz aus. Es wurden 2’-Desoxy- und 2’,3’-Didesoxyanaloga der BCNA verwendet, bei denen sich entweder ein Propylsubstituent oder kein Substituent an der Nucleobase befand. Neben der Untersuchung des Hydrolyseverhaltens wurden drei Verbindungen für Zellaufnahmestudien verwendet. Dazu wurden HeLa-Zellen mit Zellkulturmedium, das die jeweilige Verbindung enthielt, inkubiert und anschließend mittels Fluoreszenzmikroskopie untersucht. Für alle drei Verbindungen konnte gezeigt werden, dass sie in der Zelle in ausreichendem Maße akkumulieren, um unter den Versuchsbedingungen nachgewiesen werden zu können. Allerdings zeigten sie lediglich ein schwaches Fluoreszenzsignal. Neben der nicht optimalen Anregungswellenlänge ist dies im Wesentlichen auf ein schnelles Photobleaching der Fluorophore zurückzuführen, sodass sich keine verlässlichen Rückschlüsse auf die intrazellulären Konzentrationen ziehen lassen.

In einem dritten Ansatz sollte ein Verfahren zur Bestimmung der intrazellulären Konzentration der Hydrolyseprodukte von cycloSal- und DiPPro-Nucleotiden entwickelt werden, die auf der Kopplung von HPLC und Massenspektrometrie (MS) basiert. Es wurden Methoden entwickelt, die die Analyse der lipophilen Prodrugs mittels RP-HPLC und die Untersuchung der Nucleosidmono- und -diphosphate durch HILIC mit anschließender massenspektrometrischer Detektion ermöglichen. Die Anwendung dieser Methoden auf die Verfolgung der Hydrolyse von C6-d4UDP in CEM/0-Zellextrakt zeigte, dass sich alle Hydrolyseprodukte nachweisen lassen. Mit den Prodrugs 3-Me-cycloSal-d4UMP und C6-d4UDP wurden Zellaufnahmestudien unter Verwendung von CEM-Zellen durchgeführt. Die Untersuchung des Zelllysats nach der Inkubation mit der entsprechenden Verbindung zeigte im Fall von 3-Me-cycloSal-d4UMP deutlich das Vorhandensein von d4UMP, während kein Di- und Triphosphat detektiert werden konnte. Bei der Analyse des Zelllysats nach Inkubation mit C6-d4UDP konnte ebenfalls nur das Signal von d4UMP beobachtet werden. Das Di- und Triphosphat konnte nicht detektiert werden.

Analogs of natural nucleosides are widely used in antiviral and anticancer therapy. In order to possess antiviral activity, these compounds need to be phosphorylated to their biologically active triphosphates. These phosphorylations are accomplished in three steps that are catalyzed by different kinases. Due to high substrate specificity of these enzymes the phosphorylation reactions are often inefficient resulting in low intracellular concentration of the triphosphates and low antiviral activity. One option to overcome this obstacle is the use of lipophilically masked phosphorylated nucleoside analogs (pronucleotides) that release the corresponding nucleoside phosphates by cleavage of the masking units inside the cell. The aim of this thesis was to develop new methods to investigate the intracellular release of nucleoside mono- and diphosphates from cycloSal- and DiPPro-nucleotides.

Using DiPPro-nucleotides of 2’,3’-dideoxyuridine (ddU) and 2’,3’-Dideoxy-2’,3’-didehydrouridine (d4U), the intracellular release of the corresponding nucleoside diphosphates should be examined by means of their antiviral activity against HIV-1 and HIV-2. For this purpose, four DiPPro-nucleotides of both nucleosides with different masking units (iPr-, C6-, C9-, C11- substituted) were synthesized. Additionally, the 3-Me-cycloSal triesters and of both nucleosides were prepared. Analysis of the hydrolysis behavior of the DiPPro-nucleotides in different media revealed that hydrolysis in phosphate buffer (pH 7.3) proceeds in accordance with the mechanism described via the mono substituted intermediate that releases the nucleoside diphosphate. By competing cleavage of the anhydride bond the nucleoside monophosphate and respective phosphate diester are formed as well. Examination of the antiviral activity against HIV-1 and HIV-2 showed no activity of the nucleosides and and cycloSal compounds and. For the DiPPro-nucleotides no or only moderate activity (EC50 = 23 µM - 42 µM) was observed as well.

With the goal to study the cellular uptake of prodrugs using fluorescence microscopy, cycloSal- and DiPPro-nucleotides of bicyclic nucleoside analogs were synthesized. Besides their antiviral activity e. g. against varicella-zoster virus, they also exhibit strong autofluorescence. 2’-Deoxy- and 2’,3’-dideoxy analogs of BCNAs either bearing a propyl group or no substituent at the nucleobase were used. Investigation of the hydrolytic behavior in phosphate buffer (pH 7.3) indicated, that DiPPro-nucleotides of BCNAs are also hydrolyzed in compliance with the aforementioned mechanism releasing the nucleoside diphosphates as well as nucleoside monophosphates. In addition to the investigation of their hydrolysis behavior, three compounds were used to investigate their cellular uptake. For this purpose, HeLa cells were incubated with cell culture medium that contained the respective compound and subsequently examined using fluorescence microscopy with an excitation wavelength of 350 nm. All compounds accumulated inside the cell sufficiently to be detected under used conditions. However, they only showed weak fluorescence. The primary reasons for this result are the non-optimal excitation wavelength and rapid photobleaching of the fluorophores.

In a third approach, a procedure to determine intracellular concentrations of hydrolysis products of cycloSal- and DiPPro-nucleotides based on HPLC coupled with mass spectrometry was developed. Initially, methods were developed that allow for the analysis of lipophilic prodrugs using RP-HPLC and polar nucleoside phosphates applying HILIC with mass spectrometric detection. For both methods a mixture of acetonitrile and ammonium formiate buffer (10 mM, pH 8.3) was used as eluent. Utilization of these methods to study the hydrolysis of C6-d4UDP in CEM/0 cell extract revealed that all hydrolysis products could be detected in cellular medium. Prodrugs 3-Me-cycloSal-d4UMP and C6-d4UDP were used to study their cellular uptake in CEM-SS cells. Examination of cell lysate after incubation with the respective compound showed in case of 3-Me-cycloSal-d4UMP the presence of d4UMP but no di- or triphosphate. Analysis of the cell lysate after incubation with C6-d4UDP the presence of d4UMP was observed as well, whereas no di- or -triphosphate was detectable.
