Zusammenfassung: 
Diese Arbeit beschäftigt sich mit der Entwicklung von HPTLC-Methoden zur Analyse von Phenol-Protein-Wechselwirkungen. Die innerhalb dieser Arbeit entwickelten HPTLC-Methoden sollten dazu dienen, die unverdauten Phenol-Protein-Derivate hinsichtlich ihrer Polarität, UV-Aktivität, ihres Molekulargewichtes, ihrer antimikrobiellen und antigenen Eigenschaften zu untersuchen. Nach einem tryptischen Verdau sollten außerdem die Polarität, UV-Aktivität und die Radikalfängerfängereigenschaften der Peptide in einem 1D- und 2D-HPTLC-System untersucht werden. Das Peptidspektrum der Phenol-Protein-Derivate wurde mittels HPTLC-DESI/MS und HPTLC-MALDI/TOF/MS untersucht.
Grundlage zur Analyse der Phenol-Protein-Derivate stellt eine gute Trennung der Peptide und Proteine im chromatographischen System dar. Daher wurde innerhalb dieser Arbeit zunächst eine Methodenentwicklung zur 1D- und 2D-Trennung von Peptiden/Proteinen durchgeführt. Die speziell zur Peptid-/Protein-Analyse angebotenen HPTLC-ProteoChrom®-Kieselgel-Glasplatten zeigten im Vergleich zu den analogen HPTLC Ausführungen eine etwas bessere Trennleistung. Allerdings eignen sich auch die traditionellen HPTLC-Kieselgel-60-Schichten und HPTLC-Cellulose-Schichten sowie RP-Schichten zur Peptid-/Proteintrennung. Als mobile Phase in der 1D-HPTLC erzielte ein Gemisch aus 2-Butanol/Pyridin/Ammoniak (25 %)/Wasser (39/34/10/26, v/v/v/v) gute Ergebnisse. In Kombination mit 2-Butanol/Pyridin/Eisessig/Wasser (10/8/3/5, v/v/v/v) in der zweiten Dimension wurde eine gesteigerte Trennleistung erreicht.
Die postchromatographische Analyse der Radikalfängereigenschaften erfolgt traditionell mit dem HPTLC-DPPH-assay, aber auch der Einsatz eines HPTLC-ABTS-assays ist denkbar. Beide assays wurde innerhalb dieser Arbeit für die Detektion von Radikalfängereigenschaften einfacher phenolischer Verbindungen validiert und miteinander verglichen.
Die postchromatographische Analyse der antimikrobiellen Eigenschaften wird durch die Kombination von der HPTLC und der Bioautographie realisiert. Diese Technik sollte zur Analyse der antimikrobiellen Eigenschaften der Phenol-Protein-Derivate genutzt werden. Dazu wurden zunächst die in der HPTLC traditionell verwendeten stationären und mobilen Phasen auf eine Beeinflussung des Mikroorganismenwachtsums untersucht. Eine negative Beeinflussung des Wachstums wurde für Ameisensäure, Eisessig, Ammoniak (25 %) und n-Hexan sowie für HPTLC-KG-60-NH2-F254S-Glasplatten und TLC-KG-60-RP-8-F254S-Glasplatten festgestellt. Außerdem wurde der Einfluss der optischen Dichte (d.h. Bakterienkonzentration) und der Inkubationsdauer auf das Analysenergebnis der HPTLC-Direkte Bioautographie untersucht.
Die postchromatographische Analyse der antigenen Eigenschaften wird mit Hilfe der HPTLC-Immunfärbung (IF) erreicht. Die Untersuchung der Eignung von stationären Phasen ergab, dass Kieselgel-60-Schichten für die HPTLC-IF von Myoglobin gut geeignet sind. Bei der Verwendung von Kieselgel-RP-18-Schichten wurde Myoglobin nicht mittels Immunfärbung detektiert.
Die Kopplungen von der HPTLC zur ESI/MS, DESI/MS und MALDI/TOF/MS wurden im Rahmen dieser Arbeit erfolgreich für die Analyse von unmodifizierten sowie modifizierten Peptiden eingesetzt. Mit Hilfe der HPTLC-DESI/MS und HPTLC-MALDI/TOF/MS wurden modifizierte Peptide auf der HPTLC-Platte lokalisiert und identifiziert, wobei die Stellung der Modifikation im Peptidmolekül durch eine HPTLC-DESI/MS/MS Analyse bis auf zwei Aminosäureseitenketten eingegrenzt werden konnte.
Die Reaktion von Modellproteinen und Chinonen sowie Semichinon-Intermediaten phenolischer/verwandter Verbindungen, die in der Literatur beschrieben werden, wurde innerhalb dieser Arbeit bestätigt. Zusätzlich wurden mit Hilfe der in dieser Arbeit entwickelten HPTLC-Methoden Ergebnisse, z.B. in Bezug auf die UV-Aktivität und auf die antigenen Eigenschaften, erhalten, die mit den bisherigen analytischen Methoden nicht erzielt werden konnten. Die Reaktivität der in dieser Arbeit untersuchten phenolischen/verwandten Verbindungen gegenüber den Modellproteinen Myoglobin, Lysozym, Casein und Ovalbumin nimmt in der Reihenfolge Chinasäure, Ferulasäure, p-Cumarsäure, Syringasäure, Sinapinsäure, Kaffeesäure, Gallussäure und Tanninsäure zu. Es kann festgestellt werden, dass die kovalente Bindung von Chinonen und Semichinon-Intermediaten phenolischer/verwandter Verbindungen an Proteine entscheidend durch Parameter, wie die Stellung der Hydroxyl- und Methoxygruppen sowie der Aminosäurekomposition der Proteine beeinflusst wird. Diese Beeinflussung wurde vielfach in Studien erwähnt und kann durch die Ergebnisse dieser Arbeit bestätigt werden. Diese Reaktion führt zu unterschiedlichen Phenol-Protein-Derivaten, die sich in den hier untersuchten Eigenschaften signifikant voneinander unterscheiden (Polarität, UV-Aktivität, Färbung mit Derivatisierungsreagenzien, Radikalfängereigenschaften, Antigenität, Molekulargewicht).

This work deals with the development of HPTLC methods for the analysis of phenol-protein interactions. HPTLC methods developed in this work should serve to investigate the undigested phenol-protein derivatives with respect to their polarity, UV activity, their molecular weight, and their antimicrobial and antigenic properties. After tryptic digestion, the peptides are analyzed in a 1D- and 2D-HPTLC system regarding their polarity, their UV-activity and radical scavenging properties. Furthermore, the peptide profile of phenol-protein derivatives was examined by HPTLC-DESI/MS and HPTLC-MALDI/TOF/MS.
A good separation in the chromatographic system represents the basis for the analysis of phenol-protein derivatives. Therefore, a method development for the 1D- and 2D-separation of peptides and proteins was initially performed within this work. ProteoChrom® HPTLC silica gel glass plates showed a better separation efficiency compared to the analog HPTLC plates. However, the traditional HPTLC silica gel 60, HPTLC cellulose layers, and reversed phase layers are also valid for peptide and protein separation. The mobile phase used in the 1D-HPTLC, a mixture of 2-butanol/pyridine/ammonia (25 %)/water (39/34/10/26, v/v/v/v), gave good results. In combination with 2-butanol/pyridine/glacial acetic acid/water (10/8/3/5, v/v/v/v) in the second dimension, an increased separation efficiency was achieved. The evaluation of 1D-chromatograms may be performed before or after postchromatographic derivatization using a wavelength scan. 2D-chromatograms can be evaluated using the software ImageMaster 2D Platinum.
The postchromatographic analysis of radical scavenging properties is traditionally done using the HPTLC-DPPH-assay, but also the use of HPTLC-ABTS-assays is possible. In this work, both assays were validated regarding the detection of simple phenolic compounds’ free radical scavenging properties and results were compared.
The postchromatographic analysis of antimicrobial properties is provided by the combination of HPTLC and bioautography. This technique was used for analysis of phenol-protein derivatives’ antimicrobial properties. For this purpose, the influence of traditionally used stationary and mobile phases was evaluated. A negative effect on the growth of microorganisms was observed for formic acid, glacial acetic acid, ammonia (25 %) and n-hexane as well as HPTLC silica gel NH2 F254S glass plates and TLC silica gel RP-8 F254S glass plates. Furthermore, the influence of the optical density and incubation period on the result of the HPTLC-direct bioautography analysis was examined. An optical density of 0.1 to 1 and an incubation period of one to four hours led to a good result.
The postchromatographic analysis of the antigenic properties is achieved by means of HPTLC-immunostaining (IS). The study of the stationary phase suitability showed that silica gel 60 layers are well suited for the HPTLC-IS analysis of myoglobin. When using silica gel RP-18 layers, myoglobin was not detected by immunostaining.
The coupling of HPTLC and ESI/MS, DESI/MS and MALDI/TOF/MS have been successfully used in this work for the analysis of unmodified and modified peptides. Using HPTLC-MALDI/TOF/MS and HPTLC-DESI/MS modified peptides were located and identified on the HPTLC plate. The modifications’ position within the peptide molecule could be localized to two amino acid side chains by a HPTLC-DESI/MS/MS analysis.
The reaction of quinones and semiquinone intermediates of phenolic/related compounds with model proteins, which is already described in the literature, was confirmed in this study. In addition, HPTLC methods developed in this work gave additional results, e.g. in terms of UV-activity and antigenic properties. These observations could barely be achieved with the traditional analytical methods. The reactivity of the examined phenolic/related compounds against the model protein myoglobin, lysozyme, casein and ovalbumin increases in the order of quinic acid, ferulic acid, p-coumaric, syringic, sinapic acid, caffeic acid, gallic acid and tannic acid. It can be noted, that the covalent attachment of quinones and semiquinone intermediates of phenolic/related compounds to proteins is influenced by crucial parameters, such as the position of the hydroxyl and methoxy groups, and proteins’ amino acid composition. This influence was already mentioned in literature and can be confirmed by the results of this work. This reaction leads to different phenol-protein derivatives, which significantly differ in investigated properties (polarity, UV activity, free radical scavenging properties, antigenicity, molecular weight).
