Der Transkriptionsfaktor NF-κB reguliert eine Vielzahl biologischer Prozesse und spielt vor allem in der angeborenen Immunantwort, dem Zelltod und der Zell¬proli¬feration eine wichtige Rolle. Die Aktivierung von NF-κB kann sowohl positive als auch negative Auswirkungen auf die virale Replikation haben. Im Zuge der angeborenen Immunantwort induziert NF-κB die Produktion pro-inflammatorischer Zytokine, Chemokine und Zell¬adhäsionsmoleküle, was der Bekämpfung der Virusinfektion dient. NF-κB induziert jedoch auch die Expression anti-apoptotischer und proliferationsfördernder Proteine, was das Überleben infizierter Zellen begünstigt. Aufgrund dieser ambivalenten Funktion von NF-κB regulieren viele Viren die NF κB-Aktivität auf komplexe Weise und exprimieren häufig sowohl NF-κB-aktivierende als auch NF-κB-inhibierende Proteine.
Das murine Cytomegalovirus (MCMV) induziert zu Beginn der Infektion eine kurzzeitige Aktivierung von NF-κB. Zu späteren Zeitpunkten wird die NF-κB-Aktivierung jedoch blockiert. In vorangegangenen Studien wurde gezeigt, dass das virale Protein M45 für die NF-κB-Inhibition verantwortlich ist. M45 interagiert mit der IκB-Kinase (IKK)-Untereinheit NEMO (NF κB essential modulator) und induziert deren Degradation, was zur Blockierung aller kanonischen NF-κB-Signalwege führt.
Die Ergebnisse der vorliegenden Arbeit zeigen erstmals, dass M45 auch die initiale NF-κB-Aktivierung nach Infektion induziert. In murinen Fibroblasten, die mit einem M45-defizienten Virus infiziert werden, findet keine sofortige NF-κB-Aktivierung statt. Die initiale NF-κB-Aktivierung ist blockiert in Zellen, die kein NEMO besitzen und stark reduziert in Zellen, denen RIP1 (receptor-interacting protein 1) fehlt, ein bekannter Interaktionspartner von M45. Bei Plasmid-vermittelter Expression induzieren alle M45-Verkürzungsmutanten, die mit RIP1 und NEMO interagieren können, die Aktivierung eines NF-κB-abhängigen Reportergens. Im Kontext der Virusinfektion ist die Interaktion mit RIP1 und NEMO jedoch nicht ausreichend für die NF-κB-Aktivierung. Zusätzlich ist die Inkorporation von M45 in Viruspartikel nötig. Dies ist abhängig von einem Bereich im C-Terminus des M45-Proteins, der nicht für die Interaktion mit RIP1 und NEMO benötigt wird. Nur das virion-assoziierte M45, nicht jedoch das neusynthetisierte M45, ist in der Lage, NF-κB in infizierten Zellen zu aktivieren.
Die Resultate dieser Arbeit erweitern das Verständnis der komplexen und dynamischen Immunmodulation durch MCMV. Sie zeigen, dass M45 eine duale Rolle bei der NF κB-Regulation in infizierten Zellen spielt und ermöglichen die Aufstellung folgenden Modells: Das mit den Viruspartikeln in die Zelle gelangende M45 interagiert mit RIP1 und NEMO, was initial die Aktivierung von NF-κB induziert. Dem folgt die M45-vermittelte Degradation von NEMO, die zu einer dauerhaften NF-κB-Inhibition führt.

Transcription factor NF-κB regulates numerous biological processes and plays an especially important role in the innate immune response, cell death and cell proliferation. The activation of NF-κB can have both beneficial and detrimental effects on viral replication. NF-κB induces the production of proinflammatory cytokines, chemokines and cell adhesion molecules, which are essential components of the innate immune response serving to counteract viral infection. However, NF-κB also induces the expression of anti-apoptotic and proproliferative proteins, which promotes the survival of infected cells. Due to this ambivalent function of NF κB many viruses regulate the NF-κB activity in a complex manner and often express both NF-κB-activating and NF-κB-inhibiting proteins.
The murine cytomegalovirus (MCMV) induces a brief and transient activation of NF-κB immediately after infection, but blocks NF-κB activation at later times. Previous studies have shown that the viral protein M45 is responsible for the NF-κB inhibition. M45 interacts with the IκB kinase (IKK) subunit NEMO (NF κB essential modulator) and induces its degradation, which leads to the inhibition of all canonical NF-κB pathways.
The results presented here show for the first time that M45 also induces the activation of NF κB immediately after infection. In murine fibroblasts infected with an M45-deficient virus, no rapid NF-κB activation can be observed. The initial NF-κB activation is abolished in cells that do not express NEMO and strongly reduced in cells lacking RIP1 (receptor-interacting protein 1), a known interaction partner of M45. When M45 is expressed from a plasmid, all M45 mutants that are capable of interacting with RIP1 and NEMO can also induce the expression of an NF-κB-dependent reporter gene. However, in the context of a viral infection, the interaction with RIP1 and NEMO is not sufficient to induce NF-κB activation. In addition, the incorporation of M45 into viral particles is required. This depends on a C-terminal region of the M45 protein that is not required for the interaction with RIP1 and NEMO. Only the virion-associated M45, but not the newly synthesized M45, is capable of activating NF-κB upon viral infection.
The data of this study expand the knowledge and understanding of the complex and dynamic immune modulation of MCMV. They demonstrate a dual role of M45 in the regulation of NF κB in infected cells and allow the proposition of the following model: M45 that is delivered to the cell by viral particles interacts with RIP1 and NEMO, which initially activates NF-κB. Subsequently, M45 induces the degradation of NEMO leading to a permanent inhibition of NF-κB.
