Titel: Phenolische Inhaltsstoffe in Cistus incanus Tee – Charakterisierung und Stabilität innerhalb der Teezubereitung

Zusammenfassung: 
Im Rahmen der vorliegenden Arbeit wurden 32 phenolische Substanzen in C. incanus Tee mittels LC-DAD-ESI-MS/MS identifiziert. Bei diesen Inhaltsstoffen handelt es sich um ein breites Spektrum an Phenolsäuren, monomeren und dimeren Flavanolen, Flavonolglycosiden und Ellagitanninen. Weiterhin wurden mittels LC-DAD 21 phenolische Inhaltsstoffe in sieben C. incanus Handelsproben quantifiziert. Hierbei wurden signifikante Konzentrationsunterschiede beobachtet. Diese wurden größtenteils auf die Partikelgröße des Teekrautes sowie die darin enthaltenen C. incanus Blatt- und Stängelanteile zurückgeführt. Eine Quantifizierung der phenolischen Substanzen in gesiebten (Partikelgrößen: 0,3 mm-2,8 mm) sowie in handsortierten Proben mit definierten Blatt- und Stängelanteilen (0 % w/w-100 % w/w Blattanteil) bestätigte dies. Weiterhin wurde eine Methode zur Bestimmung des Holzanteiles im Teekraut anhand des Cellulosegehaltes für C. incanus Tee etabliert. Hierbei korrelierte ein hoher Cellulosegehalt stark mit dem Holzanteil der Probe. Insgesamt resultierten aus Teekraut mit einer feinen Partikelgröße (0,3 mm) höhere Konzentrationen phenolischer Inhaltsstoffe, als aus Proben mit einer groben Partikelgröße (2,8 mm). Zusätzlich waren bei hohen Blattanteilen, bzw. niedrigen Stängelanteilen des Teekrautes, signifikant höhere Konzentrationen der phenolischen Substanzen im trinkfertigen C. incanus Tee enthalten.

Weiterhin erfolgte eine Charakterisierung der C. incanus Proben anhand der Gesamtphenolgehalte (Folin-Ciocalteu-Assay), Tanningehalte (Methylcellulose-Präzipitations-Assay) und gesamten antioxidativen Kapazitäten (Elektronenspinresonanz-Spektroskopie). Die Ergebnisse dieser Summenparameter korrelierten ebenfalls mit der Beschaffenheit des Teekrautes (Partikelgröße und Blatt-/ Stängelanteil des Teekrautes). In Handelsproben mit einer feinen Partikelgröße (0,3 mm) wurden höhere Werte für diese Parameter gemessen. Ein ähnliches Ergebnis resultierte aus hohen Blatt- bzw. niedrigen Stängelanteilen des Teekrautes. Insgesamt enthielten die C. incanus Handelsproben hohe Gesamtphenol- und Ellagitanningehalte. Diese phenolischen Verbindungen waren in C. incanus Tee für die gesamte antioxidative Kapazität und damit für einen Großteil der Bioaktivität verantwortlich. Die absoluten antioxidativen Kapazitäten von insgesamt 22 phenolischen Inhaltsstoffen in C. incanus Tee wurden mittels LC-onlineTEAC (Trolox Equivalent Antioxidant Capacity) bestimmt. Die Höhe der antioxidativen Kapazitäten dieser Substanzen war hierbei stark von deren Molekülstruktur abhängig. Darüber hinaus wurden 14 identifizierte Substanzen mittels präparativer LC-DAD isoliert und für weitere Bioaktivitätstests zur Verfügung gestellt. Unter diesen Substanzen waren Phenolsäuren, monomere und dimere Flavanole sowie Flavonolglycoside. Die Ergebnisse zu den Wirkungen dieser Substanzen dürfen aus Gründen der Geheimhaltung nicht in der vorliegenden Arbeit präsentiert werden.

Weiterhin wurden Effekte verschiedener Faktoren aus dem Prozess der Teezubereitung auf die Konzentrationen bioaktiver Substanzen in C. incanus Tee untersucht. Hierbei wurden durch hohe Kochtemperaturen (95 °C) die höchsten Konzentrationen erzielt. Zudem waren die quantifizierten phenolischen Substanzen bei diesen Temperaturen auch über längere Kochzeiten stabil. Ein Abbau wurde erst ab einer Kochzeit über 20 min beobachtet. Einen bedeutenden Faktor bezüglich hoher Konzentrationen bioaktiver Inhaltsstoffe in C. incanus Tee stellte der Einsatz von mineralstoffarmen Kochwasser innerhalb der Teezubereitung dar. Eine Verwendung von Mineralwasser führte hingegen beim Abkühlen des Tees zu einer Komplexierung phenolischer Inhaltsstoffe. Dies zeigte sich optisch in der Bildung eines Präzipitates sowie eines dünnen Oberflächenfilmes auf dem Tee (Tea Cream). Für beide Ausprägungen der Tea Cream wurden in C. incanus Tee hohe gesamte antioxidative Kapazitäten mittels Elektronenspinresonanz-Spektroskopie nachgewiesen. Für den wässrigen Überstand (Tee) resultierte durch Bildung der Tea Cream eine geringere gesamte antioxidative Kapazität im Vergleich zu einem Tee ohne eine solche Komplexierung bioaktiver Substanzen. Weiterhin konnte in einem Modellversuch zur Stabilität von Catechin in Abhängigkeit vom pH-Wert des Kochwassers sowie der Substanzkonzentration Epimerisierungs- sowie Autoxidations-reaktionen nachgewiesen werden. Durch die Reaktion von Catechin, insbesondere bei basischem pH-Wert (pH 11) des Kochwassers und bei vergleichbaren Catechinkonzentrationen (0,01 mg/mL) wie in C. incanus Tee, wurde eine Epimerisierung zu Epicatechin sowie eine Autoxidation zu Dehydrodicatechin A beobachtet. Beide Substanzen wurden auch in C. incanus Tee nach längerer thermischer Behandlung (60 min bei 95 °C) mit Mineralwasser (pH 7,6) detektiert.

Within the present work 32 phenolic substances were identified in C. incanus infusions by the use of LC-DAD-ESI-MS/MS. These compounds included a wide range of phenolic acids, monomeric and dimeric flavanols, flavonol glycosides as well as ellagitannins. Further on, 21 of these phenolic compounds were quantified in infusions brewed from seven commercially available C. incanus herbal teas by LC-DAD. Significant differences in the concentrations of these substances were the result. These varying contents were ascribed mainly to the particle size of the herb material and to the processed contents of leaves and stems therein. A quantification of phenolic substances in sieved (particle sizes: 0.3 mm-2.8 mm) and manually sorted (0 % w/w-100 % w/w leaf content) C. incanus herbal tea samples confirmed this assumption. Furthermore, a method for determining the real wood content in herbal material for C. incanus infusions was established in this work. Within this method the cellulose content correlated strongly with the wood content of the herbal tea samples. Altogether brewing of C. incanus herb with a small particle size (0.3 mm) resulted in higher concentrations of phenolic substances in the infusion, than brewing herb with a larger particle size (2.8 mm). Additionally, high leaf or low stem contents of herbal tea achieved higher concentrations of these compounds in C. incanus infusions.

In the second part of this work characterizations of the C. incanus infusions, based on determinations of total phenol contents (Folin-Ciocalteu assay), tannin contents (methylcellulose precipitation assay) and total antioxidant capacities (electron spin resonance spectroscopy), were performed. The results for the aforementioned analytical parameters correlated with the constitution of herb material (particle size and leaf or stem content) too. In the commercially available C. incanus herbal tea samples with a small herbal particle size (0.3 mm), higher values were measured for these parameters. Similar results were observed for high leaf or low stem contents of the herbal material. Altogether the seven analyzed C. incanus infusions contained high total phenol and high ellagitannin contents. These phenolic substances were responsible for the total antioxidant capacity and therewith for most of the bioactivity of C. incanus herbal infusions. By use of LC-onlineTEAC (Trolox Equivalent Antioxidant Capacity) the absolute antioxidant capacity of 22 phenolic ingredients of the herbal infusions were determined. The TEAC depended on the molecular structure of these single compounds. For a further investigation of bioactivity and potential health benefiting effects, 14 of the identified substances were isolated by preparative LC-DAD and provided for external analyses. These isolated compounds included phenolic acids, monomeric and dimeric flavanols as well as flavonol glycosides of C. incanus herbal infusions. The results of the external analyses were not allowed to be shown in the present work due to secrecy reasons.

In the last part of this work the effects of chosen factors of the brewing process on the concentrations of bioactive substances in C. incanus infusions were investigated. The highest contents of these compounds were achieved by high temperatures (95 °C) during the brewing process. In addition these substances were heat-resistant at the described temperature even for long brewing times. A thermal degradation was not observed before a brewing time of 20 min. The use of brewing water with low mineral contents was identified as an important factor in achieving high concentrations of bioactive substances in C. incanus infusions. The application of mineral water led to a complex formation of phenolic substances during cooling of the infusions. The result was a tea cream formation in terms of a precipitate on the base of the cup and a thin film on the surface of the infusion. For both, precipitate and surface film, high total antioxidant capacities were proven by electron spin resonance spectroscopy. For the aqueous supernatant (tea) a much lower antioxidant capacity was measured due to complex formation of bioactive substances. In infusions without such a tea cream formation higher total antioxidant capacities were observed. Additionally, in a model test the resistance of catechin against epimerization and autoxidation was dependent of the pH-value of the brewing water as well as the substance concentration in the test solution. Through reaction of catechin, especially at an alkaline pH-value (pH 11) of brewing water and a catechin concentration of the test solution comparable to C. incanus herbal infusions (0.01 mg/mL), an epimerization to epicatechin as well as an autoxidation to dehydrodicatechin A was observed. Both reaction products of catechin were detected in C. incanus herbal infusions after a prolonged heat treatment (60 min at 95 °C) with mineral water (pH 7.6).
