Im Jahr 2015 starben noch immer mehr als eine Million Menschen durch das HI-Virus und über zwei Millionen haben sich neu mit HIV infiziert. Die Entwicklung neuer Wirkstoffe und die Suche nach Impfstoffen sind daher fortwährend Gegenstand der Forschung. Einen möglichen Ansatzpunkt stellen therapeutisch einsetzbare Antikörper gegen das virale Oberflächenprotein gp120 dar, die das Virus langfristig zu neutralisieren vermögen. Die häufig in Infizierten gefundenen Antikörper binden meist das 307IHIGPGRAF315-Motiv in dem V3-Loop des gp120, welches auch prinzipiell neutralisierende Domäne (PND) genannt wird. Diese Antikörper zeigten jedoch keine langfristige und breite Neutralisierung des Virus. Im Rahmen dieser Arbeit sollten daher V3-Peptide und V3-Glycopeptide ohne PND synthetisiert und charakterisiert werden, die zukünftig zur Selektion von Antikörpern gegen die V3-Region dienen können. Jene Antikörper sollen den loop an anderen Bereichen als der PND binden und somit möglicherweise zu einer längerfristigen Neutralisation führen.
Dafür wurde zusätzlich zum linearen, unglycosylierten V3-Peptid P1, bei dem die PND durch das Aminosäure-Motiv PS ersetzt wurde, das analoge, lineare V3 Glycopeptid P2 mit einer GlcNAc(β1,4)-GlcNAc-Glycosylierung an N301 synthetisiert. Die beiden linearen Peptide konnten über eine Disulfidbrücke der terminalen Cysteine zyklisiert werden, wodurch die Peptide P3 und P4 erhalte wurden. Die Glycopeptidsynthese wurde dabei in insgesamt fünf verschiedenen Varianten mit einem acetylierten und einem ungeschützten Chitobiosylasparagin-Baustein durchgeführt, um die optimalen Synthesebedingungen zu finden. Es zeigte sich, dass die vollständig automatisierte Synthese nach Variante 1 die höchsten Ausbeuten lieferte. Um auch in der Lage sein zu können Glycopeptide mit größeren Komplextyp-Kohlenhydrat-Strukturen herzustellen, wurde das Nonasaccharid 19 aus bovinem Fibrinogen isoliert. Neben dem Nonasaccharid wurde jedoch ebenfalls das fucosylierte Decasaccharid erhalten, welches sich aber nicht vollständig abtrennen ließ.
Anschließend wurden alle vier Peptide und Glycopeptide mit Hilfe von SPR-Experimenten auf ihre Bindung zum humanen CCR5-Rezeptor untersucht. Der Rezeptor wurde in Form von CCR5-überexprimierenden GHOST(3)-Hi5-Zellen eingesetzt, die 4 · 104 Rezeptoren auf ihrer Oberfläche präsentieren. Es zeigte sich, dass die Peptide trotz Trunkierung den CCR5-Rezeptor spezifisch binden. Bis auf P1 entsprachen die Affinitäten der untersuchten V3-Peptide der Annahme, dass V3-Glycopeptide stärker mit CCR5 interagieren als unglycosylierte. Ebenso führte die Zyklisierung über eine Disulfidbrücke der flankierenden Cysteine zu einer höheren Affinität als bei vergleichbaren linearen V3-Peptiden.
Der Einfluss der Trunkierung auf die Konformation wurde zunächst durch eine Strukturvorhersage mittels molecular modeling für die zyklischen V3-Peptide P3 und P4 sowie eines zu P4 analogen Glycopeptids mit einem Nonasaccharid statt Chitobiose untersucht. Die erhaltenen Strukturen zeigten ebenso wie die Röntgenstruktur des nativen V3-Loops eine langgestreckte, aber stärker verdrillte Konformation. Die Glycosylierung hatte dabei einen Einfluss auf die Anzahl intramolekularer Wasserstoffbrückenbindungen. Die reinen In-Silico-Strukturen von P3 und P4 wurden mit experimentell ermittelten Strukturen einer NMR-Konformationsanalyse verglichen. Aus NOE-Aufbaukurven ließen sich Abstände und aus 3J Kopplungskonstanten φ-Winkel berechnen, mit denen über einen Distance-geometry-Algorithmus Konformationen erhalten wurden. Diese experimentell erhaltenen Konformationen waren ebenfalls langgestreckt, im Fall von P3 insgesamt aber etwas weiter und offener. P4 zeigte wie die In-Silico-Struktur eine stärkere Verdrillung als P3. Somit konnte gezeigt werden, dass Strukturvorhersagen von V3-Peptiden mit molecular modeling Tendenzen aufzeigen können.
Zuletzt wurden der trunkierte und der native V3-Loop, jeweils mit Nonasaccharid am N301, in die mit Maraviroc cokristallisierte Röntgenstruktur des CCR5-Rezeptors modelliert. Dabei sollte herausgefunden werden, ob das sterisch anspruchsvolle Glycan, das im Modell von Tan et al. fehlt, bei gleicher Orientierung des loops passt. Das Tan-Modell konnte weder für den trunkierten noch für den nativen loop reproduziert werden, jedoch störte das Glycan in keiner getesteten Orientierung. Der native V3-Loop wechselwirkte allerdings weniger mit CCR5 als der trunkierte.

In 2015 still more than a million people died of HIV and over two million new infections were registered. Therefore, the development of new active compounds and the quest for a vaccine are a crucial part of research. A possible approach to long-term neutralization of the virus could be therapeutic antibodies against the viral envelope protein gp120. Antibodies, frequently found in patients, mostly bind the 307IHIGPGRAF315 motif in the V3 region of gp120, also called principal neutralizing domain (PND). However, those antibodies show no broad and long-term neutralization of the virus.
Thus, in this thesis V3 peptides and glycopeptides without PND were to be synthesized and characterized, that can be used to select V3-specific antibodies. Those antibodies should bind the V3 loop at a different site than the PND and so might ensure a longer neutralization.
To achieve this goal, additional to the linear unglycosylated V3 peptide P1, in which the PND is replaced by the amino acid motif PS, the analogous linear V3 glycopeptide P2 with a GlcNAc(β1,4)-GlcNAc glycosylation at N301 was synthesized. Both linear peptides were also cyclized by a disulfide bond between the flanking cysteines, which gave the peptides P3 and P4. The glycopeptide synthesis was performed in five variants with an acetylated and an unprotected chitobiose-asparagine building block to find the optimal conditions. The best yields were provided by the fully automated synthesis of variant 1.
To be able to synthesize glycopeptides with bigger complex type carbohydrate structure the nonasaccharide 19 was isolated from bovine fibrinogen. As a byproduct of the nonasaccharide the fucosylated decasaccharide was found, which could not be separated completely from the nonasaccharide.
All four peptides and glycopeptides were analyzed by SPR considering their binding affinity to the human CCR5 receptor. The receptor was available as CCR5 over-expressing GHOST(3) Hi5 cells, which present 4 · 104 receptors at their surface. The peptides specifically bound the CCR5 receptor despite their truncation. Except for P1, the affinities of the analyzed V3 peptides correlated to the assumption that V3 glycopeptides interact stronger with CCR5 than unglycosylated peptides. Also the cyclization via the terminal cysteines led to higher affinities than comparable linear V3 peptides.
The effect of the truncation on the peptide conformation was first examined by an in silico prediction of the structures of the cyclic peptides P3 and P4 and a glycopeptide analogous to P4 with chitobiose replaced by a nonasaccharide. Like the crystal structure, the peptide structures showed an oblong and but more twisted conformation. The glycosylation had an impact on the amount of intramolecular hydrogen bonds. The in Silico structures of P3 and P4 were also compared to experimental structures from a NMR conformation analysis. Distances were calculated from NOE build-up curves and φ angles from 3J coupling constants, which were both used to calculate conformations by a distance geometry algorithm. Those experimental conformations were also long-stretched. In the case of P3 the conformation was a bit wider and more open than the in silico conformation. P4 was as well as the in silico structure more twisted than P3. Therefore, it was demonstrated that structure prediction of V3 peptides by molecular modeling can provide tendencies.
Finally, the truncated and the native V3 loop, each carrying a nonasaccharide at N301, were modeled into the with Maraviroc co-crystallized x-ray structure of CCR5. The model of Tan et al. does not include the N301 glycosylation which was to be examined in this study. The orientation of the Tan model could not be reproduced for the truncated or the native V3 loop but the glycan did not disturb or clash into the receptor in any tested orientation. However, the native V3 loop interacted less with CCR5 than the truncated.
