Das Pankreaskarzinom ist eine der aggressivsten Tumorerkrankungen und zeichnet sich durch ein schnelles Wachstum mit frühzeitiger Metastasierung, eine schlechte Prognose und eine hohe Resistenz gegenüber Chemo- und Strahlentherapie aus. Um alternative Therapiemöglichkeiten zu finden, werden zunehmend molekulare Vorgänge in der Zelle untersucht und das komplexe Netzwerk der Signalwege analysiert. Diese Arbeit möchte dazu einen Beitrag leisten indem es die Inhibierbarkeit von MALT1 und dessen Auswirkung auf die c-Rel Kerntranslokation untersucht.
Die Transkriptionsfaktoren der NF-κB-Familie sind von zentraler Bedeutung bei der Regulation der Immunantwort, bei Entzündungsvorgängen und bei der Entwicklung des Immunsystems aber auch für die Entstehung von Tumoren. Unter dem Namen NF-κB werden die Proteine RelA, RelB und c-Rel, p105 (NF-κB1), p100 (NF-κB2), welche Vorstufen von p50 und p52 sind, zusammengefasst. Diese NF-κB-Untereinheiten bilden Dimere und befinden sich im Zytoplasma, durch Bindung an IκB-Proteine, in einem inaktiven Zustand. Durch die Parakaspase MALT1 zusammen mit Bcl10 und CARD11 wird der IKK-Komplex, bestehend aus den Untereinheiten IKKα, IKKβ und IKKγ, aktiviert. Dieser Komplex phosphoryliert die IκB-Proteine, die dann durch Ubiquitinylierung zum Abbau durch das Proteasom markiert werden. So werden NF-κB-Dimere freigesetzt und translozieren in den Zellkern, wo sie durch Bindung an spezifische responsive Elemente der DNA die Genexpression beeinflussen können.
Ein Ziel dieser Arbeit war es die Wirksamkeit der Medikamente Mepazin, Biperiden und der Kombination aus beiden Wirkstoffen auf die Parakaspase MALT1 in den Zellen des Pankreaskarzinoms nachzuweisen. Hierfür wurde ein MALT1-Aktivitätsassay durchgeführt, der die proteolytische Aktivität von MALT1 misst. Es wurde erwartet, dass die proteolytische Aktivität in den Zellen nach Mepazin- und Biperidenbehandlung geringer ausfällt als in den Zellen der DMSO-Kontrollgruppe.
Zur Untersuchung der Kerntranslokation wurden die Zellen unterschiedlich lange mit Mepazin, Biperiden und der Kombination beider Wirkstoffe behandelt. Anhand von Kernfraktionierung und anschließendem Western Blot wurde die Quantität von c-Rel im Zellkern bestimmt. Als Resultat zeigte sich, dass nach fünfzehnminütiger Behandlung mit Biperiden und auch mit Mepazin signifikant weniger c-Rel im Zellkern zu finden war. Hier bestätigte sich die Arbeitshypothese und legt die Vermutung nahe, dass durch Biperiden und Mepazin MALT1 gehemmt wird und dadurch weniger c-Rel in den Zellkern translozieren kann, wodurch die antiapoptotische Wirkung von c-Rel ausbleibt und die Pankreaskarzinomzellen vermehrt in Apoptose gehen.
Auch für die weitere von uns untersuchte Zelllinie L3.6pl Res zeigte sich nach der Behandlung mit Biperiden und Mepazin eine inhibierende Tendenz auf die c-Rel-Kerntranslokation. Wie sich schon bei den Panc-1 Zellen zeigt, erfolgt auch bei den L3.6pl Res Zellen bei der Behandlung mit der Kombination aus beiden Medikamenten eher eine Zunahme der Quantität von c-Rel im Zellkern.
Diese Ergebnisse könnten richtungsweisend sein und Ansatzpunkte für die Entwicklung therapeutischer Konzepte für das Pankreaskarzinom und andere maligne Erkrankungen liefern.

Pancreatic cancer is one of the most aggressive types of cancer and is particularly dangerous because of its quick growth and early metastases. Patients affected by pancreatic cancer have a poor prognosis due to the high resistance to chemo- and radiotherapy. Alternative treatment options are urgently needed and many intracellular molecular pathways are being studied to understand the complex cellular signaling pathways. This thesis tries to contribute to the understanding and treatment of this particular kind of cancer by investigating the susceptibility to inhibition of MALT1 and the impact of this inhibition on the nuclear translocation of the antiapoptotic transcription factor c-Rel.
The transcription factors of the NF-κB family are of central importance to the regulation of immune responses, to inflammatory processes, and to the development of the immune system. However, they also play a vital role in the development of tumors.
NF-κB consists of many subunits such as RelA, RelB, c-Rel and p105 (NF-κB1) and p100 (NF-κB2) which are precursors of p50 and p52. These subunits form dimeric structures which are, bound to IκB proteins, located inactively in the cytoplasm. The activation of these NF-κB subunits occurs when paracaspase MALT1 forms the so called IKK-complex by uniting with Bcl10 and CARD11. This IKK-complex consists of three subunits IKKα, IKKβ und IKKγ. It phosphorylates the inactivating IκB proteins which in turn results in their ubiquitination to indicate the degradation of these IκB proteins by the proteasome. Now, the NF-κB subunits are no longer inactivated and are free to translocate to the nucleus regulating the gene expression by binding to specific responsive elements.
One aspiration of this thesis was to proof the inhibitory effects of the drugs Mepazine and Biperiden on paracaspase MALT1 in pancreatic cancer cell lines. In order to do so a MALT1 activity assay which measures the proteolytic activity of MALT1 was performed. It was expected to see a lower proteolytic activity in cells treated with the drugs Mepazine and Biperiden compared to the ones that were treated with the carrier substance DMSO.
To examine the nuclear translocation of c-Rel two different cell lines, Panc-1 and L3.6pl Res, were treated for different periods of time with the above mentioned drugs. By performing nuclear fractionation and subsequent western blotting the quantity of c-rel in the nucleus was measured.
As a result, the experiments showed that after the treatment with Biperiden and Mepazine for 15 minutes there was a significant smaller amount of c-Rel in the nuclear fraction than in the cells of the Panc-1 cell line which were treated with DMSO.
The working hypothesis was confirmed and it was assumed that by treating the cells with Mepazine and Biperiden MALT1 is inhibited and therefore less c-Rel is able to translocate to the nucleus. This entails in the absence of the antiapoptotic effect of c-Rel and thus pancreatic cancer cells will be able to undergo apoptosis again.
Also for the other examined cell line, L3.6pl Res, it was possible to show an inhibiting tendency to the nuclear translocation of c-rel after treating the cells with Mepazine and Biperiden individually. The result of these tests showed that the treatment with the combination of Biperiden and Mepazine causes for an increase of c-rel in the nucleus in both investigated cell lines.
The outcomes of this thesis could be pioneering and deliver starting points for the development of new treatment options for pancreatic cancer and other malignant diseases.
