In dieser Arbeit sollte untersucht werden, welche Mechanismen auf CD4+ T Zellebene zur Auslösung von hepatischer Autoimmunität führen und folglich für die Durchbrechung von hepatisch induzierter Toleranz relevant sind. Als Mausmodell für unsere Analysen dienten CRP-MBP x tg4 Mäuse, die zusätzlich zum hepatisch exprimierten MBP über einen MBP-spezifischen T Zell-Rezeptor und somit über autoreaktive MBP-spezifische T Zellen verfügen. Wir konnten zeigen, dass nahezu alle CD4+ T Zellen von Mäusen dieser Linie den transgenen MBP-spezifischen T Zell Rezeptor exprimierten und dass die MBP-spezifischen T Zellen nach Antigen-spezifischer und unspezifischer in vitro Stimulation pro-inflammatorische Zytokine wie IFN-ɣ und IL-17 produzieren konnten. Allerdings waren die Mäuse weiterhin vor einer EAE und Leberentzündung geschützt, auch nach EAE-Induktion mittels MBP-Immunisierung. Zudem fanden sich in den CRP-MBP x tg4 Mäusen erhöhte Frequenzen MBP-spezifischer Treg. Nach in vivo Treg-Depletion mittels anti-CD25 Antikörper (PC-61) waren die Mäuse weiterhin vor einer EAE und Leberentzündung geschützt. Ebenso konnte in Mäusen, die aufgrund TGF-ß-insensitiver T Zellen eine defizitäre Induktion von peripheren Treg aufwiesen (CRP-MBP x hCD2-∆κTßR II x tg4), keine EAE oder Leberentzündung induziert werden. Auch bei defizitärer Wirkung des anti-inflammatorischen Zytokins IL-10, entweder durch Behandlung mit anti-IL-10-Rezeptor Antikörper oder bei IL-10-insensitiven T Zellen (CRP-MBP x cd4-DN-IL10R x tg4), überdauerte die Toleranz. Da die T Zellen der CRP-MBP x tg4 Mäuse zwar MBP-spezifisch in vitro stimulierbar waren, aber in vivo keine EAE oder Leberentzündung induzieren konnten, vermuteten wir, dass die endogenen T Zellen durch das hepatisch exprimierte MBP Toleranz erworben hatten. Um zu untersuchen, ob nicht-tolerante MBP-spezifische CD4+ T Zellen in Mäusen mit hepatisch exprimiertem MBP Toleranz erwerben, transferierten wir MBP-spezifische CD4+ T Zellen aus tg4 Mäusen in CRP-MBP Mäuse. Wir konnten zeigen, dass ein großer Teil der transferierten Zellen einen Tr1-ähnlichen Phänotyp annahm (CD49b+LAG-3+Foxp3-) und somit das hepatisch exprimierte MBP zur peripheren Induktion einer potenziell anti-inflammatorischen Tr1-Population führte. Die Versuchstiere entwickelten nach Transfer keine EAE, jedoch konnten transient leicht erhöhte Transaminasen und entzündliche Infiltrate in der Leber nachgewiesen werden.
Unsere Befunde machen deutlich, dass die isolierte Hemmung anti-inflammatorischer Zytokine wie TGF-ß und IL-10 nicht zur Durchbrechung von hepatischer Toleranz ausreicht und nicht allein Treg für die Aufrechterhaltung der Antigen-spezifischen Toleranz verantwortlich sein können. Darüber hinaus scheint die periphere Konversion von CD4+ T Zellen zu Tr1-phänotypischen Zellen ein weiterer möglicher Mechanismus hepatisch induzierter Toleranz zu sein. Somit ist davon auszugehen, dass eine Kombination der genannten Faktoren für die Induktion und Persistenz der hepatischen Toleranz verantwortlich ist.

The liver is able to induce antigen-specific tolerance. Regulatory T cells (Treg) are known to be important mediators in the process of inducing hepatic tolerance. In a mouse model of autoimmune neuroinflammation, the Experimental Autoimmune Encephalomyelitis (EAE), it was shown that hepatic expression of myelin basic protein (MBP) led to antigen-specific tolerance and protection from EAE induction. This tolerance was mediated by TGF-ß-induced MBP-specific Treg (Lüth et al. 2008).
In this work, we wanted to identify the CD4 T cell mechanisms that are responsible for the induction of hepatic autoimmunity and consequently for the breakthrough of hepatic tolerance. In our studies, we used the CRP-MBP x tg4 mouse model that is characterized by autoreactive MBP-specific T cells due to a transgenic T cell receptor and by MBP expression in the liver. We hope to provide answers to the following questions:
1. Do CRP-MBP x tg4 mice feature MBP-reactive T cells that recognize MBP in the central nervous system (CNS) and in the liver and does this result in spontaneous inflammation in the CNS or the liver?
2. Do these MBP-reactive T cells induce an inflammatory response against MBP in CNS and liver after EAE induction by immunization with MBP?
3. How important are Treg and the cytokines IL-10 and TGF-ß for the emergence and maintenance of hepatic tolerance?
4. Do naive MBP-specific T cells evolve to either pro-inflammatory T cells (Th1, Th17) or regulatory T cells (Treg, Tr1) after being transfered in mice with hepatic MBP expression?
Our studies show that CD4 T cells of CRP-MBP x tg4 mice consistently express the transgenic MBP-specific T cell receptor. Furthermore, the MBP-specific T cells were able to produce pro-inflammatory cytokines as IFN-ɣ and IL-17 after antigen-specific and non-specific in vitro stimulation. However, CRP-MBP x tg4 mice were consistently protected from EAE induction. No inflammatory response was detected in the liver, even after MBP immunization. Additionally, we found increased MBP-specific Treg frequencies in CRP-MBP x tg4 mice. In order to examine the importance of Treg for hepatic tolerance, we analyzed CRP-MBP x tg4 mice that had been treated with anti-CD25 antibody (PC-61) for in vivo depletion of Treg. In these mice no EAE was inducible and there was no inflammatory response in the liver. Likewise, CRP-MBP x hCD2-∆κTßR II x tg4 mice with impaired peripheral Treg induction due to TGF-ß-insensitive T cells were consistently protected from EAE and inflammation of the liver.
Hepatic tolerance was also persistent in mice with impaired signalling of the anti-inflammatory cytokine IL-10: CRP-MBP x tg4 mice treated with anti-IL-10-receptor antibody as well as mice with IL-10-insensitive T cells CRP-MBP x cd4-DN-IL10R x tg4 showed persistent tolerance.
As T cells of CRP-MBP x tg4 mice were MBP-specific and reactive to stimulation but not susceptible to EAE or liver inflammation in vivo, we supposed that the endogenous T cells were consistently tolerant due to the hepatic MBP expression. To exclude potentially confounding effects due to inherent tolerance of the T cells in these mice, we transferred non-tolerant MBP-specific CD4 T cells from tg4 mice into mice with hepatic MBP expression (CRP-MBP x B10.PL). We found that a large proportion of the transferred cells developed a Tr1-like-phenotype (CD49b+LAG-3+Foxp3-), suggesting that hepatic expression of MBP might induce an anti-inflammatory Tr1-population. After cell transfer the mice were protected from EAE. Concerning liver inflammation we found inflammatory infiltrates in the liver and slightly elevated transaminases in the blood.
Our findings show that neither the isolated inhibition of anti-inflammatory cytokines, such as TGF-ß and IL-10, nor Treg impairment alone are sufficient causes for breaking hepatic tolerance. Moreover, the peripheral conversion of CD4 T cells to Tr1-like cells appears to be an additional mechanism of liver tolerance. However, the exact mechanisms of liver tolerance remain unclear. We thus suppose that a combination of the mentioned factors is responsible for induction and persistence of hepatic tolerance.
