Zahlreiche Studien
haben angedeutet, dass das IL-1-System eine Rolle in den Zell-Zell-Interaktionen
im Hoden innehat. Um eine testikuläre zelluläre Quelle von IL-1
zu identifizieren, wurden Sertoli-Zellen aus präpubertalen Mäusen
isoliert, gereinigt und in vitro kultiviert. Es wurde untersucht, ob murine
Sertoli-Zellen in der Lage sind, IL-1 (alpha, beta und ra) zu produzieren.
Um einen möglichen Effekt des IL-1-Systems auf präpubertale Sertoli-Zellen
nachzuweisen, wurde die Transferrin-Sekretion in Sertoli-Zellkulturen in
Abhängigkeit von IL-1 bestimmt. Immunhistochemische Färbungen,
Enzymimmunoassays und RT-PCR wurden als Nachweismethoden gewählt.
Die
vorliegende Arbeit zeigte, dass präpubertale Maus Sertoli-Zellen in
vitro IL-1 alpha, nicht jedoch IL-1 beta produzieren. Die Produktion von
IL-1 alpha in Sertoli-Zellen kann durch LPS, IL-1 alpha und beta, nicht
jedoch durch FSH, in jeweils unterschiedlicher Kinetik stimuliert werden.
Dabei ließ sich demonstrieren, dass der stimulatorische Effekt von
IL-1 alpha auf die IL-1 alpha-Produktion der Sertoli-Zellen durch Zugabe
von IL-1ra zu Sertoli-Zell-Media vollständig reversibel ist.
Des
weiteren zeigte diese Arbeit, dass präpubertale Sertoli-Zellen eine
testikuläre Quelle des IL-1ra darstellen. IL-1ra-Spiegel in Sertoli-Zellkulturen
konnten sowohl durch FSH, LPS, IL-1 alpha, als auch durch IL-1 beta gesteigert
werden.
Die
vorliegende Arbeit liefert Hinweise darauf, dass weder IL-1 alpha noch
IL-1ra aus präpubertalen Maus Sertoli-Zellen sezerniert werden.
Allein
IL-1 alpha, nicht jedoch IL-1 beta oder IL-1ra, zeigte stimulatorische
Effekte auf die Transferrin-Produktion in präpubertalen Sertoli-Zellen.
Die IL-1 alpha-induzierten Transferrin-Spiegel entsprachen denen FSH-stimulierter
Sertoli-Zellen. FSH und IL-1 alpha zeigten weder additive noch synergistische
Effekte auf die Transferrin-Produktion in Sertoli-Zellen.
Die
Ergebnisse der vorliegenden Arbeit können helfen, die Aktivität
von IL-1 im Hoden zu evaluieren und die Rolle dieses bislang als immunmodulatorisch
bekanntem Peptid im männlichen Reifungsprozess zu bestimmen.

Numerous
studies have indicated that the IL-1-system inherits a role in the cell-cell-interactions
within the testis. To identify a testicular source of IL-1, Sertoli-cells
were isolated from prepubertal mouse testes, purified and cultured under
in vitro conditions. It was examined if murine Sertoli-cells are capable
of producing IL-1 (alpha, beta and ra). To study possible effects of the
IL-1-system on prepubertal Sertoli-cells, the Transferrin-secretion of
Sertoli-cell-cultures in dependence of IL-1 stimulation was measured. Immunohistochemical
staining, ELISA and RT-PCR techniques were used.
The present study revealed that prepubertal
mouse Sertoli-cells produce IL-1 alpha but not IL-1 beta in vitro. The
production of IL-1 alpha in Sertoli-cells can be stimulated by LPS, IL-1
alpha and beta, but not FSH, in specific time and concentration patterns.
It was demonstrated that the stimulatory effect of IL-1 alpha on the IL-1
alpha production of Sertoli-cells can be completely reversed by the addition
of IL-1ra to Sertoli-cell media.
Furthermore this study revealed that prepubertal
Sertoli-cells are a testicular source of IL-1ra. The levels of IL-1ra in
Sertoli-cell cultures were enhanced by FSH, LPS, IL-1 alpha as well as
IL-1 beta.
The present study provides data that neither
IL-1 alpha nor IL-1ra are secreted out of prepubertal mouse Sertoli-cells.
Only IL-1 alpha, not IL-1 beta nor IL-1ra
showed stimulatory effects on the Transferrin production in prepubertal
Sertoli-cells. IL-1 alpha-stimulated Transferrin-levels were equivalent
to those of FSH-stimulated Sertoli-cells. Neither additive nor synergistic
effects could be demonstrated on the stimulatory action of FSH and IL-1
alpha on the transferrin production of Sertoli-cells.
The present study may help to better evaluate
the action of IL-1 and the role of this classically known as immunomodulatory
peptide within the male process of maturation.
