In der vorliegenden Arbeit wird über neue Naturstoffe aus marinen Invertebraten des Nord-
Atlantiks berichtet. Der Schwamm Phakellia ventilabrum, die Koralle Anthothela grandiflora
und die Tunicate Ascidia virginea wurden im Hinblick auf ihre Inhaltsstoffe untersucht.
Sesquiterpen-Isocyanide sind aus mehreren Spezies der Familie Axinellidae bekannt, zu der
auch der bisher nicht untersuchte Schwamm P. ventilabrum gehört. Die Isocyanide sind häufig
mit den Isothiocyanaten und Formamiden vergesellschaftet, die das gleiche Sesquiterpen-
Skelett aufweisen. Das Naturstoffinventar in P. ventilabrum weicht von diesem Muster ab. In
diesem Schwamm konnten erstmals Quartette aus strukturanalogen Isocyaniden, Isocyanaten,
Isothiocyanaten und Dichloriminen nachgewiesen werden. Aus dem komplexen Gemisch dieser
Inhaltstoffe ließen sich durch präparative Gaschromatographie zehn Substanzen reingewinnen,
die durch zweidimensionale NMR-Experimente vollständig charakterisiert wurden. Unter
diesen befanden sich die fünf bekannten Verbindungen Axisonitril-1 (24), Axisonitril-4 (28),
Acanthellin-1 (51), Acanthen B (52), sowie das 10-Isocyano-muurolan 45. Fünf weitere
Strukturen sind neu: die Isocyanide mit Cubeban- (31), Isodaucan- (41) und Eudesman-Skelett
(49), sowie die zwei Isocyanate Axisocyanat-1 (25) und das Eudesman-Derivat 53.
Strukturen von Neben- und Spurenkomponenten, die aufgrund ihrer geringen Konzentration
nicht in ausreichender Menge isolierbar waren, um für NMR-Experimente zur Verfügung zu
stehen, konnten durch indirekte Strukturbeweise bestimmt werden. Hierzu wurden isolierte
Isocyanide, deren Strukturen eindeutig zugeordnet worden waren, anhand von Derivatisierungsreaktionen
unter Erhalt des jeweilgen Sesquiterpen-Skeletts in die strukturanalogen Isocyanate,
Isothiocyante und Dichlorimine umgewandelt. Der gaschromatographische und massenspektrometrische
Vergleich dieser Reaktionsprodukte mit authentischem Material in den Extrakten
bestätigte die Anwesenheit weiterer 18 Sesquiterpene, von denen 10 neue Verbindungen sind.
Aus A. grandiflora wurden zwei neue monooxygenierte Sesquiterpene durch Reversed-Phase-
HPLC isoliert. Anhand ihrer NMR-Spektren wurden diese als 6-(1-Methylethyl)-4-methyl-
5a,6,7,8-tetrahydro-5H-naphtho[1,8-bc]furan (70), sowie 6-(1-Methylethyl)-4-methyl-7,8-
dihydro-6H-naphtho[1,8-bc]furan (71) identifiziert. Es handelt sich hierbei um ein Vinylfuran
und ein Benzofuran mit Cadinan-Skelett. Die Strukturverwandtschaft beider Verbidungen
konnte durch Umsetzung von 70 mit DDQ in sein Dehydro-Derivat 71 bestätigt werden. Aus der Tunicate Ascidia virginea wurden die neuen Alkaloide Ascidin A (83) und B (84)
isoliert und identifiziert. Diese intensiv roten Farbstoffe mit neuer Azachinon-Struktur erwiesen
sich im Zellkultur-Test mit den Zell-Linien HELA (humanes Zervixkarzinom) und JURKAT
(Zellen der humanen T-Zellen-Leukämie) als aktive Cytostatika.
Mit Hilfe der hochaufgelösten APCI-MS im Negativionen-Modus wurden die
Elementarzusammensetzungen von 83 und 84 zu C15H9NO5 und C15H8INO5 ermittlet. Eine
zweifach iodierte Verbindung C15H7I2NO5 (Ascidin C, 85) ließ sich zusätzlich bestimmen.
Die zweidimensionalen NMR-Spektren DQF-COSY, HMQC, HMBC und NOESY
ermöglichten die Strukturaufklärung von 83 und 84. Ascidin A (83) ist ein Isochinolin-3,7-dion,
das an C-4 einen 4’-Hydroxyphenyl-Rest trägt und an den Positionen 1 und 8 hydroxyliert ist.
Ascidin B (84) ist das 3’-Iodo-Derivat von 83.
Das doppelt iodierte Ascidin C (85) ist eine Spurenkomponente, die nicht in ausreichender
Menge zur Verfügung stand, um NMR-spektroskopisch charakterisiert zu werden. Dass es sich
hierbei um 3’,5’-Diododerivat von 83 handelt konnte durch Derivatisierung von Ascidin A (83)
gezeigt werden. Die Umsetzung von 83 mit Bispyridiniodonium Tetrafluoroborat (IPy2BF4,
Barluengas Reagenz) lieferte die iodierten Ascidine B und C.
Drei weitere Spurenkomponenten Ascidin D, E und F (86-88) werden auf der Basis
massenspektrometrischer Daten postuliert. Die Massen der Molekülionen und ihre
Isotopenmuster ließen vermuten, dass es sich hierbei um 3’-Bromo-, 3’,5’-Dibromo- und
3’-Iodo-5’-bromo-83 handelt.

This thesis presents results on the isolation and structure elucidation of natural products from
marine invertebrates of the North Atlantic Ocean. The sponge Phakellia ventilabrum, the
gorgonian coral Anthothela grandiflora and the tunicate Ascidia virginea were investigated with
respect to their secondary metabolites.
The co-occurrence of isocyanides, isothiocyanates and formamides, bearing the same carbon
skeleton is known from sponges of the family Axinellidae. This family includes P. ventilabrum,
which has not yet been chemically investigated. In P. ventilabrum the range of metabolites
differs from this well-known pattern, as quartets of structurally related isocyanides, isocyanates,
isothiocyanates and dichloroimines could be established.
From the complex mixture of sesquiterpenes, ten compounds could be isolated by preparative
gas chromatography. Their structures were elucidated by two-dimensional NMR experiments.
Among them were five known compounds: axisonitrile-1 (24), axisonitrile-4 (28), acanthellin-1
(51), acanthene B (52), as well as the 10-isocyano-muurolane 45. Another five of the isolated
sesquiterpenes are new: three isocyanides showing a cubebane- (31), an isodaucane- (41) and a
eudesmane-skeleton (49) and the two isocyanates axisocyanate-1 (25) as well as the eudesmane
derivative 53.
Minor and trace compounds, which could not be isolated because of their low concentration,
were not characterised by means of NMR-spectroscopy. The structures of these compounds
were elucidated by derivatising the isolated isocyanides into their isocyanate, isothiocyanate and
dichloroimine analogues. Comparison of gas chromatographical and mass spectroscopical data
of the reaction products with the compounds present in the natural extract revealed the
structures of additional 18 isocyanates, isothiocyanates and dichloroimines, of which ten are
new natural products.
Two new mono-oxygenated sesquiterpenes, 70 and 71, could be isolated from the extracts of
A. grandiflora by reversed phase HPLC. Structure elucidation of the compounds, based on two
dimensional NMR experiments, proved 70 to be 6-(1-methylethyl)-4-methyl-5a,6,7,8-
tetrahydro-5H-naphtho[1,8-bc]furan and 71 to be 6-(1-methylethyl)-4-methyl-7,8-dihydro-6Hnaphtho[
1,8-bc]furan. Both, the vinyl furan 70 and the benzofuran 71, show a cadinane
skeleton. Their structural relation could be shown by reaction of 70 with DDQ, to yield its
dehydro derivative 71. Two new alkaloids, named ascidine A (83) and B (84), were isolated and identified from the
tunicate Ascidia virginea. These intensively red coloured compounds showing a novel azaquinone
structure exhibited potent anticancer activity when tested in HeLa (human cervix
carcinoma) and Jurkat (human T-cell leucemia) bio assays.
High resolution APCI-MS of the negative molecular ions revealed 83 to be C15H9NO5 and 84
C15H8INO5. Furthermore a doubly iodinated compound, C15H7I2NO5 (ascidine C, 85), was
detected.
The two dimensional NMR spectra DQF-COSY, HMQC, HMBC and NOESY allowed the
structure elucidation of 83 and 84. Ascidine A (83) is a isoquinoline-3,7-dione, bearing a 4’-
hydroxyphenyl moeity at C-4 and is hydroxylated at C-1 and C-8. Ascidine B (84) is its 3’-iodo
derivative.
The doubly iodinated ascidine C (85) occurs as a trace compound, which could not be isolated
and characterised by NMR-spectroscopy. Its identity with 3’,5’-iodo-83 could be shown by
reaction of 83 with bispyridine iodonium tetrafluoroborate (IPy2BF4, Barluenga’s reagent)
which yielded the iodinated ascidines B and C.
Three further trace compounds, ascidine D, E and F (86-88), could be detected by means of
negative ion mass spectrometry. Their molecular ions and their isotopic pattern propose their
structures to be 3’-bromo-, as well as 3’,5’-dibromo- and 3’-iodo-5’-bromo-83.
