dc.description.abstractDie Wechselwirkung der pathogenen und der zellulären Isoform des Prionproteins ist das Schlüsselereignis in der Pathogenese der Transmissiblen Spongiformen Enzephalopathien (TSE), zu denen die Creutzfeldt-Jakob-Krankheit (CJK) als humane Variante zählt. Im Rahmen der Dissertation sollten peptidische Inhibitoren des Anlagerungsprozesses der beiden Konformationsisomere gefunden werden. Dazu wurde eine Bibliothek von 28 sich überlappenden Oktapeptiden aus dem globulären C-Terminus der humanen Sequenz des Prionproteins (V121-L242) synthetisiert. Die Oktapeptide wurden hinsichtlich ihrer Bindungsaktivität gegenüber rhPrPC mittels surface plasmon resonance (SPR) untersucht. Für die drei Regionen V121 R136 (a), Y149-R164 (b) und S231-L242 (c) konnte eine Interaktion mit dem Protein nachgewiesen werden. Die Peptide Region (b) YYRENMHR (8), NMHRYPNQ (9) und YPNQVYYR (10) zeigten mit 21 µM, 25 µM und 164 µM die besten Dissoziationskonstanten. Die Zusammensetzung der sich überlappenden Peptidfragmente 8 und 9 zum 12er Peptid 29 führte zu keiner Verbesserung der Bindungseigenschaft. SPR-Studien zeigten, dass die Peptide mit dem Proteinfragment PrP(90 231) interagieren. Zur Erhöhung der oralen Bioverfügbarkeit wurde im Peptid 9 das Tyrosin durch 4 Fluorphenylalanin (9a) und 4 Trifluormethylphenylalanin (9b) ersetzt. Die SPR-Bindungs-studien der Fluorderivate zeigen, dass das Tyrosin in der Wechselwirkung als Donor in einer Wasserstoffbrückenbindung fungiert. In Peptid 10 wurden alle Tyrosine durch 4 Fluorphenylalanin substituiert (10a), dieses Derivat zeigte keine spezifische Bindung mehr. Für das Peptid 9a, das in den SPR-Experimenten eine im Vergleich zu 9 dreifach schnellere Bindungskinetik aufwies, konnte das Bindungsepitop mittels STD NMR Spektroskopie charakterisiert werden. Das epitope mapping deutet auf eine Beteiligung der Sequenz RYP hin. Zur Bestätigung des Bindungsepitops wurde das Peptid RYP (30) synthetisiert. Die im Vergleich zu Peptid 9 50 fach schlechtere Dissoziationskonstante zeigt allerdings, dass die Sequenz RYP nur einen Teil des gesamten Epitops darstellt. In einem in vitro Aggregation-Inhibitions-Essay, in dem die Aggregation von rhPrP(90-231) durch metallinduzierte Oxidation eingeleitet wurde, konnte gezeigt werden, dass die Aggregation durch die Zugabe von Peptid 9 im siebenfachen Überschuss zu 80% inhibiert wird. Die Verbindung stellt somit einen potentiellen anti-CJK-Wirkstoff dar. Um den Einfluss der Glycosylierungsstelle an N197 auf die Bindungsaktivität und die Struktur des Peptidfragmentes T192-T201 zu untersuchen, wurden das Glycopeptid TTKGEN[b-D-GlcNAc-(1-4)-b-D-GlcNAc]FTET 32 und das entsprechende unglycosylierte Referenzpeptid 31 dargestellt. Die SPR-Bindungsstudien mit rhPrP(23-231) wiesen auf eine spezifische Interaktion für das Glycopeptid (455 µM) und eine unspezifische Wechselwirkung des Referenzpeptids hin. Demzufolge nimmt der Zucker entweder direkt an der Bindung teil oder stabilisiert die aktive Konformation des Peptids. Anhand von NOE-Aufbaukurven und 3JNH,Ha-Kopplungskonstanten wurde eine Konforma-tionsanalyse von 31 und 32 durchgeführt. Es ergaben sich jeweils ca. 45 interatomare Abstände, die zusammen mit den Winkelvorgaben in DG-Rechnungen einflossen, die mit dem Programm DYANA durchgeführt wurden. Die backbones der zehn besten Strukturen wurden jeweils gefittet. Beide Peptide zeigten nur geringe konformelle Abweichungen der Peptidrückgrate (rmsd < 1.7 Å), die Strukturen sind demnach gut definiert. Der Vergleich ergibt, dass die Struktur des Glycopeptids deutlich gestreckter ist. In den energieminimierten Endstrukturen einer sich anschließenden constrained MD-Simulationen sind die Vorgaben gut erfüllt. Die Strukturen weisen deutliche konformelle Unterschiede insbesondere im Bereich der Glycosylierungsstelle auf, womit die unterschied-lichen Bindungsspezifitäten von 31 und 32 gegenüber rhPrPC erklärt werden könnten. Ein zweiter Teil der Arbeit beschäftigt sich mit der Entwicklung des heteronuklearen 1D-19F{1H}-STD Experiments. Durch inverse gated Entkopplung der Protonen während der Akquisition wurde dabei eine Erhöhung der Empfindlichkeit des Experiments erreicht. Das Pulsprogramm wurde in ein Pseudo-2D-STD-Experiment umgeschrieben, durch das ebenfalls das off-resonance Spektrum zugänglich ist, das als Referenz für die Auswertung der Daten erforderlich ist. Dieses Messverfahren bietet daher eine Zeitersparnis. Als zusätzliche Information erhält man eine Aussage über das Vorzeichen der Signale. Das Pulsprogramm wurde an verschiedenen Rezeptor-Ligand-Systemen getestet: RCA120/6-Deoxy-6-fluoro-D-galactose bzw. 3-Deoxy-3-fluoro-D-galactose, BSA/Tryptophan und rhPrP(23-231)/Fluorderivat 9a. Alle vier Systeme zeigten negative Signale im 19F{1H} STD-Spektrum entsprechend einer Signalverstärkung im on-resonance-Spektrum. Trotz des bislang ungeklärten Vorzeichens kann das Verfahren zum screening fluorierter Liganden eingesetzt werden.de
dc.description.abstractThe interaction of the abnormal and the cellular isomer of the prion protein is the crucial event for the pathogenesis of transmissible spongiform encephalopathies (TSE), among them the Creutzfeldt-Jakob-Disease (CJD) is the human variant. In the course of this thesis peptide inhibitors for the interaction of the two conformational isomers should be found. Therefore, a library of 28 peptides of the C-terminal folded domain of the human prion protein (V121-L242) was synthesized each sequence with an overlap of four amino acids. The octapeptides were analyzed with respect to their binding affinity to rhPrPC by surface plasmon resonance (SPR). Three regions V121 R136 (a), Y149-R164 (b) and S231-L242 (c) showed a significant interaction. The peptides of the region (b) YYRENMHR (8), NMHRYPNQ (9) and YPNQVYYR (10) showed the best dissociation constants of 21 µM, 25 µM and 164 µM, respectively. The dodecapeptide 29 which is composed of the overlapping peptides 8 and 9 does not indicate increased binding properties. SPR studies show that the peptides interact with the shortened protein fragment PrP(90-231). In order to enhance the bioavailability of peptide 9 the tyrosine in the sequence was substituted by 4-fluoro-phenylalanine (9a) or 4-trifluoromethyl-phenylalanine (9b). SPR binding studies suggest, that the tyrosine residue directly interacts with the protein as a hydrogen bond donor. In the peptide 10 all tyrosine residues were substituted by 4 fluoro-phenylalanine (10a). This derivative did not show any characteristics of specific binding. The binding epitope of peptide 9a could be characterized by STD NMR spectroscopy. The epitope mapping suggests a contribution of the sequence RYP to the binding. In order to confirm the epitope the peptide RYP (30) was synthesized. The dissociation constant of 30 is 50 fold inferior to that of 9, which suggests, that the sequence RYP represents only a part of the binding epitope. In an in vitro aggregation-inhibition-assay, where aggregation of the prion protein was induced by metal-catalyzed oxidation, it could be shown, that aggregation could by inhibited up to 80% by addition of peptid 9 in a sevenfold excess. Hence, the compound is a potential anti-CJD drug. In order to study the influence of the glycosylation site at N197 on the binding activity and the structure of the peptide fragment T192-T201 the glycopeptide TTKGEN[b-D-GlcNAc-(1-4)-b-D-GlcNAc]FTET 32 and the unglycosylated reference peptide 31 were synthesized. SPR binding studies with rhPrP(23-231) showed a specific interaction for the glycopeptide (KD = 455 µM), while the interaction of the reference peptide was found to be unspecific. Thus, the saccharide interacts directly with the protein or stabilizes the active conformation of the peptide. A conformational analysis of 31 and 32 was carried out by analysis of NOE build-up curves, which yielded about 45 interatomic distances for both peptides. From 3JNH,Ha coupling constants dihedral angle constraints could be calculated. Both, the distance range and the angle constraints were incorporated in distance geometry calculations carried out with the program DYANA. The ten best structures were selected and the backbones were fitted. Both peptides show only small conformational variations of their backbones (rmsd < 1.7 Å). Thus, the structures seem to be well defined. When comparing the structures of 31 and 32 it becomes evident, that the glycopeptide is more stretched. The energy minimized final structures of the subsequent MD simulation were in good agreement with the experimentally determined constraints. The structures clearly exhibit conformational differences especially for the residues in the neighborhood of the glycosylation site. This yields a possible explanation for their different binding specifities with respect to rhPrPC. A second part of the thesis dealt with the development of a heteronuclear 1D-19F{1H}-STD experiment. The application of an inverse gated decoupling scheme for proton resonances lead to a higher signal intensity and thus to an enhanced sensitivity of the experiment. In order to get a more precise insight into the results of the spectra the pseudo-2D-STD experiment was developed. The acquisition of these experiments admits the access of the off-resonance spectrum which is required for the analysis of the STD data. Hence, this method reduces acquisition time. Additionally, the sign of the STD signals can be deduced from the spectra. The novel pulse program was applied to various receptor ligand systems: RCA120/6-desoxy-6-fluoro-D-galactose and 3-desoxy-3-fluoro-D-galactose respectively as well as BSA/tryptophane and rhPrPC/fluorine derivative 9a. The spectra of each system showed negative signals, indicating an enhancement of the signal intensity in case of on-resonance irradiation. However, the method can be used for the screening of fluorinated ligands even though the sign of the signals remains unclear.en
