In recent years, the percentage of highly potent active pharmaceutical ingredients among the newly developed drugs has increased significantly. Thus, safe handling is becoming increasingly important to protect employees as well as patients. The major hazard is airborne dust generated during handling of these compounds. In the present work, the generation of dust and the subsequent spatial distribution of airborne dust containing active pharmaceutical ingredients were investigated in three studies using a newly developed chamber setup. With this setup, a powder sample is atomized within an emission chamber and then passes through a defined orifice into a detection chamber, in which the spatial distribution of the dust emission may be detected using nine filter-based measurement spots. In a first study, the proof of concept of the chamber setup was focused on. Experiments with the surrogate substance acetaminophen showed that reproducible measurements of both dust emission of very small quantities of the active pharmaceutical ingredient as well as its spatial distribution are possible with the chamber setup. A linear relationship between the sample mass and the total dust detected was shown. Interestingly, the spatial distribution was not as expected, with the highest concentration close to the emission site, but the opposite. This finding was verified by computational fluid dynamics simulations and it was demonstrated that the distribution of the aerosol corresponds to the airflow generated in the chamber setup. Based on these results, the influence of the atomization parameters on the measured dust emission and the spatial dust distribution was investigated in detail in a second study. With a statistical experimental design, the linear relationship between sample mass and detected dust emission was confirmed and an interaction between the compressed air volume flow used for atomization and the atomization duration was found with regard to both the detected dust emission and the spatial dust distribution. In addition, there was an influence of the particle size distribution and the particle shape of the sample on the measured dust emissions. Based on these results and an additional optimization of the measurement procedure, the detected dust emission could be increased more than tenfold at a constant sample mass and the influence of the atomization parameters on the spatial dust distribution was successfully characterized. In a third study, the newly developed chamber setup was equipped with laser-based optical sensors, which allow the detection of the fine fraction of the dust (particles < 2.5 µm). With these sensors, the immediate spatiotemporal detection of the dust distribution was possible. The results of the study not only confirmed the spatial dust distributions already observed in the first two studies but also showed that the spatial distribution occurs within the first few minutes after atomization and that there is a subsequent equilibration of the dust concentration in the measuring system. Moreover, the optical sensors allowed to investigate the dust emission of a binary powder blend, used for direct compression in tableting, consisting of fine particulate acetaminophen and coarse lactose. This powder blend showed a decreased dust emission compared to plain acetaminophen. In summary, the three studies demonstrated that with the novel chamber setup it is not only possible to investigate dust emissions, but also the spatial distribution of dust. This work thus provides important information on how to localize sources of contamination in a production environment using stationary measurements and how to possibly reduce dust exposure through directed airflow in production facilities to protect personnel and ultimately also patients.en
