dc.description.abstractIm Rahmen der vorliegenden Arbeit wurden für die humanen Inositol Hexakisphosphat Kinasen (IP6K, Isoformen 1, 2 und 3) bisher unbekannte Reaktionen identifiziert, die die Kinasen als zelluläre Energiesensoren charakterisieren. Beschrieben wurden diese Kinasen als pyrophosphorylierende Enzyme von Inositolhexakisphosphat (InsP6). Die Kinasereaktion verläuft in Gegenwart von ATP und die IP6K übertragen das γ-Phosphat von ATP auf die Position 5 des Substrates (InsP6) und bilden 5PP-InsP5. 5PP-InsP5 ist ein Isomer welches unter anderem unterschiedliche Proteine in ihrer Aktivität regulieren kann. Anhand dieser Arbeit wird deutlich, dass die Kinasen deren Substrat nicht nur pyrophosphorylieren, sondern in Gegenwart von ADP oder eines Gemisches aus ATP und ADP auch dephosphorylieren können. Das Hauptprodukt dieser neu identifizierten Reaktion ist Ins(2,3,4,5,6)P5 und als Nebenprodukte konnten D/L-Ins(1,2,3,4,5)P5 und Ins(1,2,3,4,6)P5 identifiziert werden. Allerdings stellt die IP6K keine klassische hydrolysierende Phosphatase dar, sondern überträgt das Phosphat zur ATP Synthese auf ADP. Das Hauptprodukt Ins(2,3,4,5,6)P5 stellt das enantiomere Produkt (Ins(1,2,4,5,6)P5) der bisher einzig bekannten InsP6 Phosphatase MINPP1 dar. Die Identifizierung beider HPLC-chromatographisch nicht trennbarer Produkte erfolgte mit Hilfe einer ebenfalls bisher nicht bekannten und während dieser Arbeit identifizierten Selektivität der Inositol Polyphosphat Multikinase (IPMK) gegenüber InsP5 Isomeren. Die IPMK als Hydroxykinase für InsP3 bzw. InsP4 Isomere phosphoryliert enantioselektiv das MINPP1 Produkt Ins(1,2,4,5,6)P5 zu InsP6, nicht aber das IP6K Produkt Ins(2,3,4,5,6)P5, sodass ein hier etablierter enatioselektiver Assay die eindeutige Zuordnung der isolierten Dephosphorylierungsprodukte ermöglicht. Weitere Untersuchungen zeigten, dass die IP6K beide Dephosphorylierungsprodukte von InsP6, also Ins(1,2,4,5,6)P5und Ins(2,3,4,5,6)P5 in Gegenwart von ATP als Hydroxylgruppenkinase wieder zurück zu InsP6 und 5PP-InsP5 rephosphorylieren kann. Die Umkehrreaktion der Kinasereaktion (s.o.), d.h. von 5PP-InsP5 zu InsP6 und weiter zu Ins(2,3,4,5,6)P5 wird ebenfalls von der IP6K bei Vorhandensein von ADP katalysiert. Es wurde somit eine von [ATP] bzw. [ADP] abhängige IP6K katalysierte Phosphorylierungs- bzw. Dephosphorylierungssequenz aus den Isomeren Ins(2,3,4,5,6)P5, InsP6 und 5PP-InsP5 identifiziert. Diese Phosphorylierungs- bzw. Dephosphorylierungssequenz verläuft allerdings nicht nur in Gegenwart von ausschließlich ATP oder ADP, sondern auch bei physiologisch relevanten Konzentrationsverhältnissen beider Nukleotide. Die Kombination von unterschiedlichen ATP und ADP Konzentrationen sowie während der IP6K katalysierten Reaktionen variierte Adenylat-Energiepotentiale ([ATP]/([ATP]+[ADP])) spiegeln sich in der Synthese bzw. Konzentrationsgleichgewichten der jeweiligen Isomere wieer. Niedrige Adenylat-Energiepotentiale begünstigen die Dephosphorylierungsreaktionen, hohe Adenylat-Energiepotentiale die Phosphorylierung bzw. Pyrophosphorylierung. Die mit rekombinant exprimierten und gereinigten IP6K Isoformen in vitro erhobenen Daten konnten durch die artifizielle Variation des Adenylat-Energiepotentials in unterschiedlichen Zelllinien untermauert werden. Es konnte gezeigt werden, dass auch dort die IP6K Adenylat-Energiepotential sensitive Kinasen darstellen, die auf Veränderungen des Energiepotentials mit veränderter Produktbildung wie in vitro gezeigt reagieren. Die IP6Ks als zelluläre Energiesensoren spielen im Organismus eine wichtige Rolle, da deren bisher bekanntes Produkt 5PP-InsP5 (gebildet bei hohem Adenylat-Energiepotential) z.B. die Transkription glykolytischer Enzyme in Hefen steuert und somit direkt auf die Energie erzeugende Glykolyse einwirkt. Hochinteressant ist auch, dass 5PP-InsP5 ebenfalls die Insulinsekretion aus pankreatischen β-Zellen stimuliert und gleichzeitig die Insulinsensitivität in peripheren Geweben reguliert, was diese Kinase als Target zur Behandlung des Typ II Diabetes höchst wertvoll machen könnte. Die mögliche biologische Funktion des in dieser Arbeit neu identifizierten und physiologisch relevanten Isomers Ins(2,3,4,5,6)P5 ist allerdings in weiteren Studien noch zu zeigen.de
dc.description.abstractIn the present thesis the human inositol hexakisphosphate Kinase (IP6K) Isoforms 1, 2 and 3 are identified as sensors for the cellular adenylate energy potential. The IP6K have previously been described as inositol hexakisphosphate (InsP6) pyrophosphorylating enzymes. In this reaction the IP6K transfer the γ-phosphate from ATP exclusively to the position 5 of the inositol phosphate to synthesize 5PP-InsP5. 5PP-InsP5 is an isomer which regulates the activity of several proteins. Due to the present thesis it becomes clear that theIP6K do not only phosphorylate their substrate but also dephosphorylate InsP6 in the presence of ADP. The main product of this reaction was identified as the enantiomer Ins(2,3,4,5,6)P5. The sencondary products from this new reaction were identiied as D/L-Ins(1,2,3,4,5)P5 and Ins(1,2,3,4,6)P5, respectively. The IP6K mediated dephosphorylation of InsP6 was shown to be a phosphate transfer from InsP6 to ADP resulting in ATP synthesis. Thus the IP6K do not act as classical hydrolyzing phosphatases for InsP6. The main product Ins(2,3,4,5,6)P5 was identified to be the enantiomer of Ins(1,2,4,5,6)P5 which is the sole product of the only known InsP6 phosphatase MINPP1. Due to the impossibility to distinguish enantiomers with standard HPLC-analysis, the clear assignment of the respective enantiomer was provided by a new identified and hitherto unknown catalytic activity of the inositol polyphosphate multikinase (IPMK). The IPMK selectively phosphorylates the 3-hydroxyl moiety of Ins(1,2,4,5,6)P5 to InsP6 and not the 1-position of Ins(2,3,4,5,6)P5.Thus the enantiomers could easily be identified by the increase of InsP6 after addition of IPMK. Further investigations revealed that both dephosphorylation products from InsP6, regardless of whether they derived from MINPP1 or IP6K catalyzed reactions, are re-phosphorylated to InsP6 and further to 5PP-InsP5 by IP6K. Thus IP6K is a pyrophosphorylating kinase and a hydroxyl moiety kinase for the positions 1 and 3 of InsP5 isomers. Anyway, the IP6K did not only catalyze the phosphorylation of Ins(2,3,4,5,6)P5 to InsP6 and to 5PP-InsP5 in the presence of ATP, it additionally catalyzes the backreaction (dephosphorylation) from 5PP-InsP5 to InsP6 and to Ins(2,3,4,5,6)P5 in the presence of ADP. Both reactions, phosphorylation and dephosphorylation, were observed with a combination of both, ATP and ADP in a physiological and pathological range. These results suggest that IP6Ks are sensors for the adenylate energy potential reacting with either dephosphorylation or phosphorylation of its substrates depending on the prevalent adenylate energy potential. The detailed experiments with the recombinant isoforms of the IP6K in vitro could be confirmed with different model cell lines. Artificial ATP depletion resulted in dramatic increase of the IP6K mediated dephosphorylation product Ins(2,3,4,5,6)P5. A specific, concomitant inhibition of cellular IP6K during ATP depletion did not result in such an increase. Emerging evidence suggest a pivotal role or IP6K as cellular adenylate energy sensors for the organism or the cell. The pyrophosphorylation product from InsP6, 5PP-InsP5, is synthesized at a high adenylate energy potential. It has previously been shown that 5PP-InsP5 inhibits the transcription of glycolytic enzymes and thus directly regulating the energy providing glycolysis in yeast. Moreover 5PP-InsP5 also stimulates the insulin secretion in pancreatic ß-cells and additionally regulates the sensitivity to insulin of cells in peripheral tissue. This highlights IP6K as potential drug target for treatment of diabetes type II. The biological function of the new identified physiological relevant isomer Ins(2,3,4,5,6)P5, synthesized at a low adenylate energy potential, remains elusive so far and needs to be further investigated.en
