Zusammenfassung: 
Da Kinase-Substrat-Interaktionen transient sind, wurden bislang nur wenige Strukturen von Kinase-Substrat-Komplexen aufgeklärt. Daher wurden γ-phosphatmodifizierte ATP-Analoga als kovalente Capture-Reagenzien zur Stabilisierung transienter Kinase-Substrat-Komplexe untersucht. Eine effiziente Bildung binärer Addukte mit den Kinasen LegK4 und CK2α′ konnte mittels Kristallstrukturanalyse und intakter Massen-spektrometrie nachgewiesen werden. Es konnte jedoch weder ein Phosphotransfer noch die Bildung eines ternären Komplexes nachgewiesen werden.
Die Phosphorylierung von Hsc70 an Thr495 durch LegK4 reduziert ATPase-Aktivität und Substratprozessierung, während die entsprechende AMPylierung in BiP vergleichbare Effekte ausübt. Die strukturelle Charakterisierung von an Thr495 modifiziertem Hsc70 war bislang jedoch aufgrund fehlender robuster präparativer Methoden zur Gewinnung ausreichend modifizierten und stabilen Proteins eingeschränkt. Im Rahmen dieser Arbeit wurde ein robustes präparatives Phosphorylierungsprotokoll entwickelt, das Milligramm-Mengen stabil phosphorylierten Hsc70 mit Modifikationsgraden von etwa 70 % für den Wildtyp und bis zu etwa 86 % für die ATPase-defiziente Mutante Hsc70T204A liefert. Entscheidend waren die Minimierung der Kinasekonzentration, ein hohes ATP/ADP-Verhältnis sowie die effiziente Entfernung von LegK4, um eine kinasev-ermittelte Dephosphorylierung zu verhindern. Parallel dazu wurde ein präparatives AMPylierungsprotokoll mit hoher Ausbeute etabliert, das AMPylierungsgrade von etwa 63 % erreichte, wobei sich die effektive Entfernung von FicD als entscheidend für die Stabilität der Modifikation erwies. Small-angle X-ray scattering, analytische Größenausschlusschromatographie und limitierte Proteolyse zeigen konsistent, dass die Phosphorylierung an Thr495 die Oligomerisierung reduziert und das Konformationsensemble selbst unter ADP-gebundenen und nukleotidfreien Bedingungen in Richtung eines kompakteren, domänengedockten, ATP-ähnlichen Zustands verschiebt. Nano differential scanning fluorimetry zeigt nur geringe Auswirkungen der Phosphorylierung auf die globale thermische Stabilität. Insgesamt legen diese Ergebnisse nahe, dass die Phosphorylierung die Funktion von Hsc70 primär durch die Verschiebung von Konformationsgleichgewichten und Oligomerverteilungen moduliert und nicht durch eine Destabilisierung des Proteins.

Because kinase-substrate interactions are transient, only a few kinase-substrate complex structures have been solved. Therefore, γ-phosphate-modified ATP analogs were explored as covalent capture reagents for stabilizing transient kinase-substrate complexes. Efficient binary adduct formation with the kinases LegK4 and CK2α′ was confirmed by crystal structure and intact mass spectrometry. However, no detectable phosphotransfer or ternary complex formation was observed.
In human Hsc70, phosphorylation at Thr495 by the Legionella effector kinase LegK4 has been reported to reduce ATPase activity and client processing, while AMPylation at the homologous site in the ER-resident Hsp70 BiP results in similar effects on chaperone function. Nevertheless, structural characterization of Hsc70 modified at Thr495 has been limited, largely due to the lack of robust preparative methods yielding sufficiently modified and stable protein. In this work, a robust preparative phosphorylation protocol was developed that yields milligram quantities of stably phosphorylated Hsc70 (pHsc70) with modification levels of ~70% (wild-type) and up to ~86% for the ATPase-deficient mutant Hsc70T204A. Critical parameters included minimizing kinase concentration, maintaining high ATP:ADP ratios, and efficient removal of LegK4 by affinity chromatography to prevent kinase-mediated dephosphorylation. In parallel, a high-yield AMPylation protocol was established, achieving ~63% AMPylation. Here, efficient depletion of FicD proved essential to maintain modification stability. Structural and bio-physical characterization focused on pHsc70. Small-angle X-ray scattering, analytical size-exclusion chromatography, and limited proteolysis consistently demonstrate that phosphorylation at Thr495 reduces oligomerization and shifts the conformational ensemble toward a more compact, domain-docked, ATP-like state, even under ADP-bound and nucleotide-free conditions. Nano differential scanning fluorimetry measurements indicate that phosphorylation has only a minor effect on global thermal stability. Together, these results suggest that phosphorylation modulates Hsc70 function primarily by altering conformational equilibria and oligomeric distributions rather than by destabilizing the protein.
