Titel: Synthese und Untersuchung des photochemischen Verhaltens von cyclischen Enonen in Gegenwart von Alkenylisocyanaten

Zusammenfassung: 
Ziel dieser Arbeit war es, das photochemische Verhalten der cyclischen Sechsring-Enone (1, 2, 3) in Gegenwart der unterschiedlich substituierten Isocyanate (4, 5, 6, 7) zu untersuchen. Im ersten Schritt werden die Photoedukte synthetisiert. Alle Verbindungen sind literaturbekannt und werden nach den üblichen Methoden dargestellt, mit Ausnahme von Verbindung 7 welche nicht dargestellt werden konnte.
Die photochemischen Untersuchungen von 2,3-Dihydro-2,2-dimethyl-4H-pyran-4-on (1) in Gegenwart der drei Isocyanate (4-6) ergaben ausschließlich [2+2]-Photoaddukte. Die Reaktionen verlaufen über eine 1,4-Cyclisierung und sind wenig selektiv. Anders als erwartet, stellten bei allen drei Umsetzungen die cis-Isomere die Hauptprodukte.
Für die Umsetzung von 1 in Gegenwart von 2-Propenylisocyanat (4) wurden sechs Produkte 24a-f im Verhältnis 5 : 4 : 3 : 3 : 1.5 : 1 vorgefunden. Bei der Umsetzung von 1 in Gegenwart von Vinylisocyanat (5) konnten fünf Produkte (27a-e) im Verhältnis 2.1 : 1.8 : 1.8 : 1 : 0.6 gefunden werden. Bei diesen Reaktionen dominieren die cis-Kopf-Schwanz-Addukte. Dies wird auf den wenig flexiblen sauerstoffhaltigen Sechsring zurückgeführt. Für die Umsetzung von 1 in Gegenwart von 2-Methylpropen-1-isocyanat (6) können auch fünf Produkte (28a-e) charakterisiert werden. Dabei fällt auf, dass hier die cis-Kopf-Kopf-Addukte gegenüber den cis-Kopf-Schwanz-Produkten dominieren. Für die endo-/exo-Konfiguration wurden wie erwartet die endo-Produkte als Hauptprodukte festgestellt.
Als nächstes wurden die Umsetzungen von 5,5-Dimethylcyclohex-2-enon (2) in Gegenwart der drei Isocyanate untersucht. Dabei wurde festgestellt, dass auch diese Reaktionen wenig selektiv sind. Für die Umsetzung von (2) mit 2-Propenylisocyanat (4) wurden vier Photoprodukte gefunden. Dabei stellte sich das endo-trans-Kopf-Schwanz-Isomer 29a als Hauptprodukt heraus, alle anderen Produkte sind cis-Isomere, eines davon ist ein Kopf-Schwanz-Addukt (29b) und die letzten beiden Kopf-Kopf-Addukte (29c und 29d). Bei der Umsetzung von 2 in Gegenwart von Vinylisocyanat (5) konnten drei Produkte (30a-c) im Verhältnis 1 : 0.9 : 0.7 charakterisiert werden. Eine weitere Umsetzung wurde durchgeführt, in der 2 und 2 Methyl-propenyl-1-isocyanat (6) miteinander reagierten. Die Auswertung ergab ein Gemisch aus vier Produkten (31a-c) im Verhältnis 1 : 0.9 : 0.6 : 0.5. Die Hauptprodukte waren hier cis-Isomere, wobei sich nur geringfügig mehr Kopf-Schwanz-Produkt 31d gebildet hat. Dass sich die trans-Isomere nur als Nebenprodukte gebildet haben, bestätigt die Annahme, dass sich ein stabiles Biradikal bei der photochemischen Umsetzung gebildet hat.
Als letztes wurden die photochemischen Reaktionen von 4,4-Dimethylcyclohex-2-enon 3 mit den Isocyanaten untersucht. Für die erste Umsetzung von 3 mit 2-Propenylisocyanat (4), konnten zwei Produkte 32a-b im Verhältnis 1 : 0.4 bestimmt werden. Beide Produkte sind Kopf-Schwanz-Isomere. Als Hauptprodukt wurde das endo-trans-Kopf-Schwanz-Produkt 32a gefunden. In der Umsetzung von 3 mit Vinylisocyanat (5) wurden fünf Produkte (33a-d) im Verhältnis 1.1 : 1 : 0.9 : 0.4 : 0.4 gefunden. Die Reaktion liefert zwei trans-Kopf-Schwanz-Produkte und drei Kopf-Kopf-Produkte, wobei zum ersten Mal ein trans-Kopf-Kopf-Produkt charakterisiert werden konnte. In der letzten Umsetzung von 3 wird die Bestrahlung in Gegenwart von 2 Methyl-propenyl-1-isocyanat (6) untersucht. Dabei liefert diese Reaktion drei Produkte (34a-c) im Verhältnis 1.1: 1: 0.7. Als Hauptprodukte wurden das exo-trans-Kopf-Schwanz-Isomer 34b und das endo-cis-Kopf-Schwanz-Isomer 34a gefunden. Als Nebenprodukt trans-Kopf-Kopf-Produkt 34c.
Alle Photoprodukte konnten nicht säulenchromatographisch getrennt werden, daher wurden sie aus den Rohgemischen direkt in Amine umgesetzt. Dabei stellte sich heraus, dass die Photoprodukte aus 2,3-Dihydro-2,2-dimethyl-4H-pyran-4-on (1) nicht in die Amine umgesetzt werden konnten, da sie sich während der Reaktion zersetzten. Für die Umsetzung aller anderen Photoprodukte konnte beobachtet werden, dass nur die Kopf-Schwanz-Produkte in Amine umgesetzt werden konnten. Dabei wurden die trans-Produkte zu cis-verknüpften Aminen isomerisiert. Die Kopf-Kopf-Produkte ergaben Zersetzungsprodukte, mit Ausnahme von 29c-d, die durch eine Retro-Mannich-Reaktion in ein Diketon 35b umgesetzt wurde. Für alle Amine 35a-b bis 40a-b konnten je zwei Isomere beobachtet werden, ein endo- und ein exo-Isomer. Da auch an dieser Stelle die Diastereomere nicht getrennt werden konnten, wurden sie durch Einführen einer Schutzgruppe derivatisiert.
Mit den eingeführten Schutzgruppen gelang es, einige der Isomere zu trennen bzw. sie anzureichern. Durch Umkristallisation einiger Verbindungen konnten Röntgenstrukturanalysen durchgeführt werden, welche die vorangegangenen Ergebnisse bestätigten.
In dieser Arbeit konnte durch die erstmalige Synthese und Charakterisierung der Verbindungen 24a-f – 34a-c gezeigt werden, dass photochemische Reaktionen alpha,beta-ungesättigten Enonen in Gegenwart ungesättigter Isocyanate möglich sind. Dabei werden Photoprodukte mit einer Isocyanatfunktion am Vierring gebildet, die durch saure Hydrolyse in Amine umgewandelt werden können. Dies ist eine neue Methode, um Amine in dieser Position darzustellen. Da sie auf thermischem Wege bisher nicht möglich war, bietet dieser photosynthetischer Weg neue Möglichkeiten um an Amine als Substitutenten in Vierringen zu gelangen.

This thesis deals with the photochemical behaviour of six-membered cyclic enones 1-3 in the presence of alkenylisocyanates 4-7. The synthesis of both the cyclic enones and the alkenyl iso-cyanates has been reported in the literature.
Irradiation (350 nm) of 2,3-dihydro-2,2-dimethyl-4H-pyran-4-one (1) in presence of any of the three isocyanates 4-6 affords [2+2]-photocycloadducts exclusively. In the reaction of 1 with 2-propenylisocyanate (4), the six products 24a-f are formed in a ration of 5: 4: 3: 3: 1.5: 1. In the reaction of 1 with vinylisocyanate (5), five products (27a-e) in a ratio of 2.1: 1.8: 1.8: 1: 0.6 are formed, the cis-fused head-to-tail-cycloadducts being obtained as main products. In the conver-sion of 1 in presence of 2-methylpropen-1-ylisocyanate (6) again five products (28a-e) can be characterized. It is noticeable that here the cis-fused head-to-head-cycloadducts predominate. In general, in all these reactions the endo-diastereomers are formed in larger amounts than their exo-counterparts.
Next, the reaction of 5,5-dimethylcyclohex-2-enone (2) in the presence of the same three isocy-anates was investigated. Comparison of the product analyses here shows that the reactions occur with less regio- and diastereoselectivities than those observed for 1. Four photoproducts are formed in the reaction of 2 with 2-propenylisocyanate (4), the endo-trans-head-to-tail-isomer 29a being the main product. All other cycloadducts are cis-fused. One of it is a head-to-tail-cycloadduct (29b) and the last two are head-to-head-cycloadducts (29c and 29d). In the reaction of 2 in the presence of vinylisocyanate (5), three products (30a-c) in a ration of 1: 0.9: 0.7 could be detected. In the reaction of (excited) 2 with isocyanate 6 a mixture of four products (31a-d) in a ration of 1: 0.9: 0.6: 0.5 is obtained. The main products here are cis-fused diastereomers, with a small preference of formation for the head-to-tail-cycloadduct 31d.
Finally the photochemical behaviour of 4,4-dimethylcyclohex-2-enone (3) in the presence of the same three isocyanates was examined. In the reaction of 3 with 4, two products (32a-b) are formed in a ratio of 1: 0.4, both being head-to-tail-regioisomers, and the endo-trans-head-to-tail-cycloadduct 32a representing the main product. In the conversion of 3 with 5, five products (33a-e) in a ratio of 1.1: 1: 0.9: 0.4: 0.4 were characterized. In this reaction the formation of a trans-head-to-head-cycloadduct was observed for the first time. Last but not least, in the reaction of (excited) 3 with 6, three products (34a-c) in a ratio of 1.1: 1: 0.7 are formed, the main prod-ucts being the exo-trans-head-to-tail-cycloadduct 34b and the endo-cis-head-to-tail-cycloadduct 34a.
Chromatographic work-up of all product mixtures failed, as none of the photoproducts could separated and isolated. Therefore the product mixtures were directly hydrolyzed in order to ob-tain the corresponding bicyclic amines. Whereas this conversion was successful for the carbo-cyclic cycloadducts deriving from enones 2 and 3, the photoproducts from 2,3-dihydro-2,2-dimethyl-4H-pyran-4-one (1) decomposed under these conditions. For the conversion of all other photoproducts it was observed that (a) the trans-fused cycloadducts isomerised to the more sta-ble cis-fused amines, and (b) only head-to-tail-cycloadducts could be converted into corre-sponding amines, as the head-to-head-cycloadducts underwent (cyclobutane)-ring opening reac-tions, as shown for compounds 29c-d, wich are convertedinto diketone 35b via a retro-Mannich-reaction. All amines 35a-b to 40a-b constitute endo-exo mixtures. As the separation of these di-astereomers could not be achieved on the amine level, protection groups, e.g. BOC or trifluoroacetyl, were introduced.
With the now modified N-functionalities, chromatographic separation and isolation of the single diastereomers was successful and several structures were now additionally established by means of X-ray structure analyses.
In summary, it has been shown for the first time in this work that it is possible to accomplish photocycloadditions of cyclic enones to alkenylisocyanates, and that the resulting 7- or 8-isocyanatobicyclo[4.2.0]octan-2-ones can be cleanly converted into the corresponding amines.
