Zusammenfassung: 
Als potentielle Zielstruktur für die Entwicklung von Antiinfektiva gelten Proteine, deren Aktivität für den Parasiten lebensnotwendig ist. Alle Versuche, das MAPK-Homolog LmxMPK7 in Leishmania mexicana zu deletieren, blieben erfolglos. Dies war ein erster Hinweis, dass es sich um ein essentielles Protein handeln könnte. Als zweiter Kandidat wurde die Glykogensynthase-Kinase GSK3 auf Essentialität in Leishmanien untersucht.
Das rekombinante Protein von LmxMPK7 zeigte keine Phosphorylierungsaktivität. Die endogene Kinase scheint in beiden Lebensstadien in geringer Menge hergestellt zu werden. LmxMPK7 konnte in Promastigoten genomisch deletiert werden. Hierzu wurde die Kinase ersatzweise von einem Plasmid exprimiert. Interessanterweise verloren Amastigoten der Mutante ΔLmxMPK7(-/-) + pX6LmxMPK7TYC-ds in der Maus und Promastigoten nach Wegfall des Selektionsdruck das Plasmid und es entstand die Nullmutante ΔLmxMPK7. Für den Parasiten scheint es wichtig zu sein, dass LmxMPK7 während des Lebenszyklus in korrekter Menge produziert wird. Episomale Überexpressoren zeigten im Kulturverlauf bei hoher Zelldichte und stationärem Wachstum eine Abnahme der Proteinmenge von LmxMPK7-TY. Liegt die Kinase nicht in physiologischer Menge vor, zeigt sich die Virulenz des Parasiten bei Infektion von BALB/c-Mäusen deutlich beeinträchtigt. Dies war sowohl für die Überexpressionsmutante als auch die Nullmutante von LmxMPK7 der Fall. Im Gegensatz zur gestörten Virulenz in vivo waren Promastigoten von ΔLmxMPK7 in der Lage Peritonealmakrophagen ex vivo zu infizieren und sich als Amastigoten intrazellulär zu vermehren. Die Diskrepanz im Infektionsverhalten könnte ein Anhaltspunkt dafür sein, dass LmxMPK7 für Anpassungsprozesse im Säugetierwirt oder die initiale Immunevasion von Bedeutung ist. ΔLmxMPK7 Leishmanien differenzierten in vitro normal und waren morphologisch nicht vom Wildtyp zu unterscheiden. Interessanterweise zeigten Promastigoten als auch Amastigoten der Nullmutante eine beschleunigte Zellteilung. Gleichermaßen ging die Abnahme der Menge an episomal hergestellter LmxMPK7-TY in der Überexpressionsmutante mit einer Proliferationsbeschleunigung in beiden Lebensstadien einher. Im Lebenszyklus des Parasiten gibt es Übergänge, an denen Umwandlungsprozesse gegenüber Wachstum Vorrang haben, auch wechseln sich proliferative und nicht-proliferative Formen ab. LmxMPK7 könnte für die negative Wachstumsregulation an einem dieser Übergänge oder in einer nicht-proliferativen Form verantwortlich sein. Allerdings blieb nach Komplementation von ΔLmxMPK7, durch Einbringung von LmxMPK7 auf einem Allel, die Wachstumsbeschleunigung erhalten. Womöglich bleiben Mechanismen, welche zur Kompensation der fehlenden LmxMPK7-Funktion in ΔLmxMPK7 beitragen, bestehen und machen die „add-back“-Mutante unabhängig von LmxMPK7.
Die lange LmxGSK3α unterscheidet sich im Homologievergleich deutlich von der kurzen LmxGSK3β. Am N-Terminus von LmxGSK3α wurde ein Signalpeptid des sekretorischen Weges identifiziert. Das rekombinante Protein von LmxGSK3α besitzt Kinase-Aktivität und wird von LmxGSK3β phosphoryliert. Beide GSK3-Homologe zeigen im Kinase-Aktivitätstest Autophosphorylierungsaktivität. Durch Herstellung einer YF-Mutante konnte bewiesen werden, dass Phosphotyrosin-186 in der Aktivierungsschleife von LmxGSK3β für die Kinase-Aktivität notwendig ist. LmxGSK3β ist in beiden Lebensstadien vorhanden. Rekombinante LmxGSK3β phosphorylierte mehrere Proteine in Leishmanienlysaten. Dies könnte bedeuten, dass LmxGSK3β im Parasiten mehrere Substrate und Aufgaben besitzt. Lithiumchlorid hemmte die Kinase-Aktivität von rekombinanter LmxGSK3β und nach Zugabe zu einer Leishmanienkultur das Wachstum von Promastigoten und Amastigoten. Zu den Lithium-induzierten Effekten in Promastigoten gehörte eine signifikante Verlängerung des Flagellums. Außerdem wurde ein Defekt der Cytokineseinitiation festgestellt. Weitere Untersuchungen sind notwendig, um die beobachteten Effekte von Lithium abschließend einer GSK3-Inhibition zuzuordnen. Es ist nicht möglich eine Nullmutante von LmxGSK3β herzustellen. Die Kinase konnte erst nach Expression vom Plasmid genomisch deletiert werden. Die Mutante ΔLmxGSK3β(-/-) + pXPACLmxGSK3βPTPNterm Klon H4PPAC zeigte sich in der Maus infektiös. Läsionsamastigoten von H4PPAC verlieren die episomale Kopie von LmxGSK3β nicht (Munro, Dissertation 2013).
Fazit: Leishmanien besitzen die Möglichkeit die MPK7-Funktion zu umgehen, dies macht LmxMPK7 zu keinem idealen „Drug-Target“. LmxGSK3β ist für Leishmanien essentiell und eine hervorragende Zielstruktur für die Entwicklung von neuen Medikamenten zur Bekämpfung der Leishmaniose.

Proteins that are essential for the survival of a parasite could be potential targets for the development of anti-infectives. This thesis is concerned with the analysis of two glycogen synthase kinase homologues, LmxGSK3α and LmxGSK3β, and the mitogen-activated protein kinase homologue LmxMPK7 to functionally characterise these proteins and to establish whether any of them is an essential protein for Leishmania.
The recombinantly expressed protein of LmxMPK7 did not exhibit kinase activity. LmxMPK7 is present in both life stages but its expression appears to be weak. A plasmid carrying the gene of LmxMPK7 was introduced into the parasite. In the presence of this extra-chromosomal copy of LmxMPK7 the replacement of the genomic copies was successful. Interestingly, the resulting mutant lost the extrachromosomal copy of LmxMPK7, both in amastigotes in mice and in promastigotes cultivated without selection pressure. Hence, this led to the generation of the null mutant ΔLmxMPK7. The protein level of LmxMPK7 appears to be important for the parasite and might be tightly regulated during its life cycle. The amount of TY-tagged LmxMPK7-TY was strongly reduced during cultivation at high cell densities and during stationary growth phase in mutants over-expressing this protein. In vivo investigations revealed that non-physiological amounts of LmxMPK7 entailed a significant impairment of virulence in BALB/c-mice for both the over-expressing mutant and the null mutant. Contrary to the virulence defect in vivo, promastigotes of ΔLmxMPK7 infected isolated peritoneal macrophages and amastigotes proliferated successfully within the parasitophorous vacuole of the host cell. This discrepancy in infection behaviour could be a hint that LmxMPK7 might be involved in the adaptation of the parasite to the host environment in mammals or in processes of initial immune evasion. ΔLmxMPK7 Leishmania differentiated normally in vitro and cells displayed a normal morphology. Promastigotes and amastigotes of ΔLmxMPK7, however, presented an accelerated proliferation. Similarly the decrease in LmxMPK7-TY expressed from a plasmid was accompanied by an increased proliferation in both life stages. This indicates that the in vivo role of LmxMPK7 could be involved in the negative regulation of growth. In the life cycle of the parasite dividing forms alternate with non-dividing forms and there are transitions where transformation processes precede cell growth. It is hypothesised that the potential arrest in growth caused by LmxMPK7 implicates one of those transitions or plays an important role in a non-proliferative stage of the parasite. An add-back mutant was generated by reintroducing a wild type copy of LmxMPK7 back into one allele of the null mutant. However, the add-back mutant retained faster growth which may be because of to maintained compensation mechanisms that ΔLmxMPK7 had developed keeping the add-back mutant in a LmxMPK7-unresponsive mode.
Sequence homology analysis revealed that the long LmxGSK3α is markedly different from the short LmxGSK3β. Interestingly, LmxGKS3α presents with a N-terminal signal peptide for import into the ER. The recombinant protein of LmxGKS3α showed kinase activity and was phosphorylated by LmxGSK3β in the in vitro kinase assay. Both GSK3 homologues displayed in vitro autophosphorylation. Furthermore, phosphorylation on tyrosine 186 in the activation loop of LmxGSK3β was proved to be essential for kinase activity by generation of a YF-mutant. LmxGSK3β is present in both life stages of the parasite. Kinase assays using recombinant LmxGSK3β and L. mexicana cell lysates implied the existence of several potential substrates in the parasite. Lithium chloride could be identified as a potential inhibitor of recombinant LmxGSK3β activity in vitro. Treatment of Leishmania cultures with LiCl resulted in a reduction of cell growth in promastigotes and amastigotes. In promastigotes an elongation of flagella and a defect in initiation of cytokinesis could be observed in the presence of lithium. Further investigations are required to conclusively link the effects of lithium treatment to GSK3 inhibition. LmxGSK3β could not be deleted from the L. mexicana genome. To overcome this problem a plasmid carrying the LmxGSK3β gene was introduced into the parasite. In the presence of the additional extrachromosomal copy the replacement of the genomic copies of LmxGSK3β was successful. The resulting mutant was named H4PPAC and was infective in mice. It was demonstrated that lesion-derived amastigotes from H4PPAC had not lost the episomal copy of LmxGSK3β in mice (Munro, dissertation 2013).
In conclusion, Leishmania can overcome a lack of LmxMPK7, which makes LmxMPK7 a flawed target for drug development. LmxGSK3β is essential for Leishmania and therefore represents a potential target for a drug which could combat Leishmaniasis.
