Bestimmte Formen synaptischer Plastizität setzten die lokale Proteinsynthese in Dendriten voraus. Das Makorin RING Finger Protein-1 short (MKRN1s) ist ein potenziell positiver Regulator der lokalen Translation in Nervenzellen und als solcher an der Regulation synaptischer Plastizität beteiligt.
Dieser Arbeit lag die Beobachtung zugrunde, dass MKRN1s mit dem poly(A)-bindenden Protein (PABP) interagiert und zusammen mit PABP in granulatartigen Strukturen entlang von Dendriten lokalisiert. PABP ist eine Schlüsselkomponente verschiedener Ribonukleoprotein-Komplexe (RNPs) und übernimmt bedeutende Funktionen bei der Kontrolle der Translation, der Stabilität und der subzellulären Lokalisierung von mRNAs. MKRN1s ist im Gehirn der Ratte die primäre MKRN1-Isoform und assoziiert in RNPs mit dendritisch lokalisierten mRNAs. Nach elektrischer Stimulation von Dendriten wird MKRN1s an aktivierte Synapsen rekrutiert. Wenn MKRN1s an eine Reporter-mRNA gebunden ist, stimuliert es die Translationsrate dieser mRNA in primären Neuronen. MKRN1s enthält drei Zinkfinger-Domänen vom Typ C3H, ein PAM2- (PABC recognition motif 2) Motiv, einen MKRN-typischen Zinkfinger und eine verkürzte C3HC4-RING-Domäne.
Das Ziel dieser Arbeit war es, die Wirkweise von MKRN1s im RNA-Metabolismus besser zu verstehen. Dazu wurde die Bedeutung einzelner Domänen für die MKRN1s-vermittelte Translationsstimulation untersucht und neue Interaktionspartner von MKRN1s identifiziert. In gestressten eukaryotischen Zellen akkumulierte MKRN1s in sogenannten stress granules, zytoplasmatische Strukturen, in denen mRNPs transient der Translation entzogen werden. Für diesen Prozess ist der erste Zinkfinger (ZF1) und das PAM2-Motiv essenziell. Für die effiziente Translationsstimulation einer Reporter-mRNA sind ebenfalls ZF1 und das PAM2-Motiv essenziell.
Infolge einer massenspektrometrischen Untersuchung und anschließender Validierung putativer Interaktionspartner konnten LARP1 (La-related protein 1) und UPF1 (regulator of nonsense transcripts 1) als neue Interaktionspartner von MKRN1s identifiziert werden. Für die Wechselwirkung der zwei Proteine mit MKRN1s ist ein funktionelles PAM2-Motiv essenziell. Durch Rekrutierung von MKRN1s an eine Reporter-mRNA in Abwesenheit von LARP1 bzw. UPF1 konnte gezeigt werden, dass die Wechselwirkung von MKRN1s mit LARP1 und UPF1 für die translationsfördernde Wirkung von MKRN1s essenziell ist. MKRN1s interagiert mit LARP1 indirekt über PABP als Interaktionsgerüst. Dabei bindet MKRN1s über das PAM2-Motiv an die vier RRM- (RNA recognition motifs) und der MLLE- (mademoiselle) Domäne von PABP. LARP1 bindet über das La-Modul an die vier RRM-Domänen von PABP. Weiterhin konnte gezeigt werden, dass MKRN1s in Abhängigkeit der Bindung mit PABP mit dem Translationsinitiationskomplex eIF4F interagiert. Die effiziente translationsfördernde Wirkung von MKRN1s auf eine Reporter-mRNA ist davon abhängig, dass das vollständige Set an Translationsinitiationsfaktoren vorhanden ist. Die beobachteten Daten deuten darauf hin, dass MKRN1s in Wechselwirkung mit LARP1, PABP und UPF1 die Translationsinitiation reguliert.

Certain forms of synaptic plasticity require local protein synthesis in dendritic spines. Makorin RING Finger Protein-1 short (MKRN1s) is a potential positive regulator of local translation in nerve cells and may therefore participate in the regulation of such forms of synaptic plasticity.
This work is based on the observation that MKRN1s interacts with poly(A) binding protein (PABP) and co-localizes together with it in granular structures along dendrites. PABP is a key component of various ribonucleoprotein complexes (RNPs) and plays an important role in controlling translation rate, stability and subcellular localization of mRNAs. MKRN1s is the most common MKRN1 isoform in the rat brain and is associated with mRNAs in RNPs of dendrites. MKRN1s is recruited to activated synapses after electrical stimulation. In primary neurons, when MKRN1s is bound to a reporter mRNA, it increases its translation rate. MKRN1s contains three C3H-type zinc finger domains, a PAM2 (PABC recognition motif 2) motif, a MKRN-type zinc finger and a truncated C3HC4-RING domain.
The aim of this dissertation was to better understand the unknown mode of action of MKRN1s in RNA metabolism. For this purpose, the relevance of individual domains for MKRN1s-mediated translation stimulation was investigated and new interaction partners of MKRN1s were identified. MKRN1s accumulates in stressed eukaryotic cells in so-called stress granules: cytoplasmic structures in which mRNPs are transiently removed from translation. The first zinc finger (ZF1) and the PAM2 motif are essential for this process. ZF1 and the PAM2 motif are also essential for efficient translation stimulation of a reporter mRNA in neurons.
Mass spectrometric investigation and subsequent validation of putative interaction partners have identified two new binding partners of MKRN1s: LARP1 (La-related protein 1) und UPF1 (regulator of nonsense transcripts 1). The interaction of these proteins with MKRN1s depends on a functional PAM2 motif. The recruitment of MKRN1s to a reporter mRNA in the absence of LARP1 or UPF1 has shown that the interaction of MKRN1s with LARP1 and UPF1 is essential for the translation-promoting effect of MKRN1s. Interaction analyses have shown that the interaction of MKRN1s with LARP1 is mediated via PABP. Previous studies have shown that MKRN1s interacts with all PABP domains via its PAM2 motif. This dissertation shows that LARP1 binds to the four RRM (RNA recognition motifs) domains of PABP via its La module. Finally, it has been demonstrated that MKRN1s interacts with the eIF4F translation initiation complex in a manner dependent on of the interaction with PABP. The translation promoting effect of MKRN1s on reporter mRNA depends on the availability of the complete set of translation initiation factors. The observed data suggest that MKRN1s regulates translation initiation in association with LARP1, PABP and UPF1.
